The present study focused on the behavioral and biochemical responses of Japanese medaka (Oryzias latipes) to acute, sublethal exposure to CPF, as well as the persistence of the effects during a 3-week recovery test in CPF-free water. The medaka became hyperactive and exhibited an elevated anxiety state after a 4-day exposure to 0.024 mg/L of CPF, but they recovered from these abnormal behavioral responses within 7 days of recovery treatment. In contrast, persistent impacts on some startle responses to a sudden stimulation (induced by a ball drop) were observed in medaka exposed to CPF. The reaction latency did not change immediately after the 4-day exposure, but was significantly prolonged by as much as 21 days after the termination of exposure. The post-stimulus swimming distance within 5 s significantly decreased on the day immediately after the 4-day exposure, but it significantly increased after 7 days of recovery treatment. The activity of acetylcholinesterase (AChE) in the brains of medaka was significantly inhibited on the day immediately after the 4-day exposure, but it returned to 80% and 110% of that in control fish on days 7 and 21 of the recovery period, respectively. However, AChE activities in the eyes of exposed medaka were persistently inhibited and declined to 33%, 71%, and 72% of that in control fish on days 0 (immediately after the 4-day exposure), 7, and 21 of recovery, respectively. Correlation analysis suggested that the changes of AChE activities in the brains of medaka may underlie some of the observed acute behavioral changes, and the changes of AChE activities in the eyes may contribute to the persistence of the abnormalities in the reaction latency of the startle response. Our findings suggest that medaka need a long time to recover from acute, sublethal exposure to CPF, and the persistence of the behavioral abnormalities might affect their fitness in natural habitats..

Relative high concentrations of 110mAg (2.1–127�Bq�kg-wet−1), 134Cs (2.6–61�Bq�kg-wet−1), and 137Cs (3.5–92�Bq�kg-wet−1) were detected in specimens from the eastern Honshu areas. Significantly lower 137Cs concentrations (0.7–1.6�Bq�kg-wet−1) were detected in specimens from western and northern Honshu. The decay-corrected 137Cs concentration was significantly inversely correlated with the distance from the Fukushima Dai-ichi Nuclear Power Plant. Thus, wharf roach may serve as a good bioindicator for monitoring radioactive contamination of its habitats..

The results revealed that the sinking rate of both diatoms exposed to diuron at a level of 50% effective concentration for growth (EC50) decreased significantly compared with the control. Photosynthetic performance (Fv/Fm and PIABS) of both diatoms also decreased significantly with diuron exposure. The number of cells per chain in S. marinoi-dohrnii decreased significantly with diuron treatment, but T. pseudonana cell diameter remained stable. Neutral lipid concentration per cell was significantly higher compared with control at 72 h in both diatom species exposed to EC50 level diuron. And water-soluble protein concentration per cell at 72 h was lower than control in the T. pseudonana EC50 group only. These biochemical changes may decrease specific gravity of cells and seems to cause a decreased sinking rate in diatoms. The positive significant correlation between the numbers of cells per chain and sinking rate in S. marinoi-dohrnii indicated that chain length is also an important factor in sinking rate regulation for chain-forming diatoms. Thus, our present study suggested that suppression of photosynthetic performance and the resultant physiochemical changes induced the decreased sinking rate that may inhibit the normal survival strategy (avoidance from the surface layer where strong light either causes photo-inhibition or interrupts resting cell formation). Therefore, the use of antifouling agents should be considered for the sustainable marine environment..

