学部教育では、口腔生化学および歯科薬理学を担当している。  大学院教育および研究活動としては、主に、エピゲノム制御の観点から、周産期や各ライフステージにおける栄養素摂取と疾患罹患感受性の分子基盤に焦点を当てた研究を拡げ、がんを含めた生活習慣病や免疫疾患におけるエピゲノム制御機構、予防や治療に向けた栄養学的アプローチに取り組んでいる。さらに、各種疾患の病態や治療におけるエピゲノム制御機構、個体の代謝制御におけるTCA回路の役割について、その分子基盤を明らかにすることを目指して活動を行っている。

    Aims: Itaconate (IA) is synthesized in the citric acid cycle via cis-aconitate decarboxylase (ACOD1); however, its biological significance in cancer remains incompletely understood. In previous studies, 4-octyl itaconate (OI) was used as a membrane-permeable form of IA, but little detailed verification of the difference in biological activities between IA and OI exists. Here, we investigated the direct effects of IA and OI on melanoma. Results: The proliferation of melanoma cells treated with OI was significantly suppressed in vitro, and our transcriptomic analysis revealed drastic changes in the expression of glutathione metabolism-related genes in OI-treated cells. Indeed, OI treatment decreased intracellular glutathione levels, followed by increased production of reactive oxygen species and expression of γH2AX, a marker of DNA damage, and β-galactosidase, a marker of cellular senescence. We further showed that the mitochondrial respiratory capacity in B16 cells was significantly decreased by OI treatment. OI administration also suppressed the growth of B16 tumor transplants in vivo, and the expression of γH2AX was increased in tumor tissues of OI-treated mice. In addition, minimal effects of OI treatment were observed in melanocytes and normal tissues. We also proved that not only exogenous IA, which enters intracellularly, but also endogenous IA has little effect on melanoma proliferation activity, via an investigation using Acod1-overexpressing transfectants and Acod1-deficient mice. Conclusion: This work revealed that OI disrupts the antioxidant system via the collapse of glutathione metabolism and inhibits cancer cell proliferation. Antioxid. Redox Signal. 42, 547-565.

    After menopause, a decline in ovarian function leads to various physical and psychological changes, potentially resulting in a range of pathological conditions, including abnormalities in energy metabolism. In recent years, environmental enrichment, which is characterized by positive and comfortable eustress, has been shown to improve various physiological and pathological conditions. This study investigated the effects of environmental factors on energy metabolism in a menopause model using an ovariectomized (OVX) mouse model. Wild-type female mice (8-week-old) were subjected to OVX or a sham operation and maintained under standard condition (SC), enriched environment (EE), or isolated (IS) condition for 4 weeks. OVX led to weight gain and disruption of circadian rhythms, along with changes in various metabolic parameters influenced by differences in housing environments; i.e., EE improved metabolic parameters, but IS deteriorated them. Physical activity and social interaction were factors that determined these differences. Menopause is usually a significant transitional period in a woman's life, and changes in the social environment during this period can contribute to a diverse range of physical and psychological symptoms. Consequently, when implementing interventions to alleviate menopause-related pathological conditions, not only physical symptoms but also the social context should be carefully considered.

    Postmenopausal women experience bone loss and weight gain. To date, crosstalk between estrogen receptor signals and nuclear factor-κB (NF-κB) has been reported, and estrogen depletion enhances bone resorption by osteoclasts via NF-κB activation. However, it is unclear when and in which tissues NF-κB is activated after menopause, and how NF-κB acts as a common signaling molecule for postmenopausal weight gain and bone loss. Therefore, we examined the role of NF-κB in bone and energy metabolism following menopause. NF-κB reporter mice, which can be used to measure NF-κB activation in vivo, were ovariectomized (OVX) and the luminescence intensity after OVX increased in the metaphyses of the long bones and perigonadal white adipose tissue, but not in the other tissues. OVX was performed on wild-type (WT) and p65 mutant knock-in (S534A) mice, whose mutation enhances the transcriptional activity of NF-κB. Weight gain with worsening glucose tolerance was significant in S534A mice after OVX compared with those of WT mice. The bone density of the sham group in WT or S534A mice did not change, whereas in the S534A-OVX group it significantly decreased due to the suppression of bone formation and increase in bone marrow adipocytes. Disulfiram, an anti-alcoholic drug, suppressed OVX-induced activation of NF-κB in the metaphyses of long bones and white adipose tissue (WAT), as well as weight gain and bone loss. Overall, the activation of NF-κB in the metaphyses of long bones and WAT after OVX regulates post-OVX weight gain and bone loss.

