    Ferroptosis, a form of cell death instigated by iron-dependent lipid peroxidation reactions (LPO), is emerging as a promising therapeutic target for cancer. While the mechanisms governing LPO induction and suppression have gradually been unveiled, questions persist regarding the specific cellular location of LPO and the utilization of iron in driving cell death. A comprehensive understanding of these aspects holds significant potential for advancing therapeutic applications in disease management. Here, we show lysosomal LPO in the initiation of ferroptosis, leveraging the hidden abilities of fluorescent detection probes. Intra-lysosomal LPO triggers iron leakage, fostering cell-wide LPO by augmenting lysosomal membrane permeabilization (LMP). Conversely, cell lines with low susceptibility to ferroptosis do not exhibit LMP. This deficiency is rectified by the concurrent administration of chloroquine, leading to LMP induction and subsequent cell death. These findings underscore enhancing LMP induction efficacy as a strategic approach to surmount resistance to therapies in cancer.

    Glucosylceramide (GlcCer) belongs to sphingolipids and is found naturally in plant foods and other sources that humans consume daily. Our previous studies demonstrated that GlcCer prevents inflammatory bowel disease both in vitro and in vivo, whose patients are increasing alarmingly. Although some lipids are vulnerable to oxidation which changes their structure and activities, it is unknown whether oxidative modification of GlcCer affects its activity. In this research, we oxidized GlcCer in the presence of a photosensitizer, analyzed the oxide by mass spectrometric techniques, and examined its anti-inflammatory activity in lipopolysaccharide (LPS)-treated differentiated Caco-2 cells as in vitro model of intestinal inflammation. The results showed that GlcCer is indeed oxidized, producing GlcCer hydroperoxide (GlcCerOOH) as a primary oxidation product. We also found that oxidized GlcCer preserves beneficial functions of GlcCer, suppressing inflammatory-related gene expressions. These findings suggested that GlcCerOOH may perform as an LPS recognition antagonist to discourage inflammation rather than induce inflammation.

    Platelet-activating factor receptor (PAFR) is involved in various physiological processes, including the immune system and inflammatory responses. In addition to PAF, several oxidized phospholipids have been shown to act as ligands for PAFR. We have previously developed a comprehensive analysis method for oxidized phospholipids, and in this study, we employed this method to test whether additional oxidized phospholipids can activate PAFR. From an oxidized phosphatidylcholine mixture, we identified that 1-palmitoyl-2-(4′-oxo-butanoyl)-sn-glycero-3-phosphocholine (POBPC) functions as a novel PAFR activator, using preparative HPLC and comprehensive LC-MS/MS analysis of fractionated oxidized phospholipids. Next, multiple assays confirmed that POBPC acts as a bona fide PAFR agonist. The H248W mutation of PAFR attenuated the response to POBPC. Finally, POBPC induced phosphorylation of extracellular signal-regulated kinase in mouse peritoneal macrophages, which endogenously express PAFR. Our findings provide valuable insight into the biological functions of oxidized phospholipids, advancing our understanding of their roles in cellular processes.

    <p>Recently, mitochondrial dysfunction has gained attention as a causative factor in the pathogenesis and progression of age-related macular degeneration (AMD). Mitochondrial damage plays a key role in metabolism and disrupts the balance of intracellular metabolic pathways, such as oxidative phosphorylation (OXPHOS) and glycolysis. In this study, we focused on oxidized low-density lipoprotein (ox-LDL), a major constituent of drusen that accumulates in the retina of patients with AMD, and investigated whether it could be a causative factor for metabolic alterations in retinal pigment epithelial (RPE) cells. We found that prolonged exposure to ox-LDL induced changes in fatty acid β-oxidation (FAO), OXPHOS, and glycolytic activity and increased the mitochondrial reactive oxygen species production in RPE cells. Notably, the effects on metabolic alterations varied with the concentration and duration of ox-LDL treatment. In addition, we addressed the limitations of using ARPE-19 cells for retinal disease research by highlighting their lower barrier function and FAO activity compared to those of induced pluripotent stem cell-derived RPE cells. Our findings can aid in the elucidation of mechanisms underlying the metabolic alterations in AMD.</p>

    加齢黄斑変性患者の網膜に蓄積するドルーゼンの主要成分である酸化LDL(oxLDL)に着目し、oxLDLが網膜色素上皮(RPE)細胞の代謝に及ぼす影響について検討した。ox-LDLへの長時間曝露はRPE細胞の脂肪酸β酸化(FAO)、酸化的リン酸化および解糖活性を変化させ、ミトコンドリアの活性酸素種産生を増加させた。代謝に対する影響は、ox-LDL処理の濃度および時間により変化した。多能性幹細胞由来RPE細胞と比べてARPE-19細胞のバリア機能およびFAO活性は低く、網膜疾患の研究にARPE-19細胞を用いることの限界が示された。

    Milk is a rich source of essential nutrients such as lipids. However, lipid oxidation can be considered a crucial factor in determining the initial stage of milk deterioration. Therefore, it is essential to identify the mechanisms of lipid oxidation, such as photo-oxidation or thermal oxidation, to efficiently prevent it by selecting proper antioxidants. In this study, the oxidation mechanisms of long-life (LL) milk were investigated, and triacylglycerol hydroperoxide isomers generated corresponding to the oxidation mechanisms were analyzed by LC-MS/MS. This study first prepared the standard of TG 4:0_16:0_18:1;OOH isomers, which are the appropriate target for evaluating LL milk's oxidation mechanism. The authentic standards provided the robust analysis of TG 4:0_16:0_18:1;OOH isomers and suggested that LL milk was susceptible to photo-oxidation rather than thermal-oxidation. Furthermore, it was discovered that radicals play a role in the oxidation of LL milk during photo-oxidation. This information could be valuable in effectively preventing photo-oxidation in LL milk. It is important to note that milk is contained in a variety of food products. Hence, these findings would be applicable not only to milk but also to various milk-containing food products.

    <p>Lipid peroxidation generates numerous oxidized lipids and has shown its essential role in cell biology and human health, in which oxidized phospholipids (OxPLs) are the crucial lipids reportedly involved in the pathogenesis and progression of numerous diseases. Although OxPLs are known to modulate various biological functions, their precise role remains an ongoing debate due to the lack of structural information and dedicated analytical methods. In this study, we used mass spectrometry and HPLC techniques to explore and isolate novel types of OxPLs, such as oxygenated and truncated acyl chains of phospholipids, and then purifying by preparative HPLC. <b>It has been found that the production of oxidized lipids varies based on the oxidation methods. Additionally, we discovered that novel OxPLs identified in this study indicate multiple activities relating to inflammation and cell signaling.</b> The modification of phospholipids by oxidation is one of the critical determinants of its interactions with other molecules, proteins, and the lipid bilayer, influencing membrane properties and activities. <b>Developing the OxPLs library using mass spectrometry and OxPLs purifying are expected to provide a comprehensive understanding of OxPLs involved in diseases and pave the way for proposing candidate compounds for innovative therapeutic strategies in lipid peroxidation-related diseases.</b></p>
