In many taxonomic research laboratories and museums, securing maintenance funds for the storage space, as well as air conditioning and humidity control necessary for maintaining a vast number of specimens, poses a significant challenge. The recent news of the National Museum of Nature and Science, Tokyo, successfully reaching its goal of 100 million yen through a large-scale crowdfunding campaign within just one day has sparked discussion. This achievement underscores the magnitude of the issue and the level of interest among individuals concerned with this matter.

The vast number of insect specimens held by research institutions are utilized as research materials but are not extensively utilized beyond this purpose. Therefore, in this project, we have initiated a pilot experiment to digitize and manage these insect collections as digital assets using blockchain technology. We have explored methods to utilize and monetize these assets, such as converting valuable insect materials owned by Kyushu University into Non-Fungible Tokens (NFTs). These tokens include ultra-high-resolution images that can be enlarged to microscopic levels. By integrating blockchain technology with Kyushu University’s insect specimens and researchers’ analytical techniques, we aim to build a new self-sustaining ecosystem capable of generating maintenance funds for these specimens from the accumulated research activities. Additionally, through NFTs, we aim to establish a community that interacts with natural history materials, thereby striving to create a socially and economically stable ecosystem.

While our project focuses on insect specimens and related materials housed at Kyushu University, the concept of utilizing natural history materials accumulated through research and conservation activities as a new source of funding can be adapted to other organisms and research fields. For instance, video footage and activity records captured during conservation activities can also be digitized and managed as NFTs. Thus, our project is expected to serve as a model case for various conservation and research activities that require continuous and stable economic foundations.

The impact of large-scale chromosomal rearrangements, such as fusions and fissions, on speciation is a longstanding conundrum. We assessed whether bursts of change in chromosome numbers resulting from chromosomal fusion or fission are related to increased speciation rates in Erebia, one of the most species-rich and karyotypically variable butterfly groups. We established a genome-based phylogeny and used state-dependent birth-death models to infer trajectories of karyotype evolution. We demonstrated that rates of anagenetic chromosomal changes (i.e., along phylogenetic branches) exceed cladogenetic changes (i.e., at speciation events), but, when cladogenetic changes occur, they are mostly associated with chromosomal fissions rather than fusions. We found that the relative importance of fusion and fission differs among Erebia clades of different ages and that especially in younger, more karyotypically diverse clades, speciation is more frequently associated with cladogenetic chromosomal changes. Overall, our results imply that chromosomal fusions and fissions have contrasting macroevolutionary roles and that large-scale chromosomal rearrangements are associated with bursts of species diversification.

As part of promoting digital transformation (DX) in the field of entomology, we present a method for drawing morphological illustrations of insects using digital devices. The method presented here uses an iPad, Apple pencil, and ibis Paint. The combination of the iPad and Apple pencil is superior in imitating the sensation of drawing with a pen on paper, and the use of the ibis Paint is advantageous in almost all the functions required to create scientific illustrations.

For figures used in scientific papers, the recommended canvas size of the ibis Paint is 2400×3200 pixels with a resolution of 600 dpi. The following steps are recommended for the ibis Paint: first, in the “Settings” set “Use Pressure Sensitivity” and “Palm Rejection” to on, “Quick Eyedropper” to off, and “Stabilizer” to the maximum value of 10 for ease of use. Next, set the following five brushes to Custom Brush: Pencil #1, Dip Pen (Hard), Dashed Line, Outline (Fade), and Felt Tip Pen (Soft).

The layer function is utilized in the drawing process. The drafting procedure differs depending on whether the tracing or grid method is used. In the tracing method, the photograph to be traced is imported and set on the bottom layer. After adjusting the position, angle, and transparency of the photo, create a new layer for the draft and use “Pencil #1” to write the draft. For the grid method, start “Filter” mode and use the filter “Table (Size)” to create a grid. After placing the grid layer on the bottom layer, create a new layer for drafting and write a draft using “Pencil #1”.

For inking (final drawing), use “Dip Pen (Hard)” to draw the outline first, and then add details in sequence. Use “Dashed Line” for structures hidden behind, “Outline (Fade)” for soft hairs, and “Felt Tip Pen (Soft)” for dots. Using different layers for different purposes, such as shadows, three-dimensional expressions, and hairs, makes revising later easier. Screen tones can be used to fill in arbitrary areas with slash or cross lines.

The method presented here can easily applied to simulate traditional drawing techniques. Thus, it can be adopted by different users, from senior researchers who already have established techniques to students who intend to learn scientific drawing. However, this method is still in its infancy, and it is necessary to continue to develop improved methods. Therefore, sharing the practical usage of digital devices with the entire entomological community is also essential.

  • The bug-killer fly Gymnosoma rotundatum (L.) (Diptera: Tachinidae) forms the respiratory funnel independently of the host's immune response

In internal parasitism, the respiration strategy within the host's body is as essential as evading attack from the host's immune system. Tachinid flies are parasitoids of terrestrial arthropods, mostly insects, during their larval stage. To obtain oxygen while living in the host body, they build a cylindrical structure known as the respiratory funnel at the aperture opened by the tachinid larva on the host integument or trachea. These funnels can be divided morphologically into sheath and cone types. Previous research on sheath-type funnels revealed that they are derived from the encapsulating substance produced by the host's immune system. In contrast, the cone-type funnels cover part of the body of the larval tachinid and may be constructed independently from the host immune system. To determine the mechanisms of cone-type funnel formation, histological observations were carried out on Gymnosoma rotundatum (L.) (Diptera: Tachinidae), which possesses this type of funnel. The respiratory funnel of G. rotundatum was found to be derived from the tube-shaped faeces wrapped with the peritrophic membrane and excreted by the fly larva, not from host tissue or haemocytes. Additionally, secretory glands putatively involved in the funnel formation were discovered around the larval anal plate of G. rotundatum. A comparison of funnel types within Tachinidae revealed that Phasiinae and Dexiinae have cone-type funnels, which may be created by the same mechanism as in G. rotundatum. These new findings suggest that funnel formation that does not use the host immune system is relevant to tachinid phylogeny.

One of the factors contributing to the ecological success of ants is the acquisition of colony founding strategies, such as claustral independent colony foundation (claustral ICF) and colonial budding. We investigated tissue changes during claustral ICF in Lasius japonicus and explored differentially expressed genes associated with each strategy. Our findings revealed specialized esophageal structures in claustral founding queens for thoracic crop formation, along with elevated expression of genes involved in flight muscle development and energy metabolism. Although we couldn't identify gene expression associated with colonial budding, our study expanded the resources for comprehensive gene expression information. Further utilization of these resources is needed to identify genes associated with colonial budding in future research.
