    The variation of gene expression of seeds or leaves of Arabidopsis thaliana was investigated by irradiation with oxygen and air plasmas. The irradiation with oxygen plasma reported on the growth promotion and induced the consequence of gene expression in plant cells by neutral active oxygen species. The increase in leaf area ratio by oxygen plasma to seeds was due to epigenetics such as activation of DNA demethylation transcription factors and the growth enhancement effect induced by the plasma irradiation of seeds was inherited by next-generation cells through cell division even after germination. In oxygen irradiation for 10 s, expression of each de-DNA methylation-related gene increased, and DNA methylation-related genes decreased in expression. DNA acetylation that induces gene expressions was suppressed. However, irradiated for 20 s by oxygen, both demethylation suppression and promotion and methylation/acetylation suppression and promotion were obtained. On the other hand, methylation and demethylation may occur at the same time but were not significant and the acetylation was suppressed by air plasma irradiation. In both cases, active oxygen species was the key factor for the variation of gene expression.

    The sterilization characteristics of active species generated by an atmospheric dielectric barrier discharge plasma using air and oxygen at the inner surface of silicone tubing were investigated. A dielectric barrier discharge torch plasma device was installed at one end of the tube and generated long-lived active species that flowed into the tube. A strip-type biological indicator with a 105-cell bacterial spore was placed at the opposite end of the 60 cm tube. Sterilization was completed within 30 min by active particles generated from the air plasma. The main factors contributing to the sterilization by air plasma were HNO3 and N2O5. When organic materials (keratin, aspartic acid, and dipicolinic acid) reflecting components of the bacterial spore, were treated by the sterilization procedure there was little effect on dipicolinic acid. Keratin was oxidized by ozone and NOx generated from the oxygen and air plasmas, respectively. Aspartic acid underwent little change in composition from ozone generated from the oxygen plasma, whereas nitro (NO2), nitroso (NO), and aldehyde (CHO) groups were formed from ozone and NOx generated from the air plasma.

    Gene expression variations of plant leaf are investigated by irradiating seed and leaf with oxygen or air plasmas. Enhancement of leaf growth is induced by oxygen plasma irradiation on seeds, which is supported by increased gene expression for protein synthesis, oxidative-reduction reactions, and decreased gene expression concerning DNA methylation and histone modification. Suppression of leaf growth is observed by the oxygen plasma, which would be owing to increased gene expression concerning heat shock protein and redox reaction, and decreased expression of photosynthesis and glycoprotein. Moreover, gene expression variation due to air plasma irradiation is almost the same as that of oxygen plasma. Active oxygen species are major factors in both oxygen and air plasmas for the variation of gene expressions in plants.

    Recently, pulsed power technologies, including pulsed electric fields (PEFs) and time-modulated plasmas, are starting to be applied actively in agriculture and food processing. In the applications, compact pulsed power generators with moderate peak power and repetitive operation are developed for controlling discharge plasmas and electric field distribution. These applications are mainly based on the biological effects of a spatially distributed electric field and the chemically active species in the plasma. The PEFs are caused by applying pulse voltage between the electrodes and contribute to form pores on the cell membrane or to change conformation of protein. When the applied voltage exceeds the discharge onset criterion, plasmas are generated through the avalanche process of electron accelerated with intense electric field in a gas or liquids medium.

    Zinc ion dissolved in water is attempted to be removed by generating the oxides of zinc using the oxygen gas in DBD plasma system. The removal rate of zinc oxides’ production (ZnO and Zn (OH2)) were measured at different treatment periods by the oxygen plasma penetration in water. The removal rate of the deposit increases initially and then decreases with the treatment period. The maximum removal rate (29%) of zinc from water is achieved at the treatment period of 10 min, where pH is minimum. From FTIR the generation properties of zinc oxide can be recognized. Initially the amount the deposit increases with the ozone treatment period due to production of both ZnO and Zn (OH) 2. After that, the production of Zn (OH) 4 2-increases even when the total removal rate of the deposit decreases. Therefore, to remove zinc ion from water forming metal oxide deposit, the penetration amount of the active oxygens to the water must be controlled to keep the pH lower than around 7.5. Because with increasing pH amount of removal rate of ZnO deposit decreases. The pH of the zinc dissolved water treated by ozone depends on both zinc and ozone concentration in water.

