作物学はフィールド(群落)・個体・細胞分子・地球環境にわたる複合的学問であり、農業(栽培)現場から作物生理を捉え,個体・細胞・分子レベルで解析を行い、その知見を再び農業現場へ還元することを土台としたいと考えている。昨今の地球環境変動や人口増加傾向による食糧問題は,作物収量の改善が世界的な重要課題であることを再認識させている。また、国内に目を向ければ,地球温暖化による水稲の収量・品質の低下、外国産大豆との収量性の差異,外国産小麦との収量・品質差など、日本国内においても作物収量の改善はもちろんのこと、子実品質の改善が再重要視されている。そこで、作物収量および品質において重要形質である子実(種子)に着目した研究を推進したいと考えている。ダイズにおいては、子実収量に関する形質として子実重改変へ向けてシンク形成機構から子実重の改変を試みる。また、水稲の高温障害やコムギの穂発芽など、子実品質の低下要因には登熟期間の環境要因が強く影響していることから、子実品質と登熟環境との関連性について明らかにする。作物子実の収量及び品質向上へ向けて,子実重の改変に関わる形質や登熟環境と子実品質との関係について,従来の作物学的手法に加えて,様々な遺伝資源やゲノム情報を活用し,分子生物学・生化学的手法から作物学研究に取り組む。

    Rice grain chalkiness occurs when grains fill under heat stress, greatly reducing grain quality. But the effects of heat stress during grain filling on the subsequent development and adaptive traits remain to be elucidated. Here, we evaluated the effects of heat stress during parental grain filling on the thermotolerance of subsequent plants grown under heat stress after anthesis. Subsequent plants were grown from control (25 °C) and heat-exposed (30 °C) parental seeds under natural conditions until anthesis. Then plants were divided into three treatment groups—control [parental plants] – control [subsequent plants] (CC, 25 °C), control–heat (CH, 30 °C), and heat–heat (HH, 30 °C). Plants grown from heat-stressed seeds had thicker and shorter flag leaves, which delayed leaf senescence and improved photosynthesis under heat stress. HH plants also had significantly less chalkiness than those of CH plants. DNA methylation analysis revealed that heat stress during grain filling significantly hypomethylated promoters of starch biosynthesis genes and hypermethylated those of α-amylase genes. Consequently, HH plants had significantly higher expression of starch biosynthesis genes and suppressed expression of starch-degrading α-amylase genes than CH plants under heat stress. We propose that heat stress during grain filling induces adaptive transgenerational memory that allows subsequent plants to better cope with heat stress.

    We analysed the effects of waterlogging during different reproductive stages in soybean. Short-term waterlogged condition at the full pod and beginning seed stages was the strong for decreasing yield with less pod number. The yield loss by waterlogged conditions was mainly caused by decreasing pod number. Additionally, soybean plants under waterlogged conditions decreased photosynthesis rate and the number of nodules. The expression of GmATG8c, autophagy-related gene, increased after waterlogged conditions. Foliar nitrogen application after waterlogged conditions mitigated yield loss. Photosynthetic rate was restored by foliar nitrogen application, but the number of nodules was not recovered. The expression of GmATG8c induced by waterlogged conditions was restored to control after the application. Taken together, soybean was the most sensitive to waterlogging at R4, leading to yield loss with decreasing in pod number.

    Pod is an important organ for seed production in soybean. Pod size varies among soybean cultivars, but the mechanism is largely unknown. Here we reveal one of the factors for pod size regulation. We investigate pod size differences between two cultivars. The longer pod of ‘Tachinagaha’ is due to more cell number than in the short pod of ‘Iyodaizu’. POD SIZE OF SOYBEAN 8 (GmPSS8), a member of the heat shock protein 70 (HSP70) family, is identified as a candidate gene for determining pod length in a major QTL for pod length. Expression of GmPSS8 in pods is higher in ‘Tachinagaha’ than ‘Iyodaizu’ and is highest in early pod development. The difference in expression is the result of an in/del polymorphism which includes an enhancer motif. Treatment with an HSP70 inhibitor reduces pod length and cell number in the pod. Additionally, shorter pods in Arabidopsis hsp70-1/-4 double mutant are rescued by overexpression of GmPSS8. Our results identify GmPSS8 as a target gene for pod length, which regulates cell number during early pod development through regulation of transcription in soybean. Our findings provide the mechanisms of pod development and suggest possible strategies enhancing yield potential in soybean.

