    L1CAM is a member of the immunoglobulin superfamily involved in cell adhesion, migration, and neural development. While several studies have shown an association between L1CAM and glioblastoma (GBM) cell motility, its mechanobiological function in patient-derived primary cultured GBM cells remains insufficiently understood. This study focuses on L1CAM-dependent motility mechanisms in patient-derived GBM cells at the single-cell level. Patient-derived GBM cells were established from GBM tissues using the neurosphere method. L1CAM expression was evaluated by qPCR, western blotting, flow cytometry, and immunocytochemistry. Cell motility was assessed by single-cell tracking. Functional assays included forced expression and antibody-mediated inhibition of L1CAM. Adhesion-clutch behavior and retrograde F-actin flow were analyzed. For in vivo assessment, doxycycline-inducible L1CAM-overexpressing GBM cells were transplanted into mouse brains. L1CAM-high GBM cells showed increased motility on laminin compared to L1CAM-low cells. Anti-L1CAM antibody suppressed migration of L1CAM-high GBM cells, whereas ectopic expression enhanced migration in L1CAM-low GBM cells. L1CAM-positive GBM cells preferentially migrated on laminin rather than fibronectin, while L1CAM-low cells showed no such difference, indicating L1CAM-dependent haptotaxis. Forced L1CAM expression reduced retrograde actin flow velocity and promoted adhesion-clutch formation. In vivo, GBM cells with L1CAM overexpression displayed greater infiltrative capacity 12 weeks posttransplantation. L1CAM expression on GBM cells' surface regulates cell motility through adhesion-clutch mechanisms and substrate sensing. These mechanobiological features suggest that targeting L1CAM may serve as a promising strategy to suppress GBM cells infiltration and improve GBM treatment outcomes.

    SUMMARY

    Actin dynamics mediate cell morphogenesis. Actin filaments polymerize outward at cell protrusions such as the leading edge of migrating cells, thereby pushing the membrane to protrude. The current paradigm explains that actin dynamics are regulated by cell signaling. However, it is unclear how cells spontaneously form actin-based protrusions even without a specific local signaling cue. We found that arrays of treadmilling actin filaments emerge widely in migrating cells and move in the direction of polymerization as actin waves. Their arrival at the cell periphery pushes the plasma membrane to protrude. Furthermore, they accumulate at protrusions without local signaling cues, similar to self-propelled particles colliding with a boundary. This leads to further growth of protrusions, thereby promoting spontaneous formation of the leading edge for migration. We propose that the self-propelled actin waves drive robust formation of protrusions for cell morphogenesis, through their abilities to self-accumulate into protrusions and push the membrane.

    The growth cone, a motile structure located at the tip of growing axons, senses extracellular guidance cues and translates them into directional forces that drive axon outgrowth and guidance. Axon guidance directed by chemical cues on the extracellular adhesive substrate is termed haptotaxis. Recent studies reported that netrin-1 on the substrate functions as a haptotactic axon guidance cue. However, the mechanism mediating netrin-1–induced axonal haptotaxis remains unclear. Here, we demonstrate that substrate-bound netrin-1 induces axonal haptotaxis by facilitating physical interactions between the netrin-1 receptor, DCC, and the adhesive substrates. DCC serves as an adhesion receptor for netrin-1. The clutch-linker molecule shootin1a interacted with DCC, linking it to actin filament retrograde flow at the growth cone. Speckle imaging analyses showed that DCC underwent either grip (stop) or retrograde slip on the adhesive substrate. The grip state was more prevalent on netrin-1–coated substrate compared to the control substrate polylysine, thereby transmitting larger traction force on the netrin-1–coated substrate. Furthermore, disruption of the linkage between actin filament retrograde flow and DCC by shootin1 knockout impaired netrin-1–induced axonal haptotaxis. These results suggest that the directional force for netrin-1–induced haptotaxis is exerted on the substrates through the adhesion-clutch between DCC and netrin-1 which occurs asymmetrically within the growth cone.

