研究  現在主に行っている研究テーマは『病態モデル動物を用いた新規な高コレステロール血症発症機構に関する研究』です。  高コレステロール血症モデル動物である外因性高コレステロール血症(ExHC)ラットの遺伝子解析により高コレステロール血症発症機構の解明を行っています。現在は原因遺伝子同定の過程で発見したSmek2遺伝子について、その機能の解析を行っています。また、Smek2を利用した新たな疾患モデル動物の作製にも取り組んでいます。 教育  おもに、大学院生の修士論文および博士論文指導を行っています。また、学部学生は卒論指導および学生実験を行っています。

    Vitamin D is a hormone-like vitamin that plays a primary physiological role in regulating calcium metabolism. In addition to endogenous synthesis through sunlight exposure, dietary supplementation with vitamin D is indispensable for maintaining optimal vitamin D levels. To ensure precision and efficacy in quantifying daily vitamin D intake among the Japanese people, we established a method to measure vitamin D in food. This method optimizes instrument costs and streamlines cumbersome processes. Initially, thin-layer chromatography (TLC) was used to separate vitamin D from food samples, followed by a quantitative analysis using gas chromatography-mass spectrometry (GC-MS) with deuterated vitamin D as an internal standard. Despite the elevated temperature conditions of GC-MS, which lead to the generation of thermal decomposition compounds, the method remains accurate and effective, with an limit of quantitation (LOQ) as low as 0.8 ng for vitamin D. Subsequently, we applied this method to quantify the vitamin D content in 120 meal samples. The meals, designed by a registered dietitian (RD), were served in the dormitory and cafeteria of a Japanese company. The total vitamin D content {mean ± standard deviation (SD)} was 14.7 ± 13.7 µg/day. The measured total vitamin D content in the meal samples, showing a congruent range of 15.7 ± 12.6 µg/day, closely aligned with the corresponding estimate derived from Excel Eiyou-kun. In summary, this study established a method using GC-MS to simultaneously measure vitamin D2 and D3 in meals, which may be suitable for analyzing food samples for their vitamin D content in large quantities.

    Deficit of oxygen and nutrients in the tumor microenvironment (TME) triggers abnormal angiogenesis that produces dysfunctional and leaky blood vessels, which fail to adequately perfuse tumor tissues. Resulting hypoxia, exacerbation of metabolic disturbances, and generation of an immunosuppressive TME undermine the efficacy of anticancer therapies. Use of carefully scheduled angiogenesis inhibitors has been suggested to overcome these problems and normalize the TME. Here, we propose an alternative agonist-based normalization approach using a derivative of the C-type natriuretic peptide (dCNP). Multiple gene expression signatures in tumor tissues were affected in mice treated with dCNP. In several mouse orthotopic and subcutaneous solid tumor models including colon and pancreatic adenocarcinomas, this well-tolerated agent stimulated formation of highly functional tumor blood vessels to reduce hypoxia. Administration of dCNP also inhibited stromagenesis and remodeling of the extracellular matrix and decreased tumor interstitial fluid pressure. In addition, treatment with dCNP reinvigorated the antitumor immune responses. Administration of dCNP decelerated growth of primary mouse tumors and suppressed their metastases. Moreover, inclusion of dCNP into the chemo-, radio-, or immune-therapeutic regimens increased their efficacy against solid tumors in immunocompetent mice. These results demonstrate the proof of principle for using vasculature normalizing agonists to improve therapies against solid tumors and characterize dCNP as the first in class among such agents.

    血圧に対するコール酸(CA)補給食の効果について検討した。WKAH/HkmSlcラット(3週齢)をAIN-93に基づく対照食またはCA補給食(0.5gCA/kg)で13週間飼育した。CA食は肝臓の脂質濃度と共に、収縮期と弛緩期の血圧を上昇させた。血液のナトリウム濃度には変化がなかった。CAで飼育したラットでは、尿のアルブミン濃度が上昇した。血圧と相関するATP-binding cassette subfamily B memberの肝臓での発現が上昇し、肝門のタウロコール酸濃度上昇と尿アルブミン濃度上昇が観察された。以上の結果から、12α-ヒドロキシル化胆汁酸は肝臓機能の変化を介して、血圧上昇とアルブミン尿症に関与すると考えられた。