Hai, Xuchun Qiu, Hai Xu, Masato Honda, Mitsuyasu Yabe, Kiwao Kadokami, Yohei Shimasaki, Yuji Oshima, Contaminants in Liquid Organic Fertilizers Used for Agriculture in Japan, Bulletin of Environmental Contamination and Toxicology, 10.1007/s00128-017-2081-y, 99, 1, 131-137, 2017.01, [URL], To provide an overview of anthropogenic contaminants in liquid organic fertilizers (LOFs), products from four biogas plants in Kyushu, Japan, were analyzed for a wide range of contaminants, including copper, cadmium, tributyltin (TBT), dibutyltin (DBT), perfluorooctane sulfonate, 952 semi-volatile organic compounds, and 89 antibiotics. The highest concentrations of copper (31.1 mg/L) and cadmium (0.08 mg/L) were found in LOFs from the Hita biogas plant. Only ofloxacin and sulfapyridine were detected in total 89 antibiotics screened. TBT, DBT, and perfluorooctane sulfonate were present at low concentrations in the LOFs from all four locations. Among the 952 semi-volatile organic compounds, 78 compounds were detected in at least one sample and were present at concentrations between 1.2 and 139.6 mg/L. On the basis of comparisons with previous studies and quality standards for the use of organic fertilizers, the concentrations of contaminants in the studied LOFs indicate that they might be safe for agricultural purposes..

In fish, it is metabolized to 3-hydroxybenzo[c]phenanthrene (3-OHBcP). In the present study, we observed the effects of 1 nM 3-OHBcP on the development and gene expression of Japanese medaka (Oryzias latipes) embryos. Embryos were nanoinjected with the chemical after fertilization. Survival, developmental stage, and heart rate of the embryos were observed, and gene expression differences were quantified by messenger RNA sequencing (mRNA-Seq). The exposure to 1 nM 3-OHBcP accelerated the development of medaka embryos on the 1st, 4th, and 6th days post fertilization (dpf), and increased heart rates significantly on the 5th dpf. Physical development differences of exposed medaka embryos were consistent with the gene expression profiles of the mRNA-Seq results for the 3rd dpf, which show that the expression of 780 genes differed significantly between the solvent control and 1 nM 3-OHBcP exposure groups. The obvious expression changes in the exposure group were found for genes involved in organ formation (eye, muscle, heart), energy supply (ATPase and ATP synthase), and stress-response (heat shock protein genes). The acceleration of development and increased heart rate, which were consistent with the changes in mRNA expression, suggested that 3-OHBcP affects the development of medaka embryos. The observation on the developmental stages and heart beat, in ovo-nanoinjection and mRNA-Seq may be efficient tools to evaluate the effects of chemicals on embryos..

STC is involved in the decrease in plasma Ca'Jt level in teleosts. Comparative studies between invertebrate chordate STC and two paralogous vertebrate STCs suggest that the chordate ancestral gene of STC diverged into two paralogous genes in the evolution of vertebrates. However, this evolutionary scenario remains unclear. To clarify the diversification process of vertebrate STC, we made an attempt to isolate orthologs of STC in extant jawless fishes (cyclostomes) located most distantly in the vertebrate tree of life. In the present study, we searched the STC-like sequence by performing degenerate PCR in the inshore hagfish Eptatretus burgeri. We identified a STC homolog, designated as Eb-STC. Our molecular phyloge- netic analysis confirmed that Eb-STC is a member of the STC family. The comparison of amino acid sequences revealed that Eb-STC is similar to gnathostome STC1 and 2, in that Eb-STC shares the 10 conserved cysteine residues that form intramolecular disulfide bonds. Eb-STC and gnathostome STCs possess the conserved -glycosylated site. However, typical features discriminated between STC1 and 2, such as the distinct position of the cysteine residue associated with the intermolecular dimer, were not observed in Eb-STC. Tissue distribution analysis revealed that Eb-STC is expressed in various organs, but not in the brain. This expression pattern resembles that of teleost STC1 and 2, suggesting that Eb-STC acts as a paracrine factor. This study aimed to gather information on the molecular mechanisms underlying the physiological responses in field C. marina population during HAB. Proteomic analysis showed that the abundance of -37% protein spots (40 out of 108 detected from 2-DE gel images) significantly varied with the sampling date. Significant decreases in the abundances of proteins involved in photosystem II (LHCP 4), electron transfer chain (Cyt c553), Calvin cycle (GAPDH), and chloroplast antioxidant system (2-Cys Prx) were observed as the bloom progressed, suggesting the efficiencies of those photosynthetic pathways declined during the bloom. In addition, the abundances of the above proteins showed significant positive correlations with the Fv/Fm ratio and growth rate of C. marina and with DIN concentrations (except LHCP 4). Our findings suggested that declined expressions of those photosynthesis-related proteins presented some molecular foundation of the decreases in Fv/Fm ratio and growth rate of C. marina during the bloom, and also provided insight into mechanistic links between the external/internal factors and physiological responses of C. marina that may ultimately dictate the ecology of the bloom..

