    Benzalkonium chloride (BAC), a commonly used quaternary ammonium compound in various products like antiseptics, cosmetics, and disinfectants, has raised concerns due to its potential to contaminate aquatic environments and subsequently affect the reproductive performance of the organisms within those ecosystems. The article underscores a critical concern regarding the impact of BAC on aquatic ecosystems, particularly its effect on fish reproductive quality, using medaka (Oryzias latipes) as a model organism. Firstly, while measuring lethal dose of BAC in adult medaka, we observed a dose dependent mortality in BAC treated fish (100 and 200 ppm: 100%; 60 ppm: 51.7%; 30 ppm or less: no mortality at 24 h post treatment (hpt)) and calculated the LD50 at 96 hpt as 39.291 ppm (95% confidence interval: 28.817–53.570 ppm). Further, we assessed the molecular, cellular and histological changes through long-term exposure. Enlarged sperm pockets and reduced spermatocyte were seen in BAC exposed testis while no significant structural changes were observed in the ovaries. Following BAC exposure, drastic alterations in the gene expression and cellular localization related to sex, estrogen signaling, and autophagy were also noted from gonads and liver. Subsequently, using a short-term exposure analysis, we confirmed the sex and time responsive transcriptional kinetics and found that BAC sequentially affected the gonadal somatic cells followed by germ cell differentiation. Finally, using reproductively competent male and female medaka, we conducted progeny production and performance analysis and depicted a drastic reduction in fecundity, and fertilization and hatching rate, indicating adverse effects of BAC on reproductive success. Cumulatively, these findings emphasize the consequences of widespread use of BAC on reproductive security of aquatic animals and highlights the need for further research to comprehend the potential harm posed by such compounds to aquatic animal health and ecosystem integrity.

    Sexual development and reproduction are largely linked to epigenetic changes in many fish species. However, understanding of epigenetic regulation in scombrid species, such as tunas and mackerels, is limited. This study investigates DNA methylation’s impact on cyp19a expression, crucial for estrogen synthesis, focusing on chub mackerel. Given the commercial significance of scombrids and susceptibility of marine fish to environmental changes, elucidating epigenetic mechanisms, particularly in the context of global warming, becomes imperative. We aimed to generalize observations from chub mackerel to other scombrids. Additionally, we studied DNA methylation patterns across fish with different sexual systems to understand aromatase regulation’s phenotypic plasticity. Our in silico analysis revealed highly conserved promoter sequences within scombrids, sharing TFBS like Foxl2, FOS::JUN, ESRR, and Sox3, while CpG content varies. This indicates a conserved regulatory network controlling gene expression. We found sexual dimorphism in DNA methylation, with males hypermethylated and aromatase expression downregulated. Despite similar dnmt1 expression, tet1, tet2, and tet3 were higher expressed in females, suggesting that the observed DNA methylation patterns are maintained through active demethylation rather than differential methylation. Gonochoristic Japanese anchovy and protogynous bamboo leaf wrasse displayed similar trends, but species-specific methylation patterns highlight DNA methylation’s complex role in gonadal changes. In vitro assays confirmed methylation’s regulatory role and identified an SF-1 binding site relevant for promoter activation in chub mackerel. Another studied SF-1 site, present in both chub mackerel and bamboo leaf wrasse, showed regulatory effects, indicating potential similar regulatory mechanisms for cyp19a expression. Overall, our findings suggest that while global methylation affects cyp19a transcription, the variation in CpG density and location could be introducing nuances in its epigenetic regulation. This study contributes to our understanding of the cyp19a regulation in fish gonad maturation.

    Genome editing is a technology that can remarkably accelerate crop and animal breeding via artificial induction of desired traits with high accuracy. This study aimed to develop a chub mackerel variety with reduced aggression using an experimental system that enables efficient egg collection and genome editing. Sexual maturation and control of spawning season and time were technologically facilitated by controlling the photoperiod and water temperature of the rearing tank. In addition, appropriate low-temperature treatment conditions for delaying cleavage, shape of the glass capillary, and injection site were examined in detail in order to develop an efficient and robust microinjection system for the study. An arginine vasotocin receptor V1a2 (V1a2) knockout (KO) strain of chub mackerel was developed in order to reduce the frequency of cannibalistic behavior at the fry stage. Video data analysis using bioimage informatics quantified the frequency of aggressive behavior, indicating a significant 46% reduction (P = 0.0229) in the frequency of cannibalistic behavior than in wild type. Furthermore, in the V1a2 KO strain, the frequency of collisions with the wall and oxygen consumption also decreased. Overall, the manageable and calm phenotype reported here can potentially contribute to the development of a stable and sustainable marine product.

