Cell, 39, 87-103, 2016.10, 配偶子の形成にはDNAメチル化などのエピゲノムの制御が重要であり、その制御機構の解明は不妊や流産などの克服に資する。我々は高精度で網羅的なDNAメチル化解析法を用いて、京大・斎藤通紀らとの共同研究により、培養下の胚性幹細胞から誘導された始原生殖細胞様細胞のメチル化状態を解析し、そのメチル化状態は基本的に生体内のそれに酷似すること、転写制御因子Prdm14が始原生殖細胞様細胞におけるメチル化パターンの形成に必須であることを明らかにした。.

Commun., 6, 7870-7870, 2015.07, 常染色体劣性遺伝病であるICF症候群の原因遺伝子としてCDCA7とHELLSの2つを発見した。ICF症候群は先天性の免疫不全症で、染色体の動原体領域のDNA低メチル化が特徴的である。今回の研究成果はICF症候群の確定診断と早期治療に役立つばかりでなく、将来的に遺伝子治療の開発へと繋がるものである。また、発見された2つの遺伝子がどのように動原体のDNAメチル化に関わるのかを解明することで、染色体の安定性や免疫細胞の分化の機構が明らかになる。.

Cell , 20, 364-375, 2011.03, 生物のゲノムに寄生するトランスポゾンは挿入突然変異の原因となるため、生殖細胞はDNAメチル化やpiRNA 等の小分子RNAによりトランスポゾンを抑制する防御機構を持っている。本論文では、ホスホリパーゼDスーパーファミリーのメンバーでミトコンドリア膜の脂質カルジオリピンを加水分解するMITOPLDが、マウスにおいてpiRNAの産生に関わることを明らかにした。さらに、MITOPLDはpiRNAの生合成に関わる細胞内小器官とタンパク質の細胞内輸送に関わることを発見した。この研究により、脂質代謝と小分子RNAの産生という一見関連性の薄い事象に新たな繋がりが見つかり、生体防御機構に新たな知見をもたらした。.
