    The glycomic analysis holds significant appeal due to the diverse roles that glycans and glycoconjugates play, acting as modulators and mediators in cellular interactions, cell/organism structure, drugs, energy sources, glyconanomaterials, and more. The glycomic analysis relies on liquid-phase separation technologies for molecular purification, separation, and identification. As a miniaturized form of liquid-phase separation technology, microscale separation technologies offer various advantages such as environmental friendliness, high resolution, sensitivity, fast speed, and integration capabilities. For glycan analysis, microscale separation technologies are continuously evolving to address the increasing challenges in their unique manners. This review discusses the fundamentals and applications of microscale separation technologies for glycomic analysis. It covers liquid-phase separation technologies operating at scales generally less than 100 µm, including capillary electrophoresis, nanoflow liquid chromatography, and microchip electrophoresis. We will provide a brief overview of glycomic analysis and describe new strategies in microscale separation and their applications in glycan analysis from 2014 to 2023.

    Proteins are typically separated by an immune reaction, such as an enzyme-linked immunosorbent assay, and are detected by selective fluorescent labeling. This has potential for complicated procedures and the denaturation of proteins by labeling, and is cost consuming. In this study, we propose a technique for the selective separation and detection of a target protein using a molecularly imprinted hydrogel (PI gel) with fluorescent monomers. We focused on 8-anilino-1-naphthalenesulfonic acid (ANS), where the fluorescence intensity is easily changed by the interaction with proteins, and successfully synthesized the ANS monomer and a poly(ethylene glycol) (PEG) conjugated ANS monomer. The PI gel with the ANS monomers using bovine serum albumin (BSA) as a template showed the selective adsorption of BSA and the fluorescence intensity increased due to the adsorption of BSA.

    A hydrogel is a solid form of polymer network absorbed in a substantial amount of aqueous solution. In electrophoresis, hydrogels play versatile roles including as support media, sieving matrixes, affinity scaffolds, and compositions of molecularly imprinting polymers. Recently, the study of hydrogels has been advancing with unprecedented speed, and the application of hydrogels in separation science has brought new opportunities and possible breakthroughs. A good understanding about the roles and effects of the material is essential for hydrogel applications. This review summarizes the hydrogels that has been described in various modes of electrophoretic separations, including isoelectric focusing gel electrophoresis (IEFGE), isotachophoresis (ITP), gel electrophoresis and affinity gel electrophoresis (AGE). As microchip electrophoresis (ME) is one of the future trends in electrophoresis, thought provoking studies related to hydrogels in ME are also introduced. Novel hydrogels and methods that improve separation performance, facilitate the experimental operation process, allow for rapid analysis, and promote the integration to microfluidic devices are highlighted.

    The use of Sudan black B as coloring agent in foods is forbidden for its toxicology effect on human organs. This work proposes an efficient and sensitive method for food security inspection targeting Sudan black B. Surface-enhanced Raman spectroscopy (SERS) is applied to the analysis of trace Sudan black B. It could be detected at concentrations as low as 0.05 mg/L in standard solutions and 0.1 mg/kg in black rice extracts with the SERS method for measurement. The linear relationship between the intensity and concentration could be used for the quantitative detection of Sudan black B. The relation between dyeing time of black rice stained by Sudan black B solution and SERS intensity was studied which indirectly showed the effectiveness of the extraction method we designed. The results of the quantitative analysis reveal the practicability of using this method to detect Sudan black B in black rice. As a rapid and sensitive detection method, SERS can be extended to detect other food products and has a great application prospect in food safety inspection. (C) 2019 Elsevier B.V.     This paper demonstrated the quantitative detection of ethanol and acetone mixtures in complex solutions with Raman spectroscopy based on headspace gas analysis. By analyzing the volatile components in the headspace, their concentrations in liquid solutions were determined. We constructed our own Raman spectroscopy system to detect the headspace gas quantitatively over a solution in a sealed vial. The Raman spectra of the headspace gases over standard solutions were standardized for finding the concentrations of ethanol, acetone, and ethanol-acetone in mixture solutions. The results showed that the concentration of a gaseous component in the headspace gas was proportional to its ratio in the liquid solution. We obtained a linear relationship between the spectral intensity of volatile components in headspace and the concentration of the liquid solutions. Then, we analyzed the alcohol concentration in a white wine and a Chinese liquor called Fen Chiew by measuring the Raman spectra of the headspace gas over their liquids. For the river water sample, we also implemented our headspace gas detection with Raman spectra to obtain the concentration of acetone in the river sample. This work demonstrated the facilitation of headspace gas analysis by the qualitative and quantitative determination of volatile substances from liquid samples using Raman spectroscopy.