    For seasonal breeders, photoperiodic changes are important signals that mark the start of the breeding season. Thyroid-stimulating hormone (TSH) is a glycoprotein hormone that not only promotes the secretion of thyroid hormone but also plays a key role in regulating seasonal reproduction in birds and mammals. However, whether TSH activation has been implicated as a seasonal indicator in fish breeding has not been fully investigated. In this study, we isolated tshb as a starting point to elucidate the effect of photoperiodic changes on the activation of the reproductive axis of chub mackerel. The isolated tshb was classified as tshba, which is widely conserved in vertebrates. The quantitative PCR results showed that tshb was strongly expressed in the pituitary. When female and male chub mackerel with immature gonads were reared for six weeks under different photoperiodic conditions, the gonads developed substantially in the long-day (LD) reared fish compared to those in the short-day reared fish. Real-time PCR results showed that the expression level of tshb in the pituitary gland was significantly elevated in the LD group. Although there was no difference in the gonadotropin-releasing hormone 1 gene expression level in the preoptic area of the brain, follicle-stimulating hormone and luteinizing hormone gene expression levels in the pituitary were also significantly elevated in the LD group. In conclusion, TSH is a potential mediator of seasonal information in the reproductive endocrine axis and may induce gonadal development during the breeding season of chub mackerel.

    The gonadal sexual fate of vertebrates is either defined by genetics or environment, or a combination of both factors. Interestingly, in sequential hermaphroditism, the animal can undergo natural sex changes from female-to-male, male-to-female, and bidirectional way throughout their lives. This change exhibits the process which shifts between oogenesis and spermatogenesis and is regarded as an ideal instance of sexual plasticity. To develop the experimental model for studying the sexual plasticity of protogynous fish, the social conditions that induce sex changes were defined in wrasse, Pseudolabrus sieboldi. When six females were kept together in a tank, the largest female became a male, whereas a similar conversion did not occur when only two females were present in a tank. A semi-gonadectomy analysis developed in the present study verified the direct relationship between gonadal sex and body coloration. In P. sieboldi, the sex change is controlled by the relative body size of an individual within a group, rather than by absolute body size. When six females were kept in smaller sized tanks, delayed sex change or unchanged individuals was observed. Overall, more than 90% of the largest females demonstrated sex change after being housed with five smaller females in different sizes of tanks ranging from 80 to 500 L. Furthermore, the experiment using a transparent barrier suggested that visual stimuli are one of the major cues to initiate sex change. Our findings on the laboratory conditions leading to the initiation of sex change in wrasse suggest the usefulness of this species as a model organism for comparative studies in molecular, cellular, and physiological mechanisms of sexual plasticity, as well as on social and reproductive behaviors.

    Germ cells are pivotal for gonadal sexuality maintenance and reproduction. Sex lethal (sxl), the somatic sex determining gene of Drosophila, is the known regulator and initiator of germ cell femininity in invertebrates. However, the role of the Sxl homologue has rarely been investigated in vertebrates. So, we used medaka to clarify the role of sxl in vertebrate gonadogenesis and sexuality and identified two Sxl homologues, i.e., Sxl1a and Sxl1b. We found that sxl1a specifically expresses in the primordial germ cells (PGC), ovary, (early gonia and oocytes), while sxl1b distributions are ubiquitous. An mRNA overexpression of sxl1a accelerated germ cell numbers in 10 DAH XY fish, and sxl1a knockdown (KD), on the other hand, induced PGC mis-migration, aberrant PGC structuring and ultimately caused significant germ cell reduction in XX fish. Using an in vitro promoter analysis and in vivo steroid treatment, we found a strong link between sxl1a and estrogenic germ cell-population maintenance. Further, using sxl1a-KD and erβ2-knockout fish, we determined that sxl1 acts through erβ2 and controls PGC sexuality. Cumulatively, our study highlights the novel role of sxl1a in germ cell maintenance and sexual identity assignment and thus might become a steppingstone to understanding the commonalities of animal sexual development.