    Abstract

    A diet supplemented with cholic acid (CA), the primary 12α-hydroxylated bile acid, can induce hepatic lipid accumulation in rats without obesity. This study examined the effects of a CA-supplemented diet on blood pressure (BP). After acclimation, WKAH/HkmSlc rats (3 week old) were divided into two groups and fed with a control AIN-93-based diet or a CA-supplemented diet (0.5 g CA/kg) for 13 weeks. The CA diet increased systolic and diastolic BP as well as hepatic lipid concentrations in the rats. No changes were found in the blood sodium concentration. Urinary albumin concentration increased in CA-fed rats. An increase was observed in the hepatic expression of ATP binding cassette subfamily B member 1B that correlated BPs and urinary albumin concentration accompanied by an increase in portal taurocholic acid concentration. These results suggest that 12α-hydroxylated bile acids are involved in increased BP and albuminuria via alteration of hepatic function.

    Suppressor of mek1 (Dictyostelium) homolog 2 (Smek2), was identified as one of the responsible genes for diet-induced hypercholesterolemia (DIHC) of exogenously hypercholesterolemic (ExHC) rats. A deletion mutation in Smek2 leads to DIHC via impaired glycolysis in the livers of ExHC rats. The intracellular role of Smek2 remains obscure. We used microarrays to investigate Smek2 functions with ExHC and ExHC.BN-Dihc2BN congenic rats that harbor a non-pathological Smek2 allele from Brown-Norway rats on an ExHC background. Microarray analysis revealed that Smek2 dysfunction leads to extremely low sarcosine dehydrogenase (Sardh) expression in the liver of ExHC rats. Sarcosine dehydrogenase demethylates sarcosine, a byproduct of homocysteine metabolism. The ExHC rats with dysfunctional Sardh developed hypersarcosinemia and homocysteinemia, a risk factor for atherosclerosis, with or without dietary cholesterol. The mRNA expression of Bhmt, a homocysteine metabolic enzyme and the hepatic content of betaine (trimethylglycine), a methyl donor for homocysteine methylation were low in ExHC rats. Results suggest that homocysteine metabolism rendered fragile by a shortage of betaine results in homocysteinemia, and that Smek2 dysfunction causes abnormalities in sarcosine and homocysteine metabolism.

    In our previous study, enterohepatic 12α-hydroxylated (12α) bile acid (BA) levels were found to be correlated with hepatic triacylglycerol concentration in rats fed high-fat (HF) diet. Since BA composition is diverse depending on animal species, we evaluated whether such a relationship is applicable in mice in response to an HF diet. C57BL/6JJmsSLC (B6) male mice were fed HF diet for 13 weeks and analyzed for triacylglycerol, cholesterol, oxysterols, and other metabolites in the liver. The BA composition was determined in the liver, small intestinal contents, portal plasma, aortic plasma, and feces. Neutral sterols were also measured in the feces. The ratio of 12α BA/non-12 BA increased in the liver, portal plasma, small intestinal contents, and feces of HF-fed B6 mice. Moreover, a positive correlation was observed between the ratio of fecal 12α BAs/non-12 BAs and hepatic triacylglycerol concentration. The concentration of 7α-hydroxycholesterol was increased in the liver of HF-fed B6 mice, whereas no increase was observed in the hepatic expression of cytochrome P450 family 7 subfamily A member 1. The present study showed that the ratio of 12α BA/non-12 BA in feces is closely associated with hepatic triacylglycerol accumulation in B6 mice fed HF diet.

    Dietary phospholipids have been traditionally known to affect micelle formation. Egg yolk-derived lysophospholipids (LysoPL) are commercially available. We investigated the effects of dietary LysoPL on lymphatic lipid transport. We also compared sn-1 LysoPL and sn-2 LysoPL, which have different fatty acyl esterification positions. Thoracic lymph duct-cannulated rats were fed a diet supplemented with egg yolk-derived sn-1 LysoPL, sn-2 LysoPL, or phospholipids (PL). The amount of lymphatic lipid transport was also evaluated. Time courses of transport were applied to the one-compartment model as one of the pharmacokinetic analyses. The solubility of cholesterol in bile acid micelles was measured. Compared to the PL diet, the sn-1 and sn-2 LysoPL diets significantly reduced the lymphatic transport of cholesterol. There were no differences in the lymphatic PL and TAG transport. There was no difference in cholesterol transport between the sn-1 LysoPL group and the sn-2 LysoPL group; however, the transport rate constant at a decrease in lymphatic cholesterol was lower in the sn-1 LysoPL group than in the sn-2 LysoPL group. Cholesterol solubility in bile acid micelles was significantly decreased in the sn-1 LysoPL and sn-2 LysoPL groups compared to that in the PL group. Dietary LysoPL affects the behavior of intestinal cholesterol and suppresses lymphatic cholesterol transport.