    The activation of EL-4 T-cells is investigated when the T-cells are sensitized with monoclonal antibodies against CD3/CD28 followed by irradiation with atmospheric oxygen plasma. The proliferation of anti-CD3/CD28-sensitized EL-4 T-cells was evaluated as a function of the oxygen plasma irradiation duration. The plasma irradiation promotes cell proliferation: when plasma irradiation was applied to sensitize T-cells for 40 s, the cell number increased by five times over 24 h compared with that in a cell population that was not exposed to irradiation. Also, the plasma irradiation increases the production of interferon gamma in T-cells by a factor of two. Gene expression analysis of T-cell supports results of the proliferation and the immune function enhancements.

    Oral cancer cell HSC3 is inactivated by the direct irradiation of oxygen plasma generated by torch-type oxygen dielectric barrier discharge. Selective inactivation of the cancer cells is observed at the discharge voltage of 4.2 kV, where normal cells suffer no damages from the plasma. Inactivation mechanism of HSC3 cells irradiated by the plasma is considered as the apoptosis. Activation of mitogen-activated protein kinase and p53 proteins owing to the phosphorylation is one of the pathways to the cancer cell apoptosis.

    Sterilization of non-heat-proof equipments was
    performed by irradiation from air torch plasma produced by
    microwave discharge. The downstream region of the torch
    plasma exhibits a yellowish color. Light emission spectra and chemical indicators show that nitrogen oxide radicals are the origin of this color. Inactivation of spore-type Geobacilius stearothermophilus with the population of 105 was successful for a treatment period of 10 minutes.

    Submicron dust particles (Cu, ϕ 50 nm) were observed to be trapped in an rf plasma at relatively low pressure. The laser light scattering by particles shows the time evolution of the dust particles for several hours after pulsed injection of the particles. The dust particles were localized in the diffused region as a large volume cloud. The suspension mechanism of the dust particles in the diffused plasma was explained by an electrostatic force due to the potential structure. The spatial distribution of the charge of the dust particles was determined by this potential structure. The dispersion relations of the self-excited dust acoustic waves were adopted to confirm the spatial distribution and the time evolution of the charge of the dust particles.

    The impact of cold atmospheric plasma (CAP) treatment on the enamel of twelve primary teeth (incisors, canines, and molars) collected from six children was examined in order to evaluate the possibility of using the CAP technique in dental applications. A radio-frequency dielectric barrier discharge (DBD) plasma jet operating at a voltage of 3.25 kV using a mixture of helium and oxygen as the working gas was used for the generation of plasma as part of the electro-technological method for the treatment of biological material. The plasma exposure time for the primary teeth was 5, 10, and 20 min. The properties of tooth enamel (color, contact angles, surface roughness, surface topography, elemental composition) were examined before (control) and after the plasma treatment. As shown by the results, the plasma treatment time is a key parameter that can induce desired features, such as whitening or improved wettability. However, with prolonged plasma treatment (20 min), the enamel surface may be permanently damaged. The cold-plasma-treated samples were characterized by a higher value of the brightness L* parameter and thus a lighter color, compared to the CAP-untreated teeth. It was also evidenced that the plasma treatment increased the hydrophilicity of tooth surfaces, and the contact angles effectively decreased with the time of the CAP treatment. The tooth surface also became much more heterogeneous and rough with much greater amplitudes in heights. The surface of the primary teeth after the CAP treatment lost its homogeneity, as evidenced by the SEM micrographs. The analysis of the elemental composition revealed only minor changes after the plasma process, which may suggest that the observed morphological changes in the enamel surface are mainly physical and are not a consequence of chemical reactions between the enamel and the reactive components of the cold plasma. Plasma treatment of teeth opens up new possibilities of using this method as an alternative to whitening or pre-treatment before other dental procedures.