    The gonadotropin-releasing hormone (GnRH) is an important gene involved in fish reproduction and its action is mediated by GnRH receptor (GnRHR). Numerous GnRH-GnRHR studies have already been published in many teleost orders but information on reproduction-related genes for Clupeiformes is limited. To acquire a better understanding on the reproduction of Clupeiform fish species, GnRH isoforms and GnRHR paralogues of Japanese sardine, Sardinops sagax melanostictus (Temminck & Schlegel, 1846) were sequenced. Three GnRH isoforms (named as jsGnRH1, jsGnRH2, and jsGnRH3) and two GnRHR paralogues (named as jsGnRHR1 and jsGnRHR2) were isolated in the brain and pituitary of Japanese sardine. Based on phylogenetic analysis, jsGnRH1, jsGnRH2, and jsGnRH3 grouped into GnRH1, GnRH2, and GnRH3 forms, respectively, while jsGnRHR1 and jsGnRHR2 clustered into Type 2b and Type 1c receptors, correspondingly. The reporter gene assay showed high binding affinity of jsGnRHR1 to all three GnRH synthetic decapeptides, whereas jsGnRHR2 responded best to jsGnRH2 only. Clustering of jsGnRH1and jsGnRHR1 to other teleost GnRH1 and GnRHR2b, respectively, and activating the calcineurin/NFAT signalling pathway, suggests that these two genes are likely involved in fish reproduction.

    In vertebrates, estrogen receptors are essential for estrogen-associated early gonadal sex development. Our previous studies revealed sexual dimorphic expression of estrogen receptor β2 (ERβ2) during embryogenesis of medaka, and here we investigated the functional importance of ERβ2 in female gonad development and maintenance using a transgenerational ERβ2-knockdown (ERβ2-KD) line and ERβ2-null mutants. We found that ERβ2 reduction favored male-biased gene transcription, suppressed female-responsive gene expression, and affected SDF1a and CXCR4b co-assisted chemotactic primordial germ cell (PGC) migration. Co-overexpression of SDF1a and CXXR4b restored the ERβ2-KD/KO associated PGC mismigration. Further analysis confirmed that curtailment of ERβ2 increased intracellular Ca2+ concentration, disrupted intra- and extracellular calcium homeostasis, and instigated autophagic germ cell degradation and germ cell loss, which in some cases ultimately affected the XX female sexual development. This study is expected improve our understanding of germ cell maintenance and sex spectrum, and hence open new avenues for reproductive disorder management. In this article, Chakraborty et al. find that estrogen receptor β2 (ERβ2) plays multifaceted crucial roles in medaka gonadal development and sexuality. ERβ2 predominantly expresses in germ cells, influences the SDF1/CXCR4 chemotaxis, PGC migration, and death, and regulates germ cell proliferation and various sex-biased gene transcriptions that are essential for gonadal sex assignment and maintenance.

    The red seabream Pagrus major is a useful experimental fish for studying the endocrine control of oogenesis in teleosts. This study investigated the steroidogenic pathway for estradiol-17β (E2) biosynthesis in the ovarian follicles of red seabream. Intact follicles were isolated during vitellogenesis and incubated in vitro with different radiolabeled steroid precursors. When 17-hydroxyprogesterone (17-P), dehydroepiandrosterone (DHEA), or androstenedione (AD) were used as precursors, both testosterone (T) and estrone (E1) were synthesized by follicles, leading to estradiol-17β (E2) production. Serum steroid levels measured by enzyme-linked immunosorbent assay showed that T, E1, and E2 were present in the circulation at levels ranging from 1 ng/mL to 2 ng/mL throughout the day during the spawning season. In vitro conversion of E1 into E2, however, was 15.8-fold greater than T conversion into E2, suggesting that E2 is synthesized mainly via E1 rather than T. The results showed that E2 was synthesized from pregnenolone via 17-hydroxypregnenolone, DHEA, AD, and E1. Thus, the study demonstrated the complete steroidogenic E2 synthesis pathway in the ovarian follicles of red seabream, and revealed that E1 is the major precursor of E2.

