    May 2017 was the second-warmest May in 137 years of modern record-keeping, according to a monthly analysis of global temperatures by NASA's scientists (NASA, June 15, 2017). As our climate is changing, adaptation with the changing environment is one of the critical approaches to face the challenge of global warming induced heat stress. To facilitate the adjustment, first, we may need to know how the physiological functions are responding to high ambient temperature (HT). Most of the heat stress studies in avian species have been conducted using commercial broilers or layer chickens because of their susceptibility to heat stress. However, many people think that young chicks are not sensitive to HT since chicks need additional ambient temperature for their survival. My findings showed that young chicks from 2-weeks old were susceptible to heat stress and declined their food intake under HT, although the trend of increasing body temperature was more pronounced from 7-days old compared to 3- or 5-days old. Diencephalon, which includes thalamus and hypothalamus, was more prone to heat stress than other parts of the brain. Since heat stress causes to decline food intake and diencephalon plays a vital role in food intake regulation, expressions of some orexigenic and anorexigenic neuropeptides were analyzed in heat-exposed chicks. Although no anorexigenic neuropeptide was altered, hypothalamic orexigenic neuropeptides, namely neuropeptide Y (NPY) and gonadotropin-inhibitory hormone (GnIH) mRNA expressions were induced by heat stress. NPY played a role as an anti-stress factor in heat-exposed fasted chicks while brain GnIH mediated stress response in fed chicks under HT. The concentrations of both essential and nonessential amino acids were altered in several tissues including plasma by the exposure to heat stress. For example, plasma citrulline (Cit) was declined in the heat-exposed chicks, and plasma leucine (Leu) was decreased in the heat-exposed embryos. Subsequently, it was revealed that oral administration of L-Cit, but not the central injection, declined body temperature and afforded thermotolerance in chicks. L-Cit-mediated thermotolerance also appeared in marketing aged broilers. Oral administration of L-Cit decreased plasma glucose in chicks and increased lactic acid in broilers, which suggested that orally administered L-Cit may limit the substrate for heat production. As a result, hypothermia and thermotolerance may occur. L-Leu in ovo injection caused the reduction of body temperature at hatching and afforded thermotolerance in young broiler chicks. Interestingly, it was further found that L-Leu-mediated thermotolerance appeared in male, but not female, broiler chicks. In conclusion, declining of food intake and increasing of body temperature were confirmed in heat-exposed young chicks, which indicated that vulnerability of young chicks to heat stress. The central effects of NPY and expression of GnIH further suggest that fasting under heat stress condition would be beneficial in chickens to adapt heat stress. Some amino acids, like L-Cit and L-Leu, may play important roles to facilitate the adaptation of chickens to heat stress. Future study will clarify how the amino acids could be applied in poultry farms to promote heat adaptation during hot summer.

    High ambient temperatures (HT) can increase diencephalic neuropeptide Y (NPY) expression, and central injection of NPY attenuates heat stress responses while inducing an antioxidative state in the chick spleen. However, there is a lack of knowledge about NPY receptor expression, and its regulation by HT, in the chick spleen. In the current study, male chicks were used to measure the expression of NPY receptors in the spleen and other immune organs under acute (30 vs. 40 ± 1℃ for 3 h) or chronic (30 vs. 40 ± 1℃ for 3 h/day for 3 days) exposure to HT and in response to central injection of NPY (47 pmol, 188 pmol, or 1 nmol). We found that NPY-Y4 receptor mRNA was expressed in the spleen, but not in other immune organs studied. Immunofluorescence staining revealed that NPY-Y4 receptors were localized in the splenic pulp. Furthermore, NPY-Y4 receptor mRNA increased in the chick spleen under both acute and chronic exposure to HT. Central NPY at two dose levels (47 and 188 pmol) and a higher dose (1 nmol) did not increase splenic NPY-Y4 receptor mRNA expression or splenic epinephrine under HT (35 ± 1℃), and significantly increased 3-methoxy-4-hydroxyphenylglycol (MHPG) concentrations under HT (40 ± 1℃). In conclusion, increased expression of NPY-Y4 receptor mRNA in the spleen under HT suggest that Y4 receptor may play physiological roles in response to HT in male chicks.

Outline of Social Contribution and International Cooperation activities

  • I have been contributing to Poultry and Animal Science societies with our updated knowledge of Nutritional Physiology. I have been collaborating for my research with Massey University, Palmerston North, New Zealand, Virginia Polytechnic Institute and State University, Blacksburg, VA, USA, and State Islamic University of Sultan Syarif Kasim Riau, Indonesia. Recently a Prof. from Georgia Univ. a Prof. from Agril Univ. of Athens showed their interest in collaborating with my lab on heat stress studies.