    This study is committed to enhancing the photoresponse efficiency of titanium dioxide (TiO2) nanotubes through meticulous fabrication and optimization, focusing on using air plasma treatment. Various parameters, including oxidation voltage, concentration of ammonium fluoride (NH4F), and reaction time, exert considerable influence over the structural integrity and dimensions of the TiO2 nanotubes. The photoresponse characteristics of TiO2 nanotubes prepared at different reaction conditions under UVC light irradiation exhibited the critical role of morphology in enhancing photoresponse performance. Introducing cost-effective air plasma treatment can enhance surface defects and charge transfer efficiency, thereby improving photoresponse efficiency without damaging TiO2 nanotubes. Furthermore, the enhanced photocatalytic performance of air plasma-treated TiO2 nanotubes was studied under crystal violet degradation, and the results showed significantly improved efficiency compared to TiO2 nanotubes. These findings suggest that the synergistic optimization of fabrication processes and the incorporation of air plasma treatment contribute substantially to developing TiO2 nanotubes endowed with superior photoresponsive characteristics. The implications of this advancement extend to promising applications in environmental remediation and advanced photocatalysis.

    Planetary protection is a guiding principle aiming to prevent microbial contamination of the solar system by spacecraft (forward contamination) and extraterrestrial contamination of the Earth (backward contamination). Bioburden reduction on spacecraft, including cruise and landing systems, is required to prevent microbial contamination from Earth during space exploration missions. Several sterilization methods are available; however, selecting appropriate methods is essential to eliminate a broad spectrum of microorganisms without damaging spacecraft components during manufacturing and assembly. Here, we compared the effects of different bioburden reduction techniques, including dry heat, UV light, isopropyl alcohol (IPA), hydrogen peroxide (H2O2), vaporized hydrogen peroxide (VHP), and oxygen and argon plasma on microorganisms with different resistance capacities. These microorganisms included Bacillus atrophaeus spores and Aspergillus niger spores, Deinococcus radiodurans, and Brevundimonas diminuta, all important microorganisms for considering planetary protection. Bacillus atrophaeus spores showed the highest resistance to dry heat but could be reliably sterilized (i.e., under detection limit) through extended time or increased temperature. Aspergillus niger spores and D. radiodurans were highly resistant to UV light. Seventy percent of IPA and 7.5% of H2O2 treatments effectively sterilized D. radiodurans and B. diminuta but showed no immediate bactericidal effect against B. atrophaeus spores. IPA immediately sterilized A. niger spores, but H2O2 did not. During VHP treatment under reduced pressure, viable B. atrophaeus spores and A. niger spores were quickly reduced by approximately two log orders. Oxygen plasma sterilized D. radiodurans but did not eliminate B. atrophaeus spores. In contrast, argon plasma sterilized B. atrophaeus but not D. radiodurans. Therefore, dry heat could be used for heat-resistant component bioburden reduction, and VHP or plasma for non-heat-resistant components in bulk bioburden reduction. Furthermore, IPA, H2O2, or UV could be used for additional surface bioburden reduction during assembly and testing. The systemic comparison of sterilization efficiencies under identical experimental conditions in this study provides basic criteria for determining which sterilization techniques should be selected during bioburden reduction for forward planetary protection.

    Planetary protection is a guiding principle aiming to prevent microbial contamination of the solar system by spacecraft (forward contamination) and extraterrestrial contamination of the Earth (backward contamination). Bioburden reduction on spacecraft, including cruise and landing systems, is required to prevent microbial contamination from Earth during space exploration missions. Several sterilization methods are available; however, selecting appropriate methods is essential to eliminate a broad spectrum of microorganisms without damaging spacecraft components during manufacturing and assembly. Here, we compared the effects of different bioburden reduction techniques, including dry heat, UV light, isopropyl alcohol (IPA), hydrogen peroxide (H2O2), vaporized hydrogen peroxide (VHP), and oxygen and argon plasma on microorganisms with different resistance capacities. These microorganisms included Bacillus atrophaeus spores and Aspergillus niger spores, Deinococcus radiodurans, and Brevundimonas diminuta, all important microorganisms for considering planetary protection. Bacillus atrophaeus spores showed the highest resistance to dry heat but could be reliably sterilized (i.e., under detection limit) through extended time or increased temperature. Aspergillus niger spores and D. radiodurans were highly resistant to UV light.

    The variation of gene expression of seeds or leaves of Arabidopsis thaliana was investigated by irradiation with oxygen and air plasmas. The irradiation with oxygen plasma reported on the growth promotion and induced the consequence of gene expression in plant cells by neutral active oxygen species. The increase in leaf area ratio by oxygen plasma to seeds was due to epigenetics such as activation of DNA demethylation transcription factors and the growth enhancement effect induced by the plasma irradiation of seeds was inherited by next-generation cells through cell division even after germination. In oxygen irradiation for 10 s, expression of each de-DNA methylation-related gene increased, and DNA methylation-related genes decreased in expression. DNA acetylation that induces gene expressions was suppressed. However, irradiated for 20 s by oxygen, both demethylation suppression and promotion and methylation/acetylation suppression and promotion were obtained. On the other hand, methylation and demethylation may occur at the same time but were not significant and the acetylation was suppressed by air plasma irradiation. In both cases, active oxygen species was the key factor for the variation of gene expression.