    A dietary symbiotic supplement of okara, a by-product of tofu manufacturing with Bacillus coagulans lilac-01 found to reduce secondary bile acid (BA) in a rat study and cholic acid (CA), a primary 12α-hydroxylated (12αOH) BA, induces lipid accumulation in rats. In this study, we investigated whether dietary supplementation of a synbiotic with okara and Bacillus coagulans lilac-01 improves CA-induced hepatic lipid accumulation. Male Wistar/ST rats (4 weeks old) were fed with or without CA-supplementation combined with the synbiotic for 2 weeks. The CA diet increased hepatic triglycerides and the synbiotic diet in combination with CA normalized liver triglyceride concentration accompanied by suppression of hepatic fatty acid synthase expression, enhancement of fecal triglyceride excretion, and increase in fecal non-12αOH BA excretion. Dietary supplementation with the okara synbiotic ameliorated hepatic lipid accumulation probably by reducing de novo lipogenesis and enhancing fecal triglyceride excretion.

    There is an increasing need to explore the mechanism of the progression of non-alcoholic fatty liver disease. Steroid metabolism is closely linked to hepatic steatosis and steroids are excreted as bile acids (BAs). Here, we demonstrated that feeding WKAH/HkmSlc inbred rats a diet supplemented with cholic acid (CA) at 0.5 g/kg for 13 weeks induced simple steatosis without obesity. Liver triglyceride and cholesterol levels were increased accompanied by mild elevation of aminotransferase activities. There were no signs of inflammation, insulin resistance, oxidative stress, or fibrosis. CA supplementation increased levels of CA and taurocholic acid (TCA) in enterohepatic circulation and deoxycholic acid (DCA) levels in cecum with an increased ratio of 12α-hydroxylated BAs to non-12α-hydroxylated BAs. Analyses of hepatic gene expression revealed no apparent feedback control of BA and cholesterol biosynthesis. CA feeding induced dysbiosis in cecal microbiota with enrichment of DCA producers, which underlines the increased cecal DCA levels. The mechanism of steatosis was increased expression of Srebp1 (positive regulator of liver lipogenesis) through activation of the liver X receptor by increased oxysterols in the CA-fed rats, especially 4β-hydroxycholesterol (4βOH) formed by upregulated expression of hepatic Cyp3a2, responsible for 4βOH formation. Multiple regression analyses identified portal TCA and cecal DCA as positive predictors for liver 4βOH levels. The possible mechanisms linking these predictors and upregulated expression of Cyp3a2 are discussed. Overall, our observations highlight the role of 12α-hydroxylated BAs in triggering liver lipogenesis and allow us to explore the mechanisms of hepatic steatosis onset, focusing on cholesterol and BA metabolism.

    近年、高食肉摂取において、健康増進に寄与する一方、疾患発症・進展にも関与するという相反する効果が疫学的な調査から実証されている。しかし、この要因に関する詳細は、未だ不明である。当研究グループでは現在までに、腸内で産生される摂取タンパク質由来の腸内細菌代謝産物インドール系化合物の種類と量の違いが、各種組織の機能障害を導く一方、それら障害に対して予防・改善効果を導く事を示唆する結果を得
    ている。よって本研究では、「各種インドール系化合物が臓器機能に与える影響とその作用メカニズムの解明」、および「腸内におけるインドール系化合物の代謝促進に寄与する因子の同定」を試みる。

    本研究は代表者らが食事誘導性高コレステロール血症の原因遺伝子として同定した、遺伝子Smek2の機能性の解明に取り組むものである。Smek2の機能をより厳密に解析するため、ゲノム編集技術を利用してSmek2ノックアウトラットを確立する。また、Smek2の機能異常によって食事誘導性高コレステロール血症以外の病態の発生が予測されるため、その側面を評価し、新規モデル動物としての可能性も探索する。