    This study reports the response of lettuce seeds undergoing dormancy breaking and early senescence to DBD plasma irradiation. A heat map of germination percentages at 12 h reveals that dormancy has broken at 39 days' storage, and that one minute of plasma irradiation enhances germination in dormant seeds. Plasma irradiation does not affect those seeds where dormancy has already broken. Early senescence via storage was estimated using ESR measurements and the molecular modification of quercetin. This study reveals that lettuce seed susceptibility to plasma irradiation depends on storage duration and conditions, with dormancy state as a critical variable modulating the impact of plasma irradiation.

    <p>In mechanically harvested soybean, green stem disorder (GSD) is an undesirable trait that causes green-stained seeds, which are graded lower in Japan. To obtain DNA markers for reduced GSD, we conducted a quantitative trait locus (QTL) analysis for 2 years using F<sub>4</sub> and F<sub>5</sub> lines from a cross between ‘Suzuotome’ (less GSD) and ‘Fukuyutaka’ (more GSD). We validated the effect of a detected QTL for GSD by first identifying F<sub>4</sub> or F<sub>5</sub> plants in which one or more markers in the QTL region were heterozygous. The F<sub>5</sub> or F<sub>6</sub> progeny of each plant was used to form a pair consisting of two groups in which the QTL region was homozygous for either the ‘Suzuotome’ or ‘Fukuyutaka’ allele in a similar genetic background, and the two groups within each pair were compared for GSD. Over 3 years of testing, the ‘Suzuotome’ allele of a QTL on chromosome 6 was found to reduce the level of GSD. This novel QTL was mapped to the region around DNA marker W06_0130, and was not closely linked to QTLs for important agronomic traits including yield components. Using this marker, the low level of GSD from ‘Suzuotome’ could be conferred to ‘Fukuyutaka’ or other high-GSD cultivars.</p>

  • Health assessment of rice cultivated and harvested from plasma-irradiated seeds

    This study provides the health effects assessment of rice cultivated from plasma-irradiated seeds. The rice (Oryza sativa L.) cultivated from seeds with plasma irradiation showed a growth improvement (slope-ratios of with plasma to without plasma were 1.066, 1.042, and 1.255 for tiller, and earing, and ripening periods, respectively) and an 4% increase in yield. The cultivated rice was used for repeated oral administrations to mice for 4-week period. Distilled water and rice cultivated from seeds without plasma irradiation were also used as control. The weights of the lung, kidney, liver, and spleen, with corresponding average values of 0.22 g, 0.72 g, 2.1 g, and 0.17 g for w/ plasma group and 0.22 g, 0.68 g, 2.16 g, and 0.14 g for w/o plasma group, respectively, showing no effect due to the administration of rice cultivated from plasma-irradiated seeds. Nutritional status, liver function, kidney function, and lipid, neutral fat profiles, and glucose metabolism have no significant difference between with and without plasma groups. These results show no obvious subacute effects were observed on rice grains cultivated and harvested from the mother plant that experienced growth improvement by plasma irradiation. This study provides a new finding that there is no apparent adverse health effect on the grains harvested from the plasma-irradiated seeds.