    The sterilization characteristics of active species generated by an atmospheric dielectric barrier discharge plasma using air and oxygen at the inner surface of silicone tubing were investigated. A dielectric barrier discharge torch plasma device was installed at one end of the tube and generated long-lived active species that flowed into the tube. A strip-type biological indicator with a 10^v5-cell bacterial spore was placed at the opposite end of the 60 cm tube. Sterilization was completed within 30 min by active particles generated from the air plasma. The main factors contributing to the sterilization by air plasma were HNO_3 and N2O5. When organic materials (keratin, aspartic acid, and dipicolinic acid) reflecting components of the bacterial spore, were treated by the sterilization procedure there was little effect on dipicolinic acid. Keratin was oxidized by ozone and N_Ox generated from the oxygen and air plasmas, respectively. Aspartic acid underwent little change in composition from ozone generated from the oxygen plasma, whereas nitro (NO_2), nitroso (NO), and aldehyde (CHO) groups were formed from ozone and NOx generated from the air plasma.

    This paper presents the results of an experiment based on using a Glide-arc type plasma reactor operating at atmospheric pressure for the quality of fresh pressed tomato juice, variety Bekas. The impact of after-glow plasma gas (air) on the physicochemical, microbiological properties and morphology of the product’s samples was investigated. Five groups of juices characterized by different exposure times (30, 60, 120, 300 and 600 s), as well as untreated juice (as control) were used. The juice quality was assessed on days 1, 3, 5, and 10 of refrigerated storage. Significant increases were observed when Cold Atmospheric Plasma (CAP)-treated tomato juice was tested against total soluble solids, pH, lycopene, and vitamin C in comparison to the control treatments. Moreover, changes in the tested physicochemical values during the storage of juice subjected to the action of cold plasma did not progress as quickly as in the case of the control juice. A significant decrease was observed in total plate count, yeast, and mold after 300–600 s CAP treatment. The findings of the current study suggested that CAP treatment is a promising technique that could provide improved quality and stability during the processing of tomato juice with better physicochemical properties and bioavailable nutrients.

    Gene expression variations of plant leaf are investigated by irradiating seed and leaf with oxygen or air plasmas. Enhancement of leaf growth is induced by oxygen plasma irradiation on seeds, which is supported by increased gene expression for protein synthesis, oxidative-reduction reactions, and decreased gene expression concerning DNA methylation and histone modification. Suppression of leaf growth is observed by the oxygen plasma, which would be owing to increased gene expression concerning heat shock protein and redox reaction, and decreased expression of photosynthesis and glycoprotein. Moreover, gene expression variation due to air plasma irradiation is almost the same as that of oxygen plasma. Active oxygen species are major factors in both oxygen and air plasmas for the variation of gene expressions in plants.

    The plasma surface sterilization method has emerged as a new sterilization method for medical instruments with many advantages, including nontoxicity, short treatment time, and low thermal damage to the sterilized material, over conventional sterilization methods. In this study, low-frequency (LF)-microwave hybrid plasma is produced by irradiation of microwave to an LF plasma jet. This hybrid plasma is characterized by atmospheric-pressure nonthermal plasma with a large amount of reactive species and has been applied for plasma surface sterilization. To reduce the treatment temperature while maintaining the effect of surface sterilization for applicability to heat-sensitive materials, the LF-microwave hybrid plasma was modulated in time by using pulsed microwaves.

Because of these multitude of changes, the agricultural industry and researchers recognise a dire need for more sustainable agriculture practices and technologies. Novel technologies and methods of improving crop yields while reducing waste throughout the food supply chain are two ways that we can reduce the pressure on our already stretched food supplies.     Variations in osteoclast cell number are observed when osteoclast precursor cells are irradiated with atmospheric dielectric barrier discharge plasma. Active species generated by the oxygen plasma control the differentiation function of the osteoclast precursor cells. Long-lifetime active species such as H 2 O 2 and NO x–dissolved in the culture medium decrease the osteoclast number due to the inactivation of the differentiation function of the osteoclast precursor cells. When short-lifetime active species such as O* and OH* make contact with the osteoclast precursor cells directly, the osteoclast number tends to increase. Short-lifetime active species induce the enhancement of the gene expression of NFATc1.

