    Protein kinase Cα (PKCα) has an important role in directed cell migration. After a mechanical wounding, PKCα rapidly accumulates at cell edges adjacent to the wounded cell and regulates cell migration. However, the proteins downstream of PKCα that mediate directed signaling remain unknown. In this study, we examined the spatiotemporal dynamics of PKCα, PIP3, Moesin, Cdc42, MARCKS, Scribble, and Arf6 before directed migration. After wounding, PIP3, Moesin, and Cdc42 accumulated at the cell edge near the wounded cells later than PKCα. In contrast, MARCKS moved away from the plasma membrane without polarization, and Scribble and Arf6 exhibited no significant translocation. The inhibition of PIP3 suppressed the accumulation of Moesin and Cdc42, suggesting that PIP3 regulates Moesin and Cdc42. In particular, the inhibition of PKCα completely inhibited the translocation of all factors, indicating that PKCα is a central regulator in early signaling after wounding and before directional migration.

    Pulmonary surfactant is essential for maintaining proper lung function. Alveolar epithelial type II (AE2) cells secrete surfactants via lamellar bodies (LBs). In tidal loading during each breath, the physiological cyclic stretching of AE2 cells promotes surfactant secretion. Excessive stretching inhibits surfactant secretion, which is considered to contribute to the development of lung damage. However, its precise mechanism remains unknown. This study tested whether actin polymerization and intracellular transport are required for pulmonary surfactant secretion and the association of actin polymerization and transport in identical human AE2-derived A549 cells using live-cell imaging, not in the bulk cells population. We found that overstretching approximately doubled actin polymerization into filaments (F-actin) and suppressed LB secretion by half in the fluorescent area ratio, compared with physiological stretching (F-actin: 1.495 vs 0.643 (P < 0.01); LB: 0.739 vs 0.332 (P < 0.01)). An inhibitor of actin polymerization increased LB secretion. Intracellular tracking using fluorescent particles revealed that cyclic stretching shifted the particle motion perpendicularly to the direction of stretching according to the orientation of the F-actin (proportion of perpendicular axis motion prior particle: 0h 40.12 % vs 2h 63.13 % (P < 0.01)), and particle motion was restricted over time in the cells subjected to overstretching, indicating that overstretching regulates intracellular transport dynamics (proportion of stop motion particle: 0h 1.01 % vs 2h 11.04 % (P < 0.01)). These findings suggest that overstretching changes secretion through the cytoskeleton: overstretching AE2 cells inhibits pulmonary surfactant secretion, at least through accelerating actin polymerization and decreasing intracellular trafficking, and the change in actin orientation would modulate intracellular trafficking.

    The intracellular distribution of phosphatase and tensin homolog (PTEN) is closely related to directed cell migration. In single cells, PTEN accumulates at the rear of the cell before and during directed migration; however, the spatiotemporal distribution of PTEN in confluent cell monolayers, particularly before directed migration, remains unclear. In this study, we wounded a cell in confluent fetal rat skin keratinocytes (FRSKs) and examined the dynamics of PTEN in the cells adjacent to the wounded cell. In contrast to single-cell migration, we found that PTEN translocated to the nucleus before the beginning of directed migration. This nuclear translocation of PTEN did not occur in disconnected cells, and it was also suppressed by importin-β inhibitor and actin inhibitor. When the nuclear localization of PTEN was inhibited by an importin-β inhibitor, cell elongation in the direction of migration was also significantly inhibited. Our results indicate that PTEN translocation is induced by the disruption of cell–cell adhesion and requires the involvement of importin-β and actin cytoskeleton signaling. In addition, phosphatidylinositol 3,4,5-triphosphate (PIP3) may regulate PTEN distribution through its localized accumulation at the cell edge. Our findings suggest that the translocation of PTEN is crucial for directed cell migration and for responding to mechanical environmental changes in confluent cell monolayers.