We found the most iCT cells (53-70 cells) in the cerebral ganglion. We also detected iCT cells (4-6 cells) in the subpharyngeal ganglion. Furthermore, it was noted that iCT cells were present bilaterally in each segment of the ventral nerve cord. These results suggest that iCT cells play some functional roles in the nervous system. Next, the molecular weight (MW) of the iCT substance was examined using the western blotting method. Cerebral ganglia were collected from 200 individuals. These ganglia were homogenized and centrifuged. The separated supernatants were treated with 66% acetone and then dialyzed to remove low MW substances (less than 2,000) at 4°C. After lyophilization, the sample was reconstituted with 1 M acetic acid and then fractionated with an ultrafiltration membrane system into MWs of 3,000 to 10,000. Thereafter, the specimen was separated by SDS-polyacrylamide gel electrophoresis and analyzed by western blotting with anti-salmon calcitonin (CT) antiserum. Our results indicated that the MW of the iCT substance was close to that of teleost fish CT (3.5 kDa). The annelid Capitella teleta has two genes encoding CT-like peptides. This suggests that the iCT substance in P. aibuhitensis includes amino acid residues similar to fish CT and belongs to the CT family..

Exposure to low-oxygen had no effect on survival. Animals moved upward above the sediment under low-oxygen conditions and downward under aerobic conditions, repeating these movements for 19 d. The difference in shell height above the sediment before and after low-oxygen exposure gradually decreased; after 20 d there was no significant difference. Low-oxygen exposure also resulted in a significant decrease in glycogen content and in concentrations of malate and fumarate. Thus, repeated exposure of pen shells to low-oxygen conditions reduces glycogen content, alters metabolism, and affects vertical movements, all of which may cause ecological dysfunction. We suggest that hypoxia might be one factor contributing to the collapse of pen shell resources..

In this study, we studied variations in the social behavioral patterns, swimming behavior, and brain acetylcholinesterase (AChE) activity of Japanese medaka exposed to lethal (0.12 mg/L) and sublethal (0.012 mg/L) concentrations of CPF after different exposure times. Group behavior performance (schooling, shoaling, and solitary) was determined on day 4 of lethal exposure and on days 4, 8, and 12 of sublethal exposure. Swimming speed and brain AChE activity were measured on days 4, 8, 12, 16, and 20 of sublethal CPF exposure. We observed significant decreases in social behavior and swimming speed (i.e., hypoactivity) in fish exposed to lethal CPF concentrations for 4 days. At the sublethal concentration, there was increased schooling duration and hyperactivity of fish on day 8 but not on day 4. In contrast, 12 days of sublethal CPF exposure resulted in social behavior responses similar to those after 4 days’ lethal exposure, i.e., significant decreases in schooling frequency and duration with a notable increase in duration of solitary behavior. Brain AChE activity was inhibited in a time-dependent manner. Altered fish behavior in response to organophosphorus pesticides such as CPF may be mediated by more than AChE inhibition alone..

This study represents the first attempt that uses the accelerated failure time (AFT) model to quantify the interaction and toxicity of multi-chemical mixtures in environmental toxicology. We firstly conducted the acute immobilization tests with Daphnia magna exposed to mixtures of diazinon (DZN), fenitrothion (MEP); and thiobencarb (TB) in single, binary, and ternary formulations, and then fitted the results to the AFT model. The 48-h EC50 (concentration required to immobilize 50% of the daphnids at 48 h) values for each pesticide obtained from the AFT model are within a factor of 2 of the corresponding values calculated from the single pesticide exposure tests, indicating the methodology is able to provide credible toxicity values. The AFT model revealed either significant synergistic (DZN and MEP; DZN and TB) or antagonistic (MEP and TB) interactions in binary mixtures, while the interaction pattern of ternary mixture depended on both the concentration levels and concentration ratios of pesticides. With a factor of 2, the AFT model accurately estimated the toxicities for 78% of binary mixture formulations that exhibited significant synergistic effects, and the toxicities for all the ternary formulations. Our results showed that the AFT model can provide a simple and efficient way to quantify the interactions between pesticides and to assess the toxicity of their mixtures. This ability may greatly facilitate the ecotoxicological risk assessment of exposure to multi-chemical mixtures..