    Oxygen plasma that is a kind of external biological stimulus activates the macrophage-like cell. Active oxygen species in the oxygen plasma-irradiated macrophage-like cells in culture medium to investigate the effects of different treatment periods and applied voltages. Macrophage-like cells were activated and increased 10% after 48 h from the short period plasma irradiation of 30 s. The long period irradiation for 60 and 120 s decreased the cell number by almost 33%. The phagocytosis capability increased by 1.5 times during the shorter plasma irradiation period, which would be owing to the increase of active oxygen species inside the macrophage cells.

    The sterilization of a small vial using electron cyclotron resonance (ECR) plasma was investigated. The ECR plasma was generated throughout the inside of the small vial, which was sealed with Tybek sheet, by using magnetic configuration in which the ECR point was located inside the small vial. Moreover, it was demonstrated that the temperature of the small vial was finely controlled by the pulse width and pulse frequency of the pulsed microwave power. A biological indicator (BI), Geobacillus Stearothermophilus, containing 1×10^5 spores present inside the small vial was successfully inactivated in a total treatment time of about 14 min while maintaining treatment temperature of less than 65 ºC.

    Silkworms have recently been proposed as an animal model for safety testing in basic research. We propose using silkworms for in vivo trials of direct plasma treatment. In this study, the influence of plasma irradiation on silkworms was investigated using a non-thermal atmospheric pressure plasma. Silkworm survival rate decreased with increasing low-frequency voltage and plasma irradiation period. Further investigation of the plasma-generated agents (oxygen related radicals, UV light, and charged particles), revealed that the contribution of charged particles significantly increases silkworm mortality.

    Intermittent chaotic phenomena caused by the current-driven ion acoustic instability are experimentally observed using a Double Plasma device, in which two mesh grids are installed to excite the instability. When a dc potential is applied to one of the two mesh grids and exceeds a certain threshold, the system suddenly transits from a periodic state to a chaotic state. At the same time, the signals picked up as perturbation components of a current exhibit intermittent turbulent bursts. The calculations of the correlation dimension and the Lyapunov exponent indicate that the system reaches a chaotic state. Furthermore, it is found that the results of mathematical and statistical analysis of observed signals agree with the theoretical properties of the type-1 intermittency: the occurrence of 1/f-type low-frequency noise and the probability distribution of the duration between two bursts. Therefore, it is concluded that the present system reaches a chaotic state via the type-1 intermittency.

    Bifurcation phenomena are experimentally observed in an electron beam plasma system using a double plasma device. Upon injection of an electron beam into the target plasma, both an unstable wave and subharmonics are observed. The fundamental unstable wave is classified as to be a beam mode wave excited by electron beam plasma instability. It is confirmed that these bifurcation phenomena originate from nonlinearity of the unstable wave.

    The excitation mechanism of standing waves produced by the electron beam plasma instability is experimentally studied using a Double Plasma device. When an electron beam is injected into the target plasma, standing waves around the electron plasma frequency are excited. A test wave is propagated in an electron beam plasma system and is identified as the beam mode from the dispersion relation. The propagation direction of the beam mode is determined from the wave pattern utilizing a phase shifter. It is found that a reflected beam mode exists as well as a forward beam mode, and is generated by the reflection of the forward beam mode from a potential well produced by the injection of the electron beam. The observed standing waves are formed by superposing the beam modes propagating in opposite directions from each other.

    Enhancements of germination and growth of plants are observed after oxygen plasma irradiation to seeds. Gene expressions in plant seeds irradiated by oxygen plasma were investigated using DNA microarray bioinformatics analysis to clarify the pathways responsible for growth enhancement of plants. Gene expressions involved in photosynthesis and energy production by active oxygen species in oxygen plasma are one of the factors for the growth enhancement of plants. The observed growth enhancement effect is not passed on to the next generation by irradiating seeds, and there is no significant change in gene expressions in second-generation seeds. The observed growth enhancement of plants is brought about by epigenetics.

