(1) We create various mouse models for psychiatric disorders using C57BL/6J mice in our lab, such as, depression, schizophrenia, PTSD, cocaine addiction, and cancer-related cognitive impairment, etc. (2) We analyze behavioral phenotypes of model models for psychiatric disorders are analyzed by the test battery, such as open-field test, T-maze, Y-maze, elevated plus maze, fear conditioning, operant conditioning, CPP, FST, and PPI, etc. (3) We analyze synapses, neurons, and glial cells in the neuronal circuits of the hippocampus obtained from various model mice to understand the pathophysiology of psychiatric disorders. To this end, we use anatomical, pharmacological, and molecular biological methods. We also used the DREADD technologies with several Cre transgenic mouse lines. (4) We analyze synapses, neurons, and glial cells in the neuronal circuits of the hippocampus of normal mice using anatomical and electrophysiological methods to understand the operating principles of the brain. (5) Multidisciplinary analysis of the extracellular matrix in the central nervous system by using anatomical and electrophysiological techniques. (6) Education of undergraduate students via lecture and practical course of neuroanatomy and gross anatomy.

    Recent studies have highlighted the potential involvement of reactive oxygen species (ROS) and microglia, a major source of ROS, in the pathophysiology of schizophrenia. In our study, we explored how the second-generation antipsychotic risperidone (RIS) affects ROS regulation and microglial activation in the hippocampus using a mouse ketamine (KET) model of schizophrenia. KET administration resulted in schizophrenia-like behaviors in male C57BL/6J mice, such as impaired prepulse inhibition (PPI) of the acoustic startle response and hyper-locomotion. These behaviors were mitigated by RIS. We found that the gene expression level of an enzyme responsible for ROS production (Nox2), which is primarily associated with activated microglia, was lower in KET/RIS-treated mice than in KET-treated mice. Conversely, the levels of antioxidant enzymes (Ho-1 and Gclc) were higher in KET/RIS-treated mice. The microglial density in the hippocampus was increased in KET-treated mice, which was counteracted by RIS. Hierarchical cluster analysis revealed three morphological subtypes of microglia. In control mice, most microglia were resting-ramified (type I, 89.7%). KET administration shifted the microglial composition to moderately ramified (type II, 44.4%) and hyper-ramified (type III, 25.0%). In KET/RIS-treated mice, type II decreased to 32.0%, while type III increased to 34.0%. An in vitro ROS assay showed that KET increased ROS production in dissociated hippocampal microglia, and this effect was mitigated by RIS. Furthermore, we discovered that a NOX2 inhibitor could counteract KET-induced behavioral deficits. These findings suggest that pharmacological inhibition of ROS production by RIS may play a crucial role in ameliorating schizophrenia-related symptoms. Moreover, modulating microglial activation to regulate ROS production has emerged as a novel avenue for developing innovative treatments for schizophrenia. (Figure presented.).

    Lignans are plant-derived compounds that act as partial estrogen agonists. Chondroitin sulfate proteoglycans (CSPGs) represent one of the major components of the extracellular matrix. Here we aimed to understand the role of sesamin (SES), a major lignan compound, in the biosynthesis and degradation of CSPGs in the mouse hippocampus because CSPGs play a key role in the regulation of cognitive functions through the promotion of adult neurogenesis. The expression of the pro-inflammatory cytokine interleukin-1β was decreased by SES administration in the hippocampus of lipopolysaccharide (LPS)-treated mice, a model of neuroinflammation-induced cognitive deficits. The expression of genes related to biosynthesis and degradation of CSPGs in the hippocampus of LPS-treated mice was both increased and decreased by SES administration. Further, the diffuse extracellular matrix labeling of CSPGs by Wisteria floribunda agglutinin (WFA) in the hippocampus of LPS-treated mice was increased by SES administration. The densities of neural stem cells, late transit-amplifying cells, and newborn-granule cells in the hippocampus of LPS-treated mice were also increased by SES administration. Moreover, SES-induced alterations in gene expression, WFA labeling, and adult neurogenesis in LPS-treated mice were more evident in the dorsal hippocampus (center of cognition) than in the ventral hippocampus (center of emotion). Neither LPS nor SES administration affected locomotor activity, anxiety-like behavior, and depression-related behavior. However, impairments in contextual memory and sensorimotor gating in LPS-treated mice were recovered by SES administration. Our results show that SES can promote adult hippocampal neurogenesis through the upregulation of CSPGs, which may alleviate cognitive deficits induced by neuroinflammation.