    Intact follicles of the bambooleaf wrasse were incubated with different radioactively labeled steroid precursors during final oocyte maturation (FOM), and the maturation-inducing hormone (MIH) of this species was examined to compare the activities of the steroid metabolites produced on germinal vesicle breakdown (GVBD) in vitro. Of the metabolites obtained by thin layer chromatography, two steroids, 17,20β-dihydroxy-4-pregnen-3-one (17,20β-P) and 17,20β,21-trihydroxy-4-pregnen-3-one (20β-S), both had the greatest effect on GVBD in vitro. During FOM, a shift in steroidogenic enzymes, a decrease in C17,20-lyase activity, and an increase in 20β-HSD activity were found. In addition to this shift, continuous high 21-hydroxylase activity throughout all the oocyte developmental stages was observed, and, in all likelihood, this 21-hydroxylase activity enables ovarian follicles to synthesize enough 17,21-P. Consequently, activated 20β-hydroxysteroid dehydrogenase converted 17-P and 17,21-P to 17,20β-P and 20β-S, respectively, during FOM. Thus, the present study not only provides evidence on the physiological role of 17,20β-P and 20β-S as MIH in the bambooleaf wrasse, but also details the enzymatic kinetics of 17,20β-P and 20β-S biosynthesis.

    In our previous studies, we tentatively identified 17,20β-dihydroxy-4-pregnen-3-one (17,20β-P) as a maturation-inducing hormone (MIH) in yellowtail (Seriola quinqueradiata) through in vivo and in vitro experiments. In this study, we investigated the binding sites for radioactive 17,20β-P and characterized the receptor binding to the ovarian plasma membrane in yellowtail undergoing first stage of maturation (FSM). Equilibrium binding sites for 17,20β-P have been detected within 1 h incubation and the binding dissociated completely within 50 min at 4°C and was pH dependent (optimum pH 7.8). Scatchard analyses of specifically bound 17,20β-P showed the evidence of a single class of high affinity binding sites (KD = 22.9 nM), with limited capacity (Bmax = 2.1 pmol/g tissue) to the ovarian membrane of yellowtail undergoing FSM. Competition results revealed that ovarian membrane receptor was highly specific for 17,20β-P. There was no other steroid competed strongly with the binding sites of [3H]17,20β-P, except 17,20β-P itself. On the other hand, 17,20β-P did not bind to the membrane prepared from maturationally incompetent (MI) and ovulation (OV) stages of oocytes. As the time proceeded after the stimulation of HCG, binding activity increased significantly (0.389 ± 0.036 pmol/g tissue) in the ovarian membrane of maturationally competent (MC) oocytes by 12 h postinjection. The binding activity was further significant (0.868 ± 0.032 pmol/g tissue) at FSM by 24 h postinjection and reached its peak (0.920 ± 0.115 pmol/g tissue) temporarily at second stage of maturation (SSM) by 36 h postinjection and then sharply declined to the prestimulation levels during OV stage by 48 h postinjection. In addition to our previous findings, the present results indicate that 17,20β-P is the MIH in yellowtail.

    The bambooleaf wrasse, Pseudolabrus sieboldi, is a diandric protogynous fish with a diurnal rhythm of ovarian development, and females spawn daily during the spawning season. This study investigated the steroidogenic pathway for estradiol-17β (E2) biosynthesis in vitellogenic ovarian follicles of the bambooleaf wrasse. We incubated follicles in vitro with radioactively labeled steroid precursors, and measured serum steroid levels using microtiter plate enzyme-linked immunosorbent assays (ELISAs). ELISAs for estrone (E1) and testosterone (T) were developed. The experiments showed that E2 was synthesized from pregnenolone via 17-hydroxypregnenolone, dehydroepiandrosterone, androstenedione, and E1. T was not produced from any radiolabeled precursors, and exogenous T was not converted to E2. During the spawning season, serum levels of E2 and E1 showed similar patterns, with a diurnal rhythm of high levels at 03:00 hr associated with active ovarian vitellogenic follicles. In contrast, serum T levels were constant and relatively low compared to levels of E2 and E1. These results indicate that E2 is synthesized via E1, rather than T, in the ovarian follicles, and suggest that T detected in the blood is likely derived from extra-follicular tissues.