    Climate change due to global warming is now affecting agricultural production worldwide. In rice, one of the most important crops, water limitation due to irregular rainfall in rainfed lowlands during crop growth limits yield. Dry direct-sowing has been proposed as a water-efficient approach to cope with water stress during rice growth, but poor seedling establishment due to drought during germination and emergence is a problem. Here, we germinated indica rice cultivars Rc348 (drought tolerant) and Rc10 (drought sensitive) under osmotic stress induced by PEG to elucidate mechanisms of germination under drought. Rc348 had higher germination rate and germination index under severe osmotic stress of −1.5 MPa, above those of Rc10. Rc348 showed up-regulated GA biosynthesis, down-regulated ABA catabolism, and up-regulated α-amylase gene expression in imbibed seeds under PEG treatment compared to that of Rc10. During germination, reactive oxygen species (ROS) play important roles in antagonism between gibberellic acid (GA) and abscisic acid (ABA). Embryo of Rc348 treated with PEG had significantly greater expression of NADPH oxidase genes and higher endogenous ROS levels, together with significantly increased endogenous GA<inf>1</inf>, GA<inf>4</inf> and ABA contents compared to that of Rc10. In aleurone layers treated with exogenous GA, expression of α-amylase genes was higher in Rc348 than in Rc10, and expression of NADPH oxidase genes was enhanced with significantly higher ROS content in Rc348, suggesting higher sensitivity of GA to ROS production and starch degradation in aleurone cells of Rc348. These results suggest that the osmotic stress tolerance of Rc348 is due to enhancement of ROS production, GA biosynthesis, and GA sensitivity, resulting in a higher germination rate under osmotic stress.

  • Bulbil Formation on Water Yam (<i>Dioscorea alata</i> L.) Is Promoted by Waterlogged Soil

    The formation of bulbils, which are storage organs, is an important agronomic trait and a unique morphological feature in the vegetative reproduction of yam. We found a landrace of water yam (Dioscorea alata L.), which rarely forms bulbils, that produces bulbils during periods of high rainfall. We investigated the physiological mechanism of bulbil formation in response to over-moist soil and relevant factors at the single plant level. Waterlogging (WL) treatment markedly increased the number of bulbils initiated, predominantly toward the upper nodes. This formed-bulbil was an accessory bud developed as a storage organ in leaf axils. Photosynthetic capacity decreased under WL, attributed to stress-induced stomatal closure. WL stress also reduced dry matter partitioning to the belowground organs. During tuber enlargement in WL plants, photosynthetic products accumulated in the aboveground organs and were transported to the bulbils as a result of reduced translocation to belowground organs. We investigated the effect of abscisic acid (ABA) on bulbil formation on the basis of changes in the sink–source balance in response to WL stress. ABA treatment of leaf axils enhanced bulbil formation in unstressed plants, suggesting that increased ABA is one of the factors that initiate bulbils. Our study shows that bulbil initiation occurs as a result of changes in physiological conditions in response to WL stress. This finding may provide fundamental information for the control of bulbil production. This response of bulbil formation, as an environmentally adaptive trait of the tropical water yam, may underlie the survival strategy of vegetatively propagated plants.

    Degradation of the appearance of rice grains due to high temperatures during ripening is a serious problem being worsened by global warming. QTLs for tolerance to high temperature during ripening must be identified in order to develop new cultivars efficiently. Here, we developed recombinant inbred lines (RILs) from crosses between the high-temperature-tolerant ‘Genkitsukushi’ and the high-temperature-sensitive ‘Tsukushiroman’. The occurrence of white-back grains is significantly lower in ‘Genkitsukushi’ than in ‘Tsukushiroman’. We evaluated their high-temperature tolerance and performed quantitative trait locus (QTL) analysis. A QTL for white-back grains on chromosome (chr.) 6 and a large-effect QTL on chr. 8 were identified in the two-year experiments. The QTL on chr. 8 was close to one we previously detected using RILs derived from ‘Chikushi 52’ × ‘Tsukushiroman’. The results indicate that the QTL on chr. 6 is unique to ‘Genkitsukushi’ × ‘Tsukushiroman’, and that on chr. 8 could be identical between ‘Genkitsukushi’ and ‘Chikushi 52’. Backcrossed lines which harbor the QTL region on chr. 8 of ‘Genkitsukushi’ in the background of ‘Tsukushiroman’ verified its effects. We also analyzed the marker genotypes near the QTLs on chr. 6 and 8 in relatives of ‘Genkitsukushi’. We discuss the interaction between these QTLs and their relationship with high-temperature tolerance.