    In this study, oxygen plasma irradiation to seeds was applied to the cultivation of agricultural products. So far, by irradiating the seeds of typical agricultural products such as carrots, cucumbers, and radishes with oxygen plasma, the effects of growth enhancement and shortening the cultivation time have been obtained. In this experiment, the seeds of Angelica acutiloba, which is a typical medicinal plant but has been difficult to obtain a stable yield, are irradiated with oxygen plasma to enhance germination, growth and medicinal content.

    Gene expression analysis of plant seeds and DNA methylation quantitative analysis were carried out to confirm epigenetics as a mechanism of expression of plasma irradiation effect. The increase in leaf area and total length of Arabidopsis thaliana is related to the irradiation dose of reactive oxygen species. The growth rate of the plant grown from the seed irradiated with plasma is faster than the growth rate of the untreated seed during the growing period. Gene expression of the seeds of Arabidopsis thaliana indicates that several dozen genes are upregulated due to oxygen plasma irradiation. Chromatin structure and the structure of the chromosomal region such as DNA methylation were found to be modified, indicating enhancement of energy production system. Reduction of histone H1.2 and increase of DME slowly induces gene winding around histones and demethylation of genes, respectively. Therefore, it was revealed that the irradiation effect of the oxygen plasma inherited by the epigenetic modification of the gene.

    Gene expression analysis and DNA methylation quantification analysis have been performed to confirm the epigenetics as the inheritance mechanism of the plasma irradiation effect. Reduction of histone H1.2 and increase of DME indicate loose of gene winding around histones and demethylation of genes, respectively. Loose of winding and demethylation of DNA suggests the increase of gene expressions. The epigenetic modification of genes occurs owing to the irradiation of active oxygen species, and the inheritance mechanism of growth enhancement effect is concluded to be owing to the epigenetics.

    In this experiment, two types of plasmas those are low-pressure plasma and atmospheric discharge plasma are used for irradiation seed and leaf, respectively. Low pressure oxygen plasma for irradiation seeds is generated by the radio frequency power with 13.56 MHz and several ten W. Atmospheric oxygen discharge plasma is used for irradiation leaf with the electrical power of 10 kHz and around 5 kV. Seed and leaf of Arabidopsis thaliana wild type, Columbia-01 is used for sample. After irradiation seed with low pressure plasma, germination rate and grown leaf area is measured as index of the growth enhancement effect.

    Recently, discharge plasmas including ions, electrons and electrically neutral particles with high reactivity have attracted attentions of some fields of applications. Significant features of low-pressure plasmas are dry process, no harmful residue, higher reactivity than chemicals, so we can expect new results in medicine, and also in agriculture and engineering. The fields of application of plasma sterilization methods are now expanding from medical equipment to agricultural products and spacecraft. In this presentation, the characteristics of the oxygen plasma sterilization method is introduced.

    Cherry is treated by active oxygen species those are generated by surface discharge to keep its freshness for several weeks. Sterilization effect depends on the ozone treatment time when the ozone concentration is fixed at high concentration. IR spectra of fruits surface indicate that the reduction reaction is occurred in fruits surface by the plasma treatment. In case of high concentration of ozone and treatment time was 10 minute, almost all the mold of fruit surface was sterilized.

    The purpose of this study is to examine the increase in the number of cells and the enhancement of the phagocytosis ability of macrophages when irradiated with oxygen plasma and nitrogen plasma to macrophages, which is a type of immune cells. The plasma generated by the surface discharge was irradiated to the cells through a silicon tube. The number of cells was measured using a cell counting reagent, and phagocytosis was measured using fluorescent beads. The phagocytosis ability was enhanced up to 2.5 times by oxygen plasma irradiation and up to 4 times by nitrogen plasma irradiation.

    Differentiation of osteoclast precursor cells is attempted to be regulated by the plasma irradiation to the cells without damages on the cells. The RANKL that is the differentiation inducer in the culture medium is modified by the plasma irradiation, and lose the function. Oxygen plasma irradiation to the cells without the culture medium at lower discharge voltage leads to the increase of the cell number. The kinase ASK1 that catalyze MAPK cascade for cell growth would be activated.