    The female bambooleaf wrasse, Pseudolabrus japonicus, spawns daily during the spawning season, and exhibits a diurnal rhythm of ovarian development. In the present study, we have investigated: (1) circulating levels of 17α,20β-dihydroxy-4-pregnen-3-one (17,20β-P) and 17α,20β,21-trihydroxy-4-pregnen-3-one (20β-S) in females sampled at different times of the day during spawning season in captivity, and (2) in vitro production of 17,20β-P and 20β-S by follicle-enclosed oocytes at seven different developmental stages. In addition, we developed a microtiter plate enzyme-linked immunosorbent assay (ELISA) for 17,20β-P. Serum levels of 17,20β-P and 20β-S showed similar diurnal changes; substantial increases in these levels occurred around the time of germinal vesicle breakdown (GVBD). In vitro experiments showed that massive production of 17,20β-P and 20β-S occurred in follicles collected just before or during GVBD. Further, acute decreases in 17,20β-P and 20β-S production were found in the ovarian follicles just prior to ovulation, suggesting inactivation of the maturation-inducing hormone (MIH). These results, taken together with our previous data on the occurrence of GVBD in vitro, suggest a role for both 17,20β-P and 20β-S as MIHs in the bambooleaf wrasse.

    Gonochorism and hermaphroditism occur among teleost fishes and as in other vertebrates, sex is determined by monogenic, polygenic, and environmental means. There are at least 10 different master sex-determining genes discovered in teleosts. Regardless of the sexual system employed, once sex is determined, gonadal sex differentiation into ovaries and testes is relatively conserved. There is an antagonistic relationship between the testicular and ovarian pathways, which remains active throughout the lifetime to maintain the differentiated sexes. Estrogens play a pivotal role not only in initiating ovarian differentiation but also in maintaining the female phenotype in both gonochoristic and hermaphroditic fishes. This chapter reviews the current understanding of the diverse modes of sex determination, the relatively conserved process of gonadal sex differentiation and sexual plasticity in adulthood in teleosts.

    哺乳類では,遺伝的に性が決まり,その性に従って性分化をした後,同じ性のまま一生を過ごす。一方,魚類のなかには,遺伝的に性を決定するもののみならず,環境の影響を受けて性が決まるものや,一生の間に性を転換させるものなど,様々な性決定/性分化/性転換の様式がみられる。なかでも生息環境における社会性によって性をかえる性転換魚は,社会環境の変化を起点として脳や生殖腺を含む個体全体の性を転換させることから,脊椎動物における個体,器官(脳や生殖腺),および細胞の性的可塑性を理解する上での格好の研究材料となる。これまでに筆者らは性転換魚の実験系を独自に構築し,それを材料とした個体,組織,細胞についての解析を進めてきた。その結果,性転換魚の生殖腺において,生殖幹細胞と未分化体細胞の存在および機能の一端が見えてきた。本稿ではこうした生殖腺の性的可塑性ついての研究内容とシングルセル解析の取り組みについて紹介する。(著者抄録)

    魚類の性を自在に統御するには、それぞれの種が有する性的可塑性の特徴を細胞レベルで理解する必要がある。本研究では、性転換モデル系に空間的遺伝子発現解析を初めて導入し、生殖腺の性転換の起点になる未分化体細胞の位置情報をもとに生殖幹細胞との相互作用を分子レベルで明らかにする。同時に、各種シングルセル解析と培養系を駆使して研究を進め、“卵形成と精子形成の切り換え”の制御機構を解明する。さらに、雌雄異体魚との比較を行い、将来、全ての魚類において個体、生殖腺、および細胞の性を自在に統御するために必要な“性的可塑性の共通原理”に関する情報を得る。

    現在使われているゲノム編集技術であるTALENおよびCRISPR/Cas9に関する基本特許は外国が持っており、育種を通した日本の養殖産業の強化を視野にいれたとき、国産のゲノム編集技術の開発は極めて重要である。新しい核酸認識モジュールであるPPRタンパク質は、国産の新たなゲノム編集ツールとして期待されている。本申請課題では、完全養殖系のマサバを用いて、PPRを利用した新品種の作出を行う。さらに、食品としての安全性をオフターゲット解析およびメタボローム解析により評価する。