    In tuberous crops, tuber enlargement is one of the most important target traits for yield formation. It has long been known that tuber growth in yams is enhanced by short-day (SD) conditions, but the mechanism of tuber enlargement remains unknown. Here, we analyzed the photoperiodic regulation of tuber enlargement in water yam (Dioscorea alata L.). The photoperiod experiments in seedlings showed that tuber enlargement is initiated under SD conditions (≤10 h daylength) within 20 days of treatment. DaFT2, a FLOWERING LOCUS T (FT)-like gene, was upregulated in SD and downregulated in long-day (LD) conditions in tubers, suggesting that DaFT2 promotes tuber enlargement. DaFT1, the other FT-like gene, was significantly upregulated only in the leaves under LD, and its expression pattern was opposite to that of DaFT2 in the tubers. A night-break experiment showed that tuber growth was inhibited by red light in the dark period. These results suggest that the tuber enlargement of water yam is completely dependent on the photoperiod and that it involves an FT gene-mediated mechanism in response to the SD condition by red light sensing.

    Key message: Alterations in DNA methylation levels of ROS, GA and ABA related gene promoters cause transcriptional changes upon imbibition to induce seed germination in barley seeds exposed to heat stress during grain filling. Abstract: Environmental changes, especially changes in temperature, during seed development affect germination in several plant species. We have previously shown that heat stress during rice grain filling alters DNA methylation, an epigenetic mark important for gene silencing, regulates transcript levels of phytohormone metabolism genes, and delays seed germination. However, whether this phenomenon is present in other plant species remained to be elucidated. In this study, we compared seeds germination of barley (Hordeum vulgare L.) plants grown at 15 °C (control) or 25 °C (heat stress) during grain filling. Heat stress during grain filling significantly promoted seed germination in comparison with the control. The phytohormone gibberellic acid (GA) and reactive oxygen species produced by NADPH oxidases promote seed germination, whereas phytohormone abscisic acid (ABA) suppresses seed germination. We found that in heat-stressed seeds, genes related to ABA biosynthesis (HvNCED1 and 2) were significantly suppressed, whereas genes related to ABA catabolism (HvABA8’OH) and GA biosynthesis (HvHA20ox, HvGA3ox), and NADPH oxidase (HvRboh) genes were significantly upregulated after imbibition. Using MeDIP-qPCR, we showed that the promoters of HvNCED were hyper-methylated, and those of HvABA8’OH1, HvABA8’OH3, HvGA3ox2, and HvRbohF2 were hypo-methylated in heat treated seeds. Taken together, our data suggest that heat stress during grain filling affects DNA methylation of germination-related genes and promotes seed germination in barley.

    For sustainable food production, labor-saving cultivation systems are required. Yam, a major food crop, is cultivated mostly with supports such as stakes (staking), which increases tuber yield (TY) but involves high material and labor costs. We, therefore, focused on non-staking water yam cultivation, in which no stakes are used. The effects of different cultivation methods (staking vs. non-staking) on TY, yield components, shoot traits, and tuber shape of six cultivars were investigated in a two-year field experiment, and phenotypic traits related to yielding ability in non-staking cultivation were analyzed. Averaged across years and cultivars, TY was significantly lower (by 19%) in non-staking than in staking cultivation because of smaller single-tuber weight. TY was significantly affected by the cultivation × cultivar interaction. We found no difference among cultivars in staking cultivation. In non-staking cultivation, Yamatomakousha and Shirokoushaman 1 cultivars had higher TY than the other cultivars, which was similar to their TY in staking cultivation. Shoot dry weight and vine number were closely associated with TY in both cultivations, whereas lower tuber length-to-width ratio was strongly related to higher TY only in non-staking. Tubers of Yamatomakousha and Shirokoushaman 1 were more rounded than those of other cultivars. In non-staking cultivation, these two cultivars showed a higher vine number and, thus, maintained higher TY owing to higher above-ground growth. Therefore, rounded tubers and high vine number are target traits for non-staking cultivation of water yam.