    <p>Directional migration of eukaryotic cells results from an interplay between cell motility and environmental cues such as stiffness, roughness, and microtopological features of substrates. While the effects of aligned grooves on the substrate lithographically designed on active cell migration are well documented, the impact of unorganized and wrinkled substrates remains poorly understood. We investigated how microscale wrinkles on gelatin substrates regulate two-dimensional (2D) migration of immortalized human mesenchymal stem cells (iMSCs). We showed that there exists an optimal wrinkle size to enhance iMSC migration, where the iMSCs exhibited larger travel distance with moderate wrinkle size (30 μm) than shorter or larger wrinkle sizes (10 μm and 50 μm). These findings reveal a size-dependent effect of wrinkles on iMSC migration and provide insights into designing biomaterials to control stem cell behavior in regenerative medicine applications.</p>

    <p>The fluidity of endothelial cell (EC) membranes, consisting of a lipid bilayer and heterogeneous multicomponent, undergoes alterations in response to shear stress. Although localized variations in membrane fluidity are assumed to induce region-specific signal transduction by modulating membrane protein dynamics on almost planar bilayers, membrane protein diffusion under shear stress remains uncertain. Hence, this study aimed to quantify membrane protein diffusion on ECs under fluid shear stress. We used the photochromic fluorescent protein Dronpa to tag a glycosylphosphatidylinositol-anchored protein (GPI-AP), which diffuses across the outer membranes, and quantified its surface diffusion based on the spatiotemporal distribution of Dronpa-Green-labeled GPI-AP (DGGPI-AP) on EC membranes. We developed an experimental platform to measure the GPI-AP surface diffusion under fluid shear stress and quantified the diffusion coefficient of GPI-AP in two distinct membrane regions: upstream and downstream relative to the direction of fluid flow. Our experimental results showed that there were not statistically significant differences in GPI-AP diffusion on EC membranes between the time points or between the upstream and downstream regions of ECs for at least 15 minutes under shear stress. Our developed methodology and experimental results will be useful to understand a relationship between the membrane protein diffusion and shear-induced cellular processes.</p>

    <p>がん細胞は生体内において接着環境の弾性率を上げてしまうことはよく知られている。そのため、がん組織は正常な組織と比較して硬い構造を有しており、乳がんの触診はこの特性を応用した簡便ながん診断法である。正常肺組織のヤング率は1〜5kPaであるのに対し、線維化組織や肺腫瘍組織は30倍程度硬い。しかしながら、肺は微視的には様々な組織から構成されており、腫瘍中のがん細胞は浸潤の際に硬いコラーゲン含有繊維に接触する。</p><p>本研究では、ヒト肺胞基底上皮腺癌細胞A549の集団遊走挙動に及ぼす基材弾性率の影響を調べた。架橋剤の比率を調整することにより弾性率の異なる(1.4、3.4、18.3MPa)ポリジメチルシロキサン(PDMS)基板を作製し、スクラッチアッセイにより、PDMS基板上および7.7 GPaガラス上でのA549細胞の遊走距離と遊走軌跡を調べた。その結果、A549細胞の18.3MPaでの移動範囲は、1.4MPaおよび3.4MPaでの移動範囲よりも有意に高かった。当日は細胞遊走の直進性についても併せて検討する。</p>

    脳皮質スライス標本を材料に、ニューロンを刺激することでその近傍の細動脈にみられる反応が、酸素濃度によってどのように変化するか実験した。3~4週齢の雄性Wistarラット16匹から脳皮質スライス標本を調製した。その培養液中の酸素濃度は20~95%の範囲で変化させた。脳皮質中のII/III層の錐体細胞を同定し、記録電極をその錐体細胞内に、刺激電極をIV層に設置した。トレイン刺激(20Hz、350ms)を0.75Hz周期で5回繰り返すことで興奮性シナプス後場電位を惹起させた。その結果、酸素濃度を95%に保った場合、錐体細胞を刺激することでその近傍における細動脈の径は縮小することが判明した。それと同条件で酸素濃度のみ20%とした時には、逆に細動脈径は拡張した。以上から、海馬のニューロンについて既に報告されていたのと同様に、皮質ニューロンでも近傍血管の反応の方向性は酸素濃度によって逆転することが示唆された。

    <p>The Young’s modulus of normal lung tissues ranges from 1 to 5 kPa, whereas fibrotic or lung tumor tissues can be ~30 times stiffer. However, the lung parenchyma microscopically consists of various tissues, and cancer cells in tumors contact stiff collagen-containing fibers during invasion. In this study, we investigated the effect of stiff substrates on A549 cell migration. We fabricated stiff polydimethylsiloxane (PDMS) substrates with different stiffness levels (1.4, 3.4, and 18.3 MPa) by adjusting the cross-linking agent ratio. We examined the distance and the trajectory orientation of A549 cell migration on PDMS substrates and much stiffer 7.7 GPa glass with scratch-wound assays. A549 cells exhibited a collective sheet-like migration pattern toward the wounded area on stiff substrates. The migration range at 18.3 MPa was significantly higher than at 1.4 and 3.4 MPa, and that on glass was significantly higher than that at 18.3 MPa. The cell trajectory orientation on stiff PDMS substrates gradually increased and became constant, whereas that on glass was constant from the initial migration. Our results revealed that stiff substrates affect A549 cells migration in this range.</p>