    BACKGROUND AND PURPOSE: Chondroitin sulfate proteoglycan (CSPG) constitutes the neurogenic niche in the hippocampus. The reduction of hippocampal neurogenesis is involved in aging-related cognitive decline and dementia. The purpose of this study is to find candidates that improve cognitive function by analyzing the effects of memantine (MEM), a therapeutic agent for Alzheimer's disease, on CSPG and adult hippocampal neurogenesis. EXPERIMENTAL APPROACH: The effects of MEM on neurogenesis-related cells and CSPG content were assessed in the hippocampus of middle-aged mice. The MEM-induced alterations in gene expressions of neurotrophins and enzymes associated with biosynthesis and degradation of CSPG in the hippocampus were also measured. The effects of MEM on cognitive function were estimated using a behavioral test battery. The same set of behavioral tests was applied to evaluate the effects of pharmacological depletion of CSPG in the hippocampus. KEY RESULTS: The densities of newborn granule cells and content of CSPG in the hippocampus were increased by MEM. The expression levels of the enzyme responsible for the biosynthesis CSPG were increased by MEM. The neurotrophin-related molecules were activated by MEM. Short- and long-term memory performance was improved by MEM. Pharmacological depletion of CSPG impairs the effects of MEM on cognitive improvement in middle-aged mice. CONCLUSIONS AND IMPLICATIONS: MEM regulates the biosynthesis and degradation of CSPG, which may underlie the improvement of cognitive function via the promotion of adult hippocampal neurogenesis. These results imply that CSPG-related enzymes may be potentially attractive candidates for the treatment of aging-related cognitive decline.

    Microglia, resident immune cells in the brain, are shown to mediate the crosstalk between psychological stress and depression. Interestingly, increasing evidence indicates that sex hormones, particularly estrogen, are involved in the regulation of immune system. In this study, we aimed to understand the potential effects of chronic social defeat stress (CSDS) and genistein (GEN), an estrogenic compound of the plant origin, on neuron-microglia interactions in the mouse hippocampus. The time spent in the avoidance zone in the social interaction test was increased by CSDS 1 day after the exposure, while the avoidance behavior returned to control levels 14 days after the CSDS exposure. Similar results were obtained from the elevated plus-maze test. However, the immobility time in the forced swim test was increased by CSDS 14 days after the exposure, and the depression-related behavior was in part alleviated by GEN. The numerical densities of microglia in the hippocampus were increased by CSDS, and they were decreased by GEN. The voxel densities of synaptic structures and synaptic puncta colocalized with microglia were decreased by CSDS, and they were increased by GEN. Neither CSDS nor GEN affected the gene expressions of major pro-inflammatory cytokines. Conversely, the expression levels of genes related to neurotrophic factors were decreased by CSDS, and they were partially reversed by GEN. These findings show that GEN may in part alleviate stress-related symptoms, and the effects of GEN may be associated with the modulation of neuron-microglia signaling via chemokines and neurotrophic factors in the hippocampus.

    中枢神経系のニューロンの一部には、細胞体の周囲にペリニューロナルネット (PNN) と呼ばれる細胞外基質の特殊構造が形成されている。我々は近年、記憶と情動の中枢である海馬神経回路を構成するGABAニューロンのサブクラスとPNNの分子多様性にフォーカスし、独自の形態学的研究に取り組んできた。これまでに、海馬のWFA陽性PNNはPV含有ニューロンの中でもBasket型に選択的に形成されていることや、Aggrecan陽性PNNはBasket型に加えてO-LM型・H-S型のPV含有ニューロンにも形成されていることを報告している。また、海馬のCat-315陽性PNNの発現レベルは加齢に伴って上昇し、抗認知症薬 (memantine) によって低下することを見出している。本研究会では、海馬のCCK含有ニューロンに形成されているPSA-NCAM陽性PNNについて、シナプス入力の特殊性や、拘束ストレスモデルと抗うつ薬 (fluoxetine) を用いた実験結果等を紹介し、PNNに関する議論を深める予定である。