    <jats:title>Abstract</jats:title><jats:p>Discharge plasma irradiates seeds with reactive oxygen and nitrogen species (RONS). However, RONS introduced in seeds by plasma irradiation have not been successfully detected thus far. This study provides experimental evidence that nitrate ion NO<jats:sub>3</jats:sub><jats:sup>−</jats:sup> is introduced in lettuce seeds as RONS upon irradiation with atmospheric-pressure air dielectric barrier discharge plasma. Plasma irradiation for 5 min promotes seed germination. The components of the plasma-irradiated seeds were examined using electrospray ionization quantum mass spectrometry (ESI QMS), which revealed that the plasma irradiation introduced an ion with a mass of 62 m/z in detectable amounts. This ion was identified as NO<jats:sub>3</jats:sub><jats:sup>−</jats:sup> by liquid chromatography (LC), multiple wavelength detector (MWD), and LC-ESI QMS. A one-dimensional simulation at electron temperature T<jats:sub>e</jats:sub> = 1 eV, electron density N<jats:sub>e</jats:sub> = 10<jats:sup>13</jats:sup>/m<jats:sup>3</jats:sup>, and gas temperature T<jats:sub>g</jats:sub> = 300 K indicated the introduction of NO<jats:sub>3</jats:sub><jats:sup>−</jats:sup>, involving nitric oxide NO. NO<jats:sub>3</jats:sub><jats:sup>−</jats:sup> is one of the most important ions that trigger signal transduction for germination when introduced in seeds. The scanning electron microscopy (SEM) images revealed that there was no change on the surface of the seeds after plasma irradiation. Plasma irradiation is an effective method of introducing NO<jats:sub>3</jats:sub><jats:sup>−</jats:sup> in seeds in a dry process without causing damage.</jats:p>

    プラズマは、環境負荷の少ない植物成長促進法として注目を集めている。中でも種子へのプラズマ照射は、低コストな成長促進だけでなく、DNA修飾変動などを示す。種子へのプラズマ照射からDNA修飾変動に至る分子機構解明のためには、プラズマから種子内細胞までの分子機構がプラズマによりどのように変動を受けるのかを統合した学理の構築が必要である。本総括研究では、5年間の研究期間において、プラズマ科学と種子科学を中心に、様々な分野における研究者が一同に会して議論しプラズマ種子科学領域を創成する。

    プラズマ照射条件に応答する植物内メカニズムについて、ゲノムワイドなエピゲノム解析(WGBS、Chip-seq、ATAC-seq)や遺伝子発現解析(RNA-seq)により網羅的な要因解明を実施する(石橋)。また、エピゲノム制御はDNAメチル基転移酵素やヒストンアセチル化酵素等の酵素反応により制御されていることから、プラズマ照射によるエピゲノム制御関連酵素の構造変化を調査する(萩原)。ゲノム中には多くの転移因子(トランスポゾン)が存在しており、イネゲノムの35%はトランポゾン様の配列である。そこで、プラズマ照射によるエピゲノム制御を介したトランスポゾンの再編成の可能性を調査する(栂根)。

    本研究課題では、研究材料をイネとし登熟ステージ毎や複数回の高温環境を登熟子実に記憶させる。更に、その種子内のメチル化DNAやヒストン修飾などのエピゲノム情報や生育後の遺伝子発現など網羅的に解析すると共に生育調査を実施し、エピゲノム情報と表現型情報により、高温環境適応型の種子生産に関与するエピゲノムマーカーを特定する。本研究課題は、種子科学の理解とエピジェネティク解析技術が必要不可欠であるが、研究代表者である石橋は、これまでの種子生理科学の知見を、研究分担者のSuriyasakは、これまでのエピジェネティク研究の経験をフル活用して以下の研究課題に挑む。