    Axon outgrowth is promoted by the mechanical coupling between F-actin and adhesive substrates via clutch and adhesion molecules in an axonal growth cone. In this study, we utilized a femtosecond laser-induced impulse to break the coupling between the growth cone and the substrate, enabling us to evaluate the strength of the binding between the growth cone and a laminin on the substrate, and also determine the contribution of adhesion strength to axon outgrowth and traction force for the outgrowth. We found that the adhesion strength of axonal L1 cell adhesion molecule (L1CAM)-laminin binding increased with the laminin density on the substrate. In addition, fluorescent speckle microscopy revealed that the retrograde flow of F-actin in the growth cone was dependent on the laminin density such that the flow speed reduced with increasing L1CAM-laminin binding. However, axon outgrowth and the traction force did not increase monotonically with increased L1CAM-laminin binding but rather exhibited biphasic behavior, in which the outgrowth was suppressed by excessive L1CAM-laminin binding. Our quantitative evaluations suggest that the biphasic outgrowth is regulated by the balance between traction force and adhesion strength. These results imply that adhesion modulation is key to the regulation of axon guidance.

    Axon pathfinding is an essential step in neuronal network formation. Shootin1a is a clutch-linker molecule that is mechanically involved in axon outgrowth and guidance. It was previously shown that concentration gradients of axon guidance molecule netrin-1 in the extracellular environment elicit asymmetrically localized Pak1 kinase-mediated phosphorylation of shootin1a within axonal growth cones, which is higher on the netrin-1 source side. This asymmetric phosphorylation promotes shootin1a-mediated local actin-adhesion coupling within growth cones, thereby generating directional forces for turning the growth cone toward the netrin-1 source. However, how the spatial differences in netrin-1 concentration are transduced into the asymmetrically localized signaling within growth cones remains unclear. Moreover, the protein phosphatases that dephosphorylate shootin1a remain unidentified. Here, we report that protein phosphatase-1 (PP1) dephosphorylates shootin1a in growth cones. We found that PP1 overexpression abolished the netrin-1-induced asymmetric localization of phosphorylated shootin1a as well as axon turning. In addition, we show PP1 inhibition reversed the asymmetrically localized shootin1a phosphorylation within growth cones under netrin-1 gradient, thereby changing the netrin-1-induced growth cone turning from attraction to repulsion. These data indicate that PP1-mediated shootin1a dephosphorylation plays a key role in organizing asymmetrically localized phosphorylated shootin1a within growth cones, which regulates netrin-1-induced axon guidance.

    細胞の移動や異物取り込み等の細胞運動には、力の生成とその制御が不可欠である。例えば、細胞が誘引分子に向かって移動する走化性では、細胞は誘引物質に応答して移動のための推進力を細胞外環境に及ぼす。一方、食作用では、食細胞が異物を認識して異物に対して取り込むための力を生み出す。本研究は、「細胞外の誘引物質や異物の刺激を受容した細胞がいかにシグナル伝達を介して力を生み出し細胞移動や食作用を遂行するのか」を、応募者らが同定したシューティンを分子基盤として、1分子イメージングや力計測法等の手法を交えて解明する。本研究は、シグナル伝達を介した細胞移動および食作用の全容解明に繋がる点で創造性がある。

    グリオブラストーマは悪性度の高い脳腫瘍の代表であり、脳内を迅速に浸潤し腫瘍範囲を拡大するため、患者の予後は悪い。しかしながら、グリオブラストーマ細胞の脳内浸潤機構は不明である。これまでに申請者は、Shootin1が神経軸索の伸長および誘引に関与することを明らかにした。本研究では、Shootin1を分子基盤としてグリオブラストーマ細胞の浸潤機構を解明する。本研究の成果は、グリオブラストーマ細胞の浸潤機構の理解を深めるだけでなく、腫瘍生物学や医学、細胞生物学などの研究分野に学術的波及効果をもたらし、癌細胞の浸潤を阻害する治療薬の開発に貢献することが期待できる。