    The perineuronal net (PNN), a specialized aggregate of the extracellular matrix, is considered to play an important role in regulation of neuronal plasticity. Since the late 1980s, a number of studies have documented that perineuronal nets (PNNs) are closely associated with parvalbumin-positive (PV+) GABAergic neurons. However, most studies have not taken into account the fact that not only PNNs but also PV+ GABAergic neurons are heterogeneous. We thus have engaged in the research of PNNs and PV+ GABAergic neurons in the hippocampus from the viewpoint of their heterogeneity, and have obtained several novel findings. First, the vast majority of PV+ basket cells were surrounded with Wisteria floribunda agglutinin-positive (WFA+) PNNs while PV+ axo-axonic cells, oriens-lacunosum-moleculare (O-LM) cells, and hippocampo-septal projection (H-S) cells were rarely enwrapped by WFA+ PNNs. By contrast, the majority of PV+ O-LM cells and H-S cells were enwrapped with aggrecan+ PNNs. These findings indicate that the molecular composition of PNNs in the hippocampus may differ in a subclass-specific manner. Next, the densities of GABAergic synapses were higher around PV+ bistratified cells enwrapped with aggrecan+ PNNs than around those without aggrecan+ PNNs. We thus have hypothesized that PNNs may affect the synaptic formation around PV+ GABAergic neurons in the hippocampus. Finally, the expression levels of Cat-315 epitope, human natural killer-1 glycan on aggrecan-based PNNs, were increased during normal brain ageing, and an open-channel blocker of NMDA receptor, memantine, counteracted the age-related changes in Cat-315 epitope. I will show the results of our ongoing projects at the symposium, and discuss the future direction of PNN research.

    中枢神経系にさまざまな糖鎖が存在することは古くから知られていた.大脳皮質や海馬のGABAニューロンの周囲にペリニューロナルネットと呼ばれる糖タンパク質からなる特殊な細胞外マトリックス構造が形成されていることについては,多くの優れた解剖学的研究がある.しかしながら,その機能の研究は遅々として進まず,ペリニューロナルネットが神経可塑性の制限や精神神経疾患の病態に関わっていることがわかってきたのは,比較的最近のことである.また,神経発生において糖鎖が重要な役割を果たしていることについては研究の蓄積がある一方で,成体における神経新生を維持する微小環境(ニッチ)としての細胞外マトリックスに関する研究は少ない.我々は近年,海馬のペリニューロナルネットと神経新生のニッチに着目し,糖鎖の機能を明らかにすることを目的とする基礎研究に取り組んできた.本稿では,高次脳機能の制御基盤としての海馬の糖鎖の機能について,概説を行う.(著者抄録)

    複雑なモダリティを有する顕微鏡画像の定量化には膨大な労力を要する.この点で,研究者自身によるプログラミングは,研究の効率化に貢献するゲームチェンジャーとも言えるものであるが,あまり普及していない.一方,小学校では,2020年度からプログラミング学習が必修化されている.プログラミングを学ぶということは,難解なプログラミング言語を学ぶことだと思われがちだが,小学校におけるプログラミング学習の主な目的は,物事を論理的に考える「プログラミング的思考」を身につけさせることにある.近年の画像解析ソフトの多くにプログラミング言語が搭載されている.顕微鏡画像の解析に研究者が自らプログラミングを導入することには多くのメリットがあり,論文の生産性を向上させることにとどまらず,研究の質的な向上にもつながる.本稿によって,研究者自身によるプログラミングの有用性がより理解され,研究力の向上に役立つことが期待される.(著者抄録)

    中枢神経系の一部のニューロンは、網状の特殊な細胞外マトリックスによって覆われている。この構造はペリニューロナルネット(PNN)と呼ばれ、直鎖状の糖鎖を基本骨格に持つコンドロイチン硫酸プロテオグリカンに加え、ヒアルロン酸、フィブロネクチン、コラーゲン、ラミニンなどからなる。今回、認知と情動を担う神経回路の老化に伴う構造的・機能的変化をPNNの視点から明らかにすることを目指し、マウス海馬のPNN発現強度の長軸に沿った差異と加齢変化について組織化学的な解析を行った。結果、背側海馬のCA1領域とCA3領域の錐体細胞層におけるWFA陽性PNNの発現強度は若年成獣マウスに比べて加齢マウスで減弱していることが明らかになった。腹側海馬の各領域におけるWFA陽性PNNの発現強度は全ての層で若年成獣マウスと加齢マウスとの差異は認めなかった。