    <p>The brain requires O<sub>2</sub> and nutrition to keep functioning. It is known that O<sub>2</sub> concentration converts the polarity of vessel response near the activated neuron in the hippocampus. However, it remains unclear whether O<sub>2</sub> concentration converts the polarity in the cortex. We investigated the comparison of the arteriole response near the stimulated neurons in cerebral cortex slices between 95% and 20% O<sub>2</sub> concentration in medium. We showed that the diameter of the blood vessels decreased under 95% O<sub>2</sub> concentration when the pyramidal cells near the blood vessels were stimulated and increased under 20% O<sub>2</sub> concentration with the same stimulation.</p>

    Cell adhesion on biomaterial surfaces has been extensively studied from the perspective of the adsorption properties of adhesive ligands, while recent research on mechanobiology has been revealing a critical role of the mechanical properties of the extracellular milieu in the control of cell adhesion, such as the stiffness and viscoelasticity of the matrix. Although the effects of the lateral mobility of an adhesive ligand have been intensively investigated in a model substrate with water-soluble polymer layers, less is known about those in the setting of lateral deformability of hydrophobic condensed polymer layers. In this study, to help clarify this issue, we used PNIPAAm-grafted substrates with a well-controlled degree of graft-polymerization (DGP) as a typical hydrophobic condensed polymer surface at a cell culture temperature of 37 °C. We observed a clear negative correlation between cell spreading and DGP of PNIPAAm regardless of the amount of fibronectin adsorbed on the substrates, which was found to be attributable to the lateral deformability of a condensed PNIPAAm layer based on lateral force microscopic analysis. The surface-lateral-deformation-induced modulation in stability and maturation of focal adhesion of the cells is discussed in relation to the matrix-strain-induced alteration of the density distribution of adsorbed adhesive ligands.

    Physically cross-linked poly(vinyl alcohol) (PVA) gels are versatile biomaterials with their excellent biocompatibility and mechanical strength. Since the late 1970s, the semicrystalline PVA gels prepared by a freeze-thawing method (FT gel) have been extensively studied in characterizing the structural and functional properties for practical applications, such as artificial hydrogel cartilage. Recently, a simple preparation method for a physically cross-linked PVA gel by a cast-drying method (CD gel) was reported. Although the network nanostructures are similar, CD gels are transparent and elastic, while FT gels are opaque and less elastic. The crystallization conditions of these systems have been investigated; the gels become highly swollen and rigid by the selection of optimum preparation conditions. In this chapter, the mechanical properties of FT and CD gels, such as tearing energy, sliding friction, and abrasion loss, are reviewed in connection with the nano- and microstructures of physical PVA gels. The tribological properties of FT and CD gels are compared with that of natural cartilage. Based on experimental results, simple preparation methods to improve the mechanical and lubrication properties of physically cross-linked PVA gels are presented, and the mechanism of superior lubricity in newly developed PVA gels for artificial hydrogel cartilage is discussed.

    細胞遊走は、恒常性の維持、様々な生理機能の発現,組織・器官の発生において基礎をなす機能である。細胞は基材に応じて細胞挙動を変化させることが知られており、例えば硬さの違いを感知し、より硬い領域へ遊走する。申請者は近年、表面に微小な凹凸構造が存在するハイドロゲルを開発し、細胞が表面の微細凹凸構造の周期や大きさによって郵送挙動が異なる現象を見出した。本研究課題は、表面凹凸による細胞運動変化のメカニズムを解明することを目的とする。

    細胞遊走は、恒常性の維持、様々な生理機能の発現,組織・器官の発生において基礎をなす機能である。従来研究において、細胞は基材の硬さの違いを感知し、より硬い領域へ遊走することが知られている。この運動はがん細胞などで観察されている。これに対して申請者は、表面に微小な凹凸構造が存在する時、細胞が従来とは逆に柔らかい基材に細胞が動くことを見出した。本研究課題は、表面凹凸による細胞運動変化のメカニズムを解明することを目的とする。
