平成2年3月九州大学医学部医学科卒業。第2外科に入局し2年間の臨床研修の後、平成4年4月九州大学大学院医学系研究科に入学(外科系専攻、病理学)。平成8年3月「炎症性腹部大動脈瘤発生におけるヒトサイトメガロウイルス感染の意義(主論文:Laboratory Investigation 74; 723-736,1996)」にて学位取得。大学院では主に下に示す研究内容についての研究を進めた。同4月第2外科医員。1年半余血管外科の臨床および遺伝子治療に関する研究に携わった後、平成9年11月より英国ロンドン大学インペリアルカレッジ医学部National Heart and Lung Institute遺伝子治療部門(主任教授:prof. Eric Alton)にて、Cystic Fibrosisに関する研究。日本において新しく開発された遺伝子治療用ベクターの性能試験を行い、このセンダイウイルスベクターが既存のベクターと比較して圧倒的な遺伝子導入効率を示すことを明らかにした(英国Wellcome Trust特別海外研究員:Nature Biotechnology,18: 970-973, 2000、書評:News & Views in Nature Genetics 26:11, 2000、Molecular Medicine Today 6: 460, 2000)。平成11年5月より九州大学医学部附属病院病理部助手として復学し、平成15年6月九州大学病院病理部講師、平成16年610月九州大学大学院医学研究院基礎医学部門助教授(病理病態学)、の後、平成18年4月より千葉大学大学院医学研究院・教授(寄附講座、九州大学大学院医学研究院・特任教授併任)。 平成21年10月より九州大学大学院薬学研究院・教授(寄附講座担当)、現在に至る。 1.所属学会 1) 海外: American Heart Association (Fellow, Premium Professional Member)、 American Society of Gene Therapy (International Active Member)、FASEB/American Association of Immunologists、他 2) 国内:日本遺伝子治療学会(理事、評議員) 、日本動脈硬化学会(評議員、広報委員会幹事、学術委員会委員)、日本脈管学会(評議員、英文誌Editorial Board)、日本血管生物医学会(評議員)、日本病理学会(英文誌Editorial Board)、外科分子細胞治療研究会、日本外科学会、日本血管外科学会、日本癌治療学会、他 2.英文誌編集委員(Editorial Board Member): Human Gene Therapy, The Open Gene Therapy Journal, Annals of Vascular Diseases, Pathology International 3. 主な研究テーマ 1)動脈硬化、血管新生の分子病理学的研究。血管平滑筋細胞、血管内皮細胞の特性、細胞周期制御遺伝子、分化誘導因子、血管新生因子の生体内作用機構に関する研究。 2)遺伝子治療に関する研究。循環器疾患、先天性呼吸器疾患、悪性疾患などに対する、センダイウイルスベクター系およびレンチウイルスベクター系を用いた遺伝子治療系の確立。 3)樹状細胞・NK細胞の分子生物学的研究、抗腫瘍免疫誘導機構に関する研究。 4.教育 大学院学生教育:研究・実験・論文作成指導 研究成果は国内外の主要学会、主要欧米紙にて積極的に発表している。 また臨床応用可能な遺伝子治療系の確立を目指し、学内外、国外の研究者と積極的な共同研究を推進している。 5. 学会賞、他 第8回日本遺伝子治療学会 学会賞、The Wellcome trust Fellowship

    <p>2014年11月に医薬品医療機器等法(改正薬事法)が施行、再生医療等製品を対象として条件及び期限付製造販売承認制度(いわゆる早期承認制度)が導入された。これは患者にとって画期的治療技術へのアクセスのハードルを下げるための規制緩和として期待され、更に遺伝子治療・細胞治療(法的区分においては再生医療等製品)の開発を手掛けるスタートアップは、これまで小規模な臨床試験だけで早くから販売に到達可能となることを期待して来た。その後9年が経過し複数の製品が早期承認されたものの、その売上高は限定的であり、各社の収益に貢献しているとは言い難い。結局のところ、(1)有効性が「推定されている(確定されていない)」に留まっている再生医療等製品について、医療現場が積極的に使用する動機になりにくい、(2)早期承認レベルのデータで、特に欧米で承認を受けることは概ね不可能である、などの理由が想定される。そのため、最近では製薬企業を中心に、「一定規模の比較試験で明確な有効性を証明し、医療現場で使ってもらう素地を固めることが、結局は一番近道ではないか」という主張も聞かれるようになって来た。以上のように、現在早期承認制度については、議論を残したまま過渡期に入っている。一方で一定規模の比較試験を組むことに困難が伴う場合、つまり自家細胞の加工を要する製品や、遺伝性疾患などの極希少疾患を対象とする製品の場合、早期承認制度は依然として重要な制度である。これらはまさに遺伝子治療・再生医療にあてはまる場合が多いが、ではそのような場合にはどのようなデータを以て「有効性を推定」することが可能になるだろうか?本講演では、我が国における以上の経緯を整理した上で、2017年に米国FDAで制定されたRMAT指定制度、そしてそのドラフトガイドラインの記載内容と我が国での実例との比較をすることにより、早期承認制度のあるべき姿について議論したい。</p>