    Purpose of Review: According to the Developmental Origins of Health and Disease (DOHaD) theory, nutritional status in utero can permanently change body structure, function, and metabolism and eventually regulate the susceptibilities toward various diseases in later life. This review aims to examine and summarize the findings of previous reports, revealing how maternal nutrition during pregnancy and lactation affects oral health in offspring. Recent Findings: A limited number of studies have found an evident relationship between maternal nutrition and children’s oral health. Some evidence suggests that maternal nutrients such as sucrose, proteins, minerals, and vitamins are associated with oral and maxillofacial development, including bone and tooth size, and the risk of dental caries, periodontal disease, and oral cancer in offspring. Summary: The lessons of DOHaD theory from the perspective of the oral health of offspring reveal how in utero nutrients affect not only oral health but also total health throughout life. Oral disease and dysfunction can be prevented in the next generation by improving maternal nutrition during pregnancy and lactation periods.

    Salivary glands are physiologically orchestrated by the coordinated balance between cell differentiation, proliferation, apoptosis, and interactions between epithelial, mesenchymal endothelial, and neuronal cells, and they are frequent sites of manifestations of Sjögren's syndrome (SS) or IgG4-related disease (IgG4-RD). However, little is known about salivary gland homeostasis and its involvement in those diseases. Inhibitor of DNA binding/differentiation 4 (Id4) is an Id protein involved in the transcriptional control of many biological events, including differentiation. Studies of Id4-deficient mice revealed that Id4-deficient submandibular glands were smaller and exhibited accelerated differentiation, compared with those from wild-type littermates. In addition, dry mouth symptoms and Th17 expansion in splenocytes were also observed in the absence of Id4. Furthermore, Id4 levels in the salivary glands of patients with IgG4-RD, but not SS, were significantly decreased compared with those of healthy controls. miRNA-mRNA integrated analysis demonstrated that miR-486-5p was upregulated in IgG4-RD patients and that it might regulate Id4 in the lesion sites. Together, these results provide evidence for the inhibitory role of Id4 in salivary differentiation, and a critical association between Id4 downregulation and IgG4-RD.

    本研究では、妊娠中に高脂肪高ショ糖食や各種ビタミン欠乏食・過剰食を摂取した母親マウスの産仔について、網羅的解析により得られる DNAメチル化や遺伝子発現変化を統合的に解析することで、疾患感受性遺伝子の同定を行い、それら遺伝子についての機能解析を行う。またその結果に基づき、妊娠母体あるいは臍帯血中の当該栄養素と産子の代謝パラメータとの相関について、ヒトでの検証(出生コホートを用いた解析)を行う。

    本邦では不育症は5%と欧米より高頻度であり、不育症の克服は少子化対策に直結する。25原因不明患者に対し、欧州のRCTで流産回数が多く性器出血を伴う患者へのプロゲステロン(P4)腟錠の有効性が報告されたが、妊娠年齢の高齢化が進んでいる本邦において流産を繰り返す前に治療効果のある患者を選び出し流産を予防する戦略を開発することが重要である。本研究ではP4シグナルに関連する遺伝子多型が不育症病態におけるP4感受性に関わっているとの仮説のもと、P4応答群と不応群とでゲノムワイド関連解析(GWAS)を用いて遺伝子多型を同定する。これらの成果はヒト妊娠維持メカニズムの新たな解明に発展できる。

    生活習慣病の多くは肥満に起因する。一旦発症すると治療効果が得難いことから、その制圧には肥満の予防策を講じることが重要である。ところが今、肥満予防には、生後の取り組みに加えて新たな視点が必要となっている。
    申請者は現在までに、動物実験で妊娠母体の栄養状態によって次世代の仔に肥満が現れる事を認め、肥満の仔で生じているエピゲノム異常の共通項を肥満感受性エピゲノムとした。そこで本研究では、細胞や動物を用いた生物学実験とヒト疫学研究を組み合わせて母体の栄養(因)と児の肥満発症素因(果)の関係について検証し、肥満感受性や肥満症多様性を決定する世代を超えるエピゲノム変化の同定と検証を行う。

    閉経後の女性では、骨密度の低下と共に脂肪の増加による体重増加が起こる。我々は卵巣摘出マウスモデルにNF-κB阻害剤を投与すると、骨量の減少が抑制されるだけでなく、体重増加も抑制することを報告した。以上のような学術的背景と独自の研究成果に基づき、骨量減少と体重増加に重要な女性ホルモンの欠乏で起きるNF-κBシグナルの活性化組織・細胞を同定し、特異的にNF-κBシグナルを抑制できれば、閉経後の骨量減少とエネルギー代謝の悪化に対する革新的な治療法や予防法が確立できると考えた。本研究成果は、新たな骨代謝とエネルギー代謝間のクロストークを提唱するだけでなく、国民のQOLの延伸に貢献できる。