    Selective inactivation of the oral cancer cells is observed when the atmospheric oxygen plasma irradiates the oral cancer cells (HSC3) and normal cells (HaCaT) after removing culture medium. When the cells are irradiated with oxygen plasma, the intracellular active oxygen in cancer cells increases comparing with that in normal cells. Activity of caspase-3/7 in oral cancer cells, which is a kind of major biological reactions of the apoptosis, increases after oxygen plasma irradiation.

    Growth promotion characteristics of plants seeds are investigated by an atmospheric discharge irradiation onto plasma seeds. The torch-type barrier discharge plasma is blown outside with the gas flow in ceramics tube to radish sprouts seeds locate below the torch edge. The growth promotion of sprouts was observed in the total length of stem and root during its cultivation. The total length increases approximately 1.5 - 2 times at the cultivation time of 24 h. The growth promotion effect is found to be maintained for 40h, after sowing seeds. The mechanism of the growth promotion would be the redox reaction inside plant cells induced by oxygen radicals.

    Growth stimulation characteristics of plants seeds are investigated by an atmospheric discharge irradiation into plasma seeds. Atmospheric pressure plasma torch is consisted of alumina ceramics tube and the steel mesh electrodes wind inside and outside of the tube. When AC high voltage (8kHz) is applied to the electrode gap, the barrier discharge plasma is produced inside the alumina ceramics tube. The barrier discharge plasma is blown outside with the gas flow in ceramics tube. Radish sprouts seeds locate at 1 cm from the torch edge. The growth stimulation was observed in the length of a stem and a root after the plasma irradiation. The stem length increases approximately 2.8 times at the cultivation time of 24h. And the growth stimulation effect is found to be maintained for 40h, after sowing seeds. The mechanism of the growth stimulation would be the redox reaction inside plant cells induced by oxygen radicals.

    プラズマ照射を用いた細胞の活性制御を目的として,大気圧プラズマ照射によるパン酵母の増殖促進を試みた.実験にはバリア放電を用い,照射時間を変化させ直接粉末酵母に距離1mmで照射を行った.照射後の酵母は生育培地に懸濁し,マルチウェルプレート上に分注し振盪培養を行った.増殖曲線は波長660nmの吸光度の時間変化を測定し求めた.プラズマ照射を行った酵母は未照射の酵母と比較し,増殖曲線中における遅滞期の期間が短くなる傾向を示した.この結果は酵母の細胞周期が照射により加速されている可能性を示唆している.

    農産物へのプラズマ照射により①農産物表面の防菌防黴と②農産物の抗酸化活性向上を同時に実現することを目的とする.また,プラズマ照射による抗酸化活性向上のメカニズムの解明を遺伝子解析により試みる.なお,研究対象として当初設定した柑橘類に関しては,強い酸性の果汁により本研究を遂行する上で重要となる試薬類の多くが適用不可応であることから,本研究では実験対象果実としてブドウを選択した.ブドウで得られた防菌防黴と抗酸化活性の結果は,プラズマ処理を柑橘類に対して適用する場合にも十分な指標となると考えられる.

    アレルギー等の免疫疾患は,生体内の数種類の免疫細胞において活性のバランスが崩れることで生じる.従前の対症療法だけでなく,各免疫細胞の活性を通常のレベルに戻し生体の免疫機能を正常化させる方法を見出すことは喫緊の課題である.本研究では,プラズマ照射が免疫細胞の分化を通して免疫活性を制御するメカニズムの解明を行う.特に,T細胞に酸素プラズマを照射した場合に(i)免疫細胞の分化に対する酸素プラズマの作用機序,および(ii)酸素プラズマ照射から細胞分化を経て免疫活性制御に至る反応経路を明らかにすることを目的とする.加えて生体において崩れた免疫バランスがプラズマ照射で正常化することを明らかにする.

    By irradiating CD4+ naive T cells (EL4) with atmospheric pressure oxygen plasma under specific conditions, we achieved enhancement of growth and differentiation of T cells. Gene expression analysis suggests that the cell growth is mediated by activation of ERK and NFkB pathways in cells induced by active oxygen species. From the measurement of cytokines released by T cells, oxygen plasma-irradiated T cells did not produce IFN-γ and the amount of IL-4 released was significantly increased, indicating that T cells differentiated into helper Th2 cells. At this time, there is possibility that the autocrine phenomenon, in which Th2 cells are further activated by IL-4 produced by Th2 cells, occurred. It is concluded that the oxygen plasma is effective in activating T cells.