    Mitochondrial cloning is a promising approach to achieve homoplasmic mitochondrial DNA (mtDNA) mutations. We previously developed a microfluidic device that performs single mitochondrion transfer from a mtDNA-intact cell to a mtDNA-less (ρ0) cell by promoting cytoplasmic connection through a microtunnel between them. In the present study, we described a method for generating transmitochondrial cybrids using the microfluidic device. After achieving mitochondrial transfer between HeLa cells and thymidine kinase-deficient ρ0143B cells using the microfluidic device, selective culture was carried out using a pyruvate and uridine (PU)-absent and 5-bromo-2′-deoxyuridine-supplemented culture medium. The resulting cells contained HeLa mtDNA and 143B nuclei, but both 143B mtDNA and HeLa nuclei were absent in these cells. Additionally, these cells showed lower lactate production than parent ρ0143B cells and disappearance of PU auxotrophy for cell growth. These results suggest successful generation of transmitochondrial cybrids using the microfluidic device. Furthermore, we succeeded in selective harvest of generated transmitochondrial cybrids under a PU-supplemented condition by removing unfused ρ0 cells with puromycin-based selection in the microfluidic device.

    Memory-like NK(ML-NK)は、難治性急性骨髄性白血病AMLでの臨床試験において驚異的な治療効果を示すことから、いわゆるGVL効果の本態ではないかと注目されている。我々はこれらの動きとは全く独立に、固形がんへ極めて高い抗腫瘍活性を示す、「他家」NK細胞様製剤 GAIA-102を開発、固形腫瘍を対象に3つの治験を進めており、複数の症例で臨床的POCを示唆するデータを得ている。本研究では「ML-NK-like NK」とも呼び得るGAIA-102の分化系譜を明らかとし、なぜNK細胞にはこのようなバラエティに富んだスペクトラムがあるのかという免疫学の根源的な疑問に肉薄する事を目的とする。

    本研究は、神経芽腫に対する高活性NK細胞製剤(開発コード:GAIA-102)の抗腫瘍効果を強化するべく行われる前臨床試験である。GAIA-102は、他のNK細胞製剤と比較して歴然に優れた、固形がんへの高い浸潤能と傷害活性を示す。抗PD-1抗体を併用することで、より強力な免疫細胞誘導を生じると考えられ、神経芽腫に対するGAIA-102と抗PD-1抗体併用療法の有効性とメカニズムについて明らかにする。

    本研究は神経芽腫に対する革新的な免疫治療の開発を目的としている。これまでの研究で神経芽腫表面に発現するジシアロガングリシド(以下GD2)に対する抗体を搭載した間葉系幹細胞(GD2-MSCs)を開発し、神経芽腫自然発生モデルマウスにおいて高効率に神経芽腫にhomingし、なおかつADCC活性を誘導するメインエフェクターであるNatural Killer(以下NK)細胞の腫瘍内への誘導能を確認した。この研究を更に発展させるべく、共同研究施設で開発された高活性NK細胞様CD3陰性細胞をGD2-MSCと併用することを立案し、より効率的な養子免疫治療を実現する。

    移植グラフトを目指したミニチュアヒト肝臓の創成を目指して医工連携研究を行う。ヒト初代培養細胞やヒトiPS細胞から得る機能性ヒト細胞(肝細胞・胆管細胞)、臓器構築のscaffoldとなる内皮化されたラット脱細胞鋳型肝臓を組み合わせて臓器工学システムを構築する。更に、極小型人工肺モジュールを用いた高酸素供給が可能なex-vivo培養回路を作製し、ミニチュアヒト肝臓を灌流培養して、高い肝組織構築(肝小葉・類洞形成)を実現する。

    We have selected NKp46, 2B4, and NKp30 as target molecules on GAIA-102 and conducted selection of monobodies that specifically bind to these target molecules. All clones were confirmed to bind to GAIA-102. In addition, we designed scFv based on known antibody sequences and fusion proteins with monobody for functional evaluation of GAIA-102 binding monobodies. These research results provide valuable data for the development of advanced "antibody-cell complex" technology as intended and will now move on to the practical development steps for standardization and manufacturing towards clinical development.

    2)細胞の包埋に用いる『機能性ヘパリン導入ゼラチンゲル基剤』を最適化する。
    3)臓器構築のscaffoldとなる『内皮化されたラット脱細胞鋳型肝臓』を組み合わせて臓器工学システムを構築する。ミニチュアヒト肝臓を灌流培養して、高い肝組織構築 (肝小葉・類洞形成)を実現する。

    TRの適正・安全・迅速な推進は、世界トップレベルといわれる我が国の医療水準を今後も維持しかつ向上させるためには必須の課題である。九州大学では、ウィルス遺伝子組換え技術を基盤とした純国産バイオ医薬、及び我が国が世界に誇る工学技術を基盤とした情報型先進治療システムを中心に先進的な医療技術の開発に取り組んできた。本プロジェクトにおいては、これらを中心とした独創的かつ画期的な国産医療技術の迅速な実用化・産業化を目指したTR拠点の形成を目指す。
