リソソームは真核細胞において普遍的に存在する酸性の内部環境を有する細胞内小器官であり、ユビキチン・プロテアソーム系とともにエンドサイトーシスおよびオートファジーによる細胞内外の物質分解に主要な役割を果たしている。このような細胞内の主要なタンパク質分解システム系としてのリソソームの機能は、その内部に含まれる多くの加水分解酵素に加え、近年、ユビキチン様タンパク質による共有結合システムを必要とするオートファジーを介した非選択的な大規模なタンパク質分解システムの研究により更なる進展を遂げつつある。生体の恒常性はタンパク質の合成と分解の絶妙なバランスの基に維持されているため、このバランスの崩壊は細胞及び生体に重篤な障害をもたらすことになる。実際に、このようなリソソーム系のタンパク質分解システムの破綻が原因で多くの神経及び筋疾患を引き起こすことが報告されている。また、タンパク質分解システムに加え、リソソームは病原性細菌の除去、自然免疫・獲得免疫による抗原提示、細胞死および損傷した細胞膜の修復等様々な生命現象にも重要な役割を担っていることが明らかにされつつある。このようなリソソームの機能発現は様々なオルガネラ(エンドソーム、ファゴソーム、細胞膜など)との密接なコミュニケーションのもとに成り立っている。それ故、このようなリソソームの機能発現には、その内部に含まれる加水分解酵素に加え、リソソームを形成・維持する膜タンパク質が重要な役割を果たしていると考えられる。このような背景を踏まえ、リソソーム膜タンパク質の機能解明を通して、リソソームの生理機能の解明を目指している。

    Autophagy is a degradation pathway that delivers cytoplasmic materials to lysosomes via doubled-membrane organelles termed autophagosomes. Although autophagy can be induced by nutrient deprivation or the absence of growth factors, it also occurs at basal levels in most tissues and contributes to the routine turnover of cytoplasmic components. Here we show that a small GTPase, Rab32, is a novel protein required for the formation of autophagic vacuoles. We found that overexpression of the wild-type or GTP-bound form of human Rab32 expressed in HeLa and COS cells is predominantly localized to the endoplasmic reticulum (ER) and overexpression induces the formation of autophagic vacuoles containing an autophagosome marker protein LC3, the ER-resident protein calnexin and endosomal/lysosomal membrane protein LAMP-2, even under nutrient-rich conditions. The recruitment of Rab32 to the ER membrane was necessary for autophagic vacuoles formation, thereby suggesting involvement of the ER as a source of autophagosome membranes. Additionally, the failure of autophagic vacuole formation and the development of ubiquitin-containing proteins accumulating aggresomes in cells expressing the GDP-bound or GTP/GDP-free form of Rab32 further suggest the requirement of the normal function of Rab32 in autophagy. Indeed, treatment of these cells with autophagy-stimulating agents could not induce the formation of autophagic vacuoles. Moreover, our results show that the knockdown of Rab32 by the expression of Rab32-specific siRNA leads to the formation of aggresome-like structures containing ubiquitinated proteins and p62 under normal growth conditions. Thus, our data imply the physiological importance of Rab32 in the cellular clearance of aggregated proteins by basal constitutive autophagy. We conclude that Rab32 plays pivotal roles in not only the formation of autophagic vacuoles, but also the clearance of ubiquitin-containing protein aggregates by autophagy under constitutive conditions.