    種子の発芽は、植物の一生の始まりであり、極めて神秘的な生命現象であると同時に食料に直結する重要な農業形質の一つでもある。この種子発芽機構に、一般的に毒性のイメージが強い活性酸素が重要なシグナル分子として機能していることは、意外と知られていない。これまで、申請者らは、種子が吸水後、積極的に活性酸素を産生し、遺伝子発現調節やタンパク質修飾を介して発芽を促進することを明らかにしている。本研究課題では、国際共同研究を実施することで、国際種子発芽ネットワークが構築され、種子発芽における統御システムの構築、農学的にはモデル植物から農作物への応用展開と繋がっていくと考える。

    As global warming has been a problem, the quality of rice deteriorates due to high temperatures are also a problem. This study was conducted with the aim of clarifying the genetic factors that lead to the decline in rice quality.
    When comparing the varieties used in this study that tend to deteriorate in quality under high temperatures with those that do not easily deteriorate in quality even under high temperatures, regions that cause quality deterioration were detected in the rice genome. They also found multiple genes with different expression levels in that region. These genes include genes involved in plant hormone metabolism and DNA transcription, and are thought to be involved in the deterioration of rice quality.

    Dioscorea ssp. has unique characteristic of vegetative propagation through the formation of bulbil and underground tuber. In this study, we investigated the mechanism of tuber formation and enlargement in one species, Dioscorea alata L.(water yam).
    It was revealed that FT-mediated control mechanisms are involved in tuber enlargement in response to short-day, and two FT genes were identified that regulate enlargement through aboveground-underground communication. Also, mutant lines were developed and early maturing lines were selected.
    Bulbil formation was demonstrated to be accelerated in response to environmental stresses in the underground parts. These characteristics were thought to be related to (1) the enhancement of above-ground abscisic acid biosynthesis and other factors resulting from stress sensing in the roots, and (2) an increase in sugar concentration in the leaf axils due to suppression of translocation to underground tuber.

    The objective of this research project was to apply Drug Delivery System (DDS) technology, which has been pioneered in the medical field, to plants. Considering the eventual agronomic application, we focused on nanoparticles from various DDS technologies. As a method for nanoparticles of essential plant elements, we employed supercritical technology, which is a clean method. As a result, they succeeded in producing highly dispersible nanoparticles, which were introduced into plants by foliar spraying, and confirmed high introduction efficiency and differences in localization within plants. Furthermore, when the active ingredient was added to the nanoparticles and introduced into the plant, the efficacy was successfully confirmed at lower concentrations.

    This research aimed to determine whether the environment during grain maturation affects the growth of the next generation of rice plants. When rice was used as the test material and the environment was subjected to high temperature stress, the heat stressed seeds showed various phenotypes such as seed germination, plant height, branching, heading/flowering, and morphology of the stop leaves in the growth of the next generation of rice plants. Furthermore, whole genome methylation analysis of DNA extracted from seeds that had experienced high temperature stress suggested that DNA methylation of genes involved in the above phenotypes was involved. Additionally, these phenotypes were divided into traits that persist for several years once memorized and those that do not, suggesting that there is a degree of memory. In summary, the results of this study suggest that the environment during the crop's ripening process influences the growth of the next generation.

    In this study, we aimed to clarify "what is the optimum physiological state of seeds for plasma irradiation". We found that the sensitivity of seeds to plasma irradiation changes depending on seed aging and seed coat color. The effect of plasma irradiation changes depending on the state of seeds even under the same irradiation conditions. The study also revealed that plasma irradiation can alter DNA modifications in seeds. The response to plasma irradiation differs depending on the dormancy state of seeds. For our developed scalable dielectric barrier discharge plasma, its growth-promoting effect was greater when seeds were in a deeper dormancy state.
