Animals sense many environmental stimuli and, by responding to them, they behave properly. We are interested in mechanisms on sensory processing in neuronal circuits which regulates behavior. We use C. elegans as a model organism to elucidate molecular mechanisms of informational processing. I give lectures on basic molecular genetics and molecular biology for under graduate student. In addition for members in our lab, I have earnest discussions not only on their students studies but also on the logic and methodology of biological science.

    3D cell tracking in a living organism has a crucial role in live cell image analysis. Cell tracking in C. elegans has two difficulties. First, cell migration in a consecutive frame is large since they move their head during scanning. Second, cell detection is often inconsistent in consecutive frames due to touching cells and low-contrast images, and these inconsistent detections affect the tracking performance worse. In this paper, we propose a cell tracking method to address these issues, which has two main contributions. First, we introduce cell position heatmap-based non-rigid alignment with test-time fine-tuning, which can warp the detected points to near the positions at the next frame. Second, we propose a pairwise detection method, which uses the information of detection results at the previous frame for detecting cells at the current frame. The experimental results demonstrate the effectiveness of each module, and the proposed method achieved the best performance in comparison.

    Cross-linking of proteins by mammalian transglutaminases (TGs) plays important roles in physiological phenomena such as blood coagulation and skin formation. We show that Drosophila TG suppressed innate immune signaling in the gut. RNA interference (RNAi) directed against TG reduced the life span of flies reared under conventional nonsterile conditions but not of those raised under germ-free conditions. In conventionally reared flies, TG RNAi enhanced the expression of genes encoding antimicrobial peptides in the immune deficiency (IMD) pathway. Wild-type flies that ingested gut lysates prepared from conventionally reared TG RNAi-treated flies had shorter life spans. In conventionally reared flies, TG RNAi triggered apoptosis in the gut and induced the nuclear translocation of Relish, the NF-κB (nuclear factor κB)-like transcription factor of the IMD pathway. Wild-type flies that ingested synthetic amine donors, which inhibit the TG-catalyzed protein-protein cross-linking reaction, showed nuclear translocation of Relish and enhanced expression of genes encoding IMD-controlled antimicrobial peptide genes in the gut. We conclude that TG-catalyzed Relish cross-linking suppressed the IMD signaling pathway to enable immune tolerance against commensal microbes.
