Cooper, kiyoshi koyano, Topography influences cellular functions and RhoA activity of titanium surface-adherent cells., 9th International Wrokshop on Biomaterials in Interface Science, 2014.08, Purpose: RhoA, one of Rho-family of GTPases, is known to influence cell adhesion and motility through organization of the actin cytoskeleton in response to various stimuli. The aim of this study was to investigate cellular functions and RhoA activities of the adherent cells on titanium substrates with three kinds of topography.
Materials and Methods: Three kinds of surface topography Ti disks were prepared. Smooth (S) surfaces were passivated with 30% HNO3. HCl-treated (H1) surfaces were grit-blasted with 100-μm aluminum oxide (Al2O3) particles, subsequently treated with 5N HCl and passivated with 30% HNO3. H2O2+H2SO4-treated (H2) surfaces grit-blasted with 100-μm Al2O3 particles and subsequently treated with 30% H2O2 and 2N H2SO4. These surfaces were characterized by SEM analysis and the measurement of surface roughness. To evaluate the effect of surface topography and RhoA inactivation on cellular functions, murine mesenchymal stem cells were cultured on each disk and cell morphology, RhoA activity, wound healing rates (cell migration) and osteogenic markers’ expressions were investigated with or without RhoA inhibitor (C3).
Results: Each disk showed a specific surface topography and surface roughness analysis indicated the highest roughness in H1 surfaces (H1>H2>S). Cell morphology was influenced by surface topography and C3. RhoA was activated in a surface roughness-dependent manner. The highest wound healing rate was observed on S surface. RhoA inhibition also enhanced cell migration on S and H1 surfaces, but not H2 surface. The expression of osteogenic markers was inhibited by C3 in S and H1 adherent cells, but not in H2 adherent cells.
Conclusions: Topography influences cellular functions and RhoA activity. However, the effect of RhoA inhibition on cellular functions may be in a surface topography- dependent manner.