    ECS(電気痙攣療法の動物実験モデル)によるグリア細胞の形態学的ダイナミクスの解析から、精神疾患におけるグリア細胞の役割について検討した。成熟オスマウスに電気痙攣刺激を与え、約10秒間持続する強直間代痙攣を確認した。マウスは急性ECS群、慢性ECS群、コントロール群の3群に分けた。コントロール群のマウス海馬では、Iba1陽性ミクログリアは細く高度に分枝した突起と小型の細胞体を示したが、急性ECSでは大部分の細い突起が消失した。慢性ECS群では、同様の変化は1ヵ月後にも認めた。Iba1陽性ミクログリアの突起密度は、急性ECS施行24時間後に有意に低下したが、1ヵ月後には回復した。慢性ECS群では、1ヵ月経っても、突起密度は有意に低下していた。ミクログリアとアストロサイトの空間分布密度はECS施行による変化は認めなかった。

    Broca-桂指数,比体重,BMI,Rohrer指数,ベン指数の五つの体格指数の,青年男子の肥満指標としての妥当性について,1)体重とは高い相関を持つが身長とはできるだけ独立であること,2)標準体重による肥満度と高い相関を持つこと,3)各体格指数の判定基準で過体重の有無を正しく判定できること,の三条件を設定し,九州大学医学部男子学生の1961年~1992年の身体計測データを用いて評価を行った。BMIはこの三条件をいずれも満たしており,優れた肥満指標であることが示された。ベン指数は身長に対して独立であるだけでなく体重やObese rateとも高い相関を示すことが示されたが,この指数での確立された過体重の判定基準がないため,条件3)に関する検討を行うことができなかった。比体重,Rohrer指数,Broca-桂指数は身長に対して独立でなく,今回の研究対象における肥満指標としては適当でない

    がん細胞が産生するサイトカイン・ケモカイン・増殖因子は全身を隈なくめぐり、様々な臓器・組織に作用する。本研究で我々は、がん由来の分子が脳と相互作用する「場」として、オリゴデンドロサイトを定義し、その機能デコーディングに取り組み、がんによる認知機能障害が起こるメカニズムを明らかにすることを目指す。また、がん由来分子の解析と認知機能の行動学的解析に、ダブルパッチクランプを用いたオリゴデンドロサイト機能のデコーディングを組み合わせて、脳・身体相互作用の機序にアプローチする。これらから、身体のがんと脳のグリアの連関をターゲットにした新たな研究フィールドを創造する。

    若年者に比べて中高年者の方がうつ病の発症率が高い理由は、現在まで不明である。また、中高年者のうつ病の治療では、基本的に若年者と同様の薬物療法が行われているが、難治化する症例も少なくなく、中高年者のうつ病の新たな理解と治療戦略が求められている。本研究では、ストレスに対する「抵抗性」と「回復力」からなるレジリエンスの概念に基づき、 うつ病と発症と回復のプロセスにおけるニッチの役割にアプローチする。さらに、その創造性は、海馬のオリゴデンドロサイト新生のニッチの改変によってストレスレジリエンスを回復させる実験によって、中高年者のうつ病に最適化した新たな治療戦略の創出を目指す。

    近年、腸内細菌叢が脳機能に関わっていることが認識され、研究が急速に広まっている。興味深いことに、運動に伴って腸内細菌叢の変化が起こることも報告されている。我々は、豊かな環境による成体海馬神経新生の促進には、歯状回のコンドロイチン硫酸の増加が関わっていることを最近の報告で明らかにしたが、そのメカニズムは未解明のままである。これらを踏まえ我々は、マウスを用いた動物実験により、「運動は腸内細菌叢の中間代謝物を介して間接的に神経新生を促進する」との仮説に基づく分野横断的な検討に取り組む。本研究によって、運動による成体海馬神経新生の促進に腸内細菌叢が関与するメカニズムが明らかになると考えられる。