    本研究は、「いかにしてアクチン線維が細胞内で局所的に自己組織化をして細胞移動を引き起こすことができるのか」を、我々が最近明らかにした「アクチン波」の分子メカニズムとその特徴的な挙動から解明することを目指す。本研究では、我々のこれまでの研究成果と異分野融合型の共同研究に基づいて、細胞内1分子計測や牽引力顕微鏡、マイクロパターン技術、定量的数理モデルを交えた最先端の分子細胞生物学的手法を用いて自発的な細胞移動と走化性の発生起源に迫る。

Educational Activities

  • これまでの経験を活かし、研究遂行能力の育成や、国際的に活躍できる人材の育成、研究不正防止の教育に取り組む。以下に具体的な教育活動と抱負について記述する。
    1. 研究遂行能力の育成
    幅広い知識や高度な実験技術を備えるための指導を行うだけでなく、研究を遂行するための有効な方法について考えることができるように指導する。そのため、研究の問題点とその改善方法について頻繁に議論することが必要である。そこで、日常的に実験方法や結果の解釈などについて学生と議論するだけでなく、教員やスタッフ、学生同士が積極的に議論できる環境を作る。その様な環境を作ることで、自分以外の思考やアイディアを積極的に取り入れることができ、研究を遂行させるための有効な方法を生み出す能力が養われる。研究課題に対して客観的に考え、研究を遂行するための能力をもつ人材の育成に努める。
    2. 国際的に活躍できる人材の育成
     研究者は自身の研究成果を世界に向けて公表することで評価される。日本国内の研究水準は海外の研究に比べて劣らないが、英語論文や英語のプレゼンテーションにおいては海外の研究者より劣っている。この問題を解決するために、実験結果の報告や論文紹介などを英語で説明できるように指導するだけでなく、学生が大半の時間を過ごす研究室内において留学生とコミュニケーションをとるように促す。それにより、語学能力が向上するだけでなく、海外研究者と接する際のコミュニケーション能力が養われる。海外の研究者と対等に渡り合える能力を持った人材の育成に努める。
    3. 研究不正防止の教育
     論文文書の剽窃、データの改竄および捏造などの研究不正の問題が世界中で問題となっている。研究者の不正は研究者個人や大学のみならず、科学の信用を損なう深刻な問題であり、各研究室が研究不正防止の教育を徹底する必要がある。研究倫理、研究資料の保管、データの記録方法について指導する。これに加えて、第三者と文書や生データを確認しながら議論するように指導し、剽窃・改竄・捏造をさせない意識を向上させ、研究不正防止の教育に努める。

    ・専攻分野における実務上の経験等:
    大学院の学生時から助教にわたる約10年の間、細胞生物学や分子生物学、生化学、発生生物学、メカノバイオロジーなどに基づく知識や最先端の技術を駆使して、神経細胞の軸索伸長や軸索ガイダンスの分子機構および樹状細胞の走化性の分子機構について解析してきた経験があり、関連する分野での研究成果を収めてきた。これまで培った知識・技術・経験を活用することで、基盤幹細胞学研究室における幹細胞分化やエピジェネティック制御に関する機構の研究をさらに発展させ、プロジェクトを遂行することができると考える。以下にこれまでに取得した実験手法を記す。
    ●細胞生物学的手法:
    神経細胞や樹状細胞の初代培養、細胞の免疫染色、細胞運動のための推進力の解析、蛍光タンパク質の細胞内イメージング、1分子計測、FACSによる細胞表面マーカー解析およびセルソーティング。リンパ節内への樹状細胞のインジェクション、細胞の走化性の解析。
    ●遺伝子工学的手法:
    組み換えDNA作成、遺伝子クローニング、形質転換、細胞への遺伝子導入、子宮内電気穿孔法によるマウス胎児脳への遺伝子導入。
    ●分子生物学的・生化学的手法:
    核酸およびタンパク質抽出、ウエスタンブロッティング、タンパク質間の相互作用解析、タンパク質精製(アフィニティー精製、イオン交換クロマトグラフィー精製、ゲル濾過クロマトグラフィー精製)、質量分析、抗体の作成および精製。放射性同位体を用いたタンパク質のリン酸化反応の解析。
    ●化学工学的手法:
    シリコンゲルを用いたマイクロ流路の設計および作成(化学物質の濃度勾配を形成する)。