A single dose of PFOS was intraperitoneally injected at 0.1 mg/kg body weight with samples taken over a 14-day period. The highest concentration of PFOS was found in the plasma, 861 ng/mL at 14 days, followed by the mucus, liver, blood clot, gonads, muscles, and skin of fish. A gradual upward trend in PFOS concentration was observed in the mucus and liver whereas there was no change in the plasma, blood clot, gonad, muscle, and skin after the initial increase in PFOS concentrations following injection. No significant trend for estimated total PFOS content in whole body was observed during the experimental period. Relatively high concentrations of PFOS (690 ng/g ww after 14 days) were detected in body surface mucus that continuously oozes from the skin. These results may suggest that mucus is one of the elimination pathways of PFOS in tiger puffer fish..

The open reading frame of 2-Cys Prx was 585 bp long and encoded a protein consisting of 195 amino acids. The putative amino acid sequence contained two cysteine residues located at the 49th and 170th amino acid positions from the N-terminal methionine residue. The sequence also possessed 2-Cys Prx characteristic motifs, F (FFYPLDFTFVCPTEI) and EVCP. The position of the 2-Cys Prx gene relative to several others (ycf59–2-CysPrx–rpl35–rpl20) was the same as that found in the chloroplast genome in the raphidophyte Heterosigma akashiwo. Upstream of the 2-Cys Prx gene, possible TATA and GGA motifs recognized by nuclear-encoded plastid RNA polymerase (NEP), and a possible -10 box and -35 box recognized by plastid-encoded plastid RNA polymerase (PEP) were observed. We measured the transcript levels of 2-Cys Prx in C. marina cells grown under three different light intensities (0, 100, 1000 µmol photons m–2 s–1, 14-h light/8-h dark photoperiod) by quantitative PCR. The 2-Cys Prx transcript level in cells grown under the highest light intensity on day 3 was threefold that on day 0 but two lower light intensities resulted in relatively stable transcription levels. The 2-Cys Prx transcript level was significantly positively related to the H2O2 concentration per cell and the H2O2 scavenging activity per cell. These results suggest that C. marina 2-Cys Prx functions in the chloroplast and its transcription could be regulated by both NEP and PEP. Moreover, the 2-Cys Prx transcript level might increase to remove excessive H2O2 produced under strong light conditions in order to maintain cell proliferation activity..

PAH and alkylated PAH (alkPAH) concentrations in wharf roaches collected from nine sampling sites on the central and northern coast of Japan in November 2011 were analyzed. Total alkPAH concentrations were higher than those of PAHs at all sampling sites. Seven sampling sites had relatively low total concentrations of PAHs and alkPAHs (57.7–130 and 276–770 ng/g-dry, respectively); they included sites in Miyagi Prefecture (St. 5 and 9) that were seriously damaged by the Great East Japan Earthquake and by the following Tsunami on 11 March 2011. Two sampling sites (St. 2: Kominato, Aomori Prefecture; St. 3: Usushiri, Hokkaido) had higher concentrations of total PAHs and alkPAHs (PAHs, 1025 and 591 ng/g-dry, respectively; alkPAHs, 1490 and 1160 ng/g-dry, respectively). Diagnostic ratio calculation showed that St. 2 and 3 had PAHs of pyrolytic origin, possibly from vehicle exhaust gas or industrial fuel combustion, or both. The concentrations of these pollutants in the wharf roach were similar to those in bivalves and therefore likely reflected PAH pollution at the collection sites. The wharf roach may be a suitable candidate indicator of PAH pollution, not only in the intertidal zone inhabited by the currently used bivalve indicators, but also in the supratidal zone..
