Summary of Research Activities 1. Studies on Photosynthesis and Biomass Production Mechanisms in Crops and Their Modification Research topics include the physiological role of silicon in dry matter production and photosynthesis in rice, elucidation of photosynthetic conversion mechanisms in response to environmental fluctuations in species such as Eleocharis and the common ice plant (Mesembryanthemum crystallinum), genetic modification of rice by introducing C4 photosynthesis-related genes, and CAM induction in C3 plants for enhanced photosynthetic plasticity. 2. Development of Stable Agricultural Production Technologies Adapted to Environmental Changes This research focuses on elucidating the salt tolerance mechanisms of halophytes, utilizing stress-tolerant resource plants as genetic resources and crops, and phytoremediation using plants under various environmental conditions. 3. Exploration of Novel Physiological Functions in Plants and Their Applications in Agricultural Technology Investigations include mechanisms underlying salt-promoted growth in halophytes, NaCl-induced enhancement of cell division and ATP production, elucidation of salt accumulation in epidermal cells, and the identification and biosynthetic pathways of functional metabolites produced by valuable resource plants. 4. Research on Plant-Microbe Interactions for Sustainable Agricultural Production Studies focus on utilizing symbiotic microbial communities to improve plant growth, stress tolerance, and nutrient uptake in diverse cropping systems. 5. Research on Plant-Microbe-Based Electricity Generation Development and characterization of plant-microbe bioelectrochemical systems for renewable energy applications. --- Summary of Educational Activities 1. Lectures and research supervision on plant production physiology for master’s and doctoral students in the Agricultural Bioscience Course, Graduate School of Bioresource and Bioenvironmental Sciences. 2.     This study presents the reproductive growth characteristics of a halophyte Mesembryanthemum crystallinum L. when grown in high-salinity soil stress conditions. The experiments were arranged and carried out continuously from 2020 to 2023, the first two years at Kyushu University in Japan and the last year at Duy Tan University in Vietnam. During the time in Japan, the effects of salinity stress on the reproductive growth stages and the morphological characteristics of M. crystallinum cultured in the soil treated with saline solutions containing 0 mM, 100 mM, and 400 mM NaCl were determined. Then, the seeds harvested from these plants were reproduced to develop them as the high salt-tolerance plants in Vietnam. The results show that 400 mM NaCl-treated plants maintained their growth and developed well during the vegetative growth stage. These plants had shorter flower blooming times than the 100 mM NaCl-treated and control ones (0 mM NaCl). The seeds harvested from NaCl-treated and control plants in Japan had regenerated in vitro plants well in the Vietnamese culture rooms. Interestingly, the 400 mM NaCl-treated M. crystallinum grew healthily in the soil, and their leaves, flowers, and seedpods tended to accumulate more betacyanin and epidermal bladder cells (EBCs) than other plants. These characteristics may be desirable for rehabilitating saline soil and developing high-salinity tolerance M. crystallinum for cultivation and growth in Vietnam.

    Background: The common ice plant (Mesembryanthemum crystallinum L.) is an annual herb belonging to the genus Mesembryanthemum of the family Aizoaceae, native to Southern Africa. Methods: We performed shotgun genome paired-end sequencing using the Illumina platform to determine the genome sequence of the ice plants. We assembled the whole genome sequences using the genome assembler “ALGA” and “Redundans”, then released them as available genomic information. Finally, we mainly estimated the potential genomic function by the homology search method. Results: A draft genome was generated with a total length of 286 Mb corresponding to 79.2% of the estimated genome size (361 Mb), consisting of 49,782 contigs. It encompassed 93.49% of the genes of terrestrial higher plants, 99.5% of the ice plant transcriptome, and 100% of known DNA sequences. In addition, 110.9 Mb (38.8%) of repetitive sequences and untranslated regions, 971 tRNA, and 100 miRNA loci were identified, and their effects on stress tolerance and photosynthesis were investigated. Molecular phylogenetic analysis based on ribosomal DNA among 26 kinds of plant species revealed genetic similarity between the ice plant and poplar, which have salt tolerance. Overall, 35,702 protein-coding regions were identified in the genome, of which 56.05% to 82.59% were annotated and submitted to domain searches and gene ontology (GO) analyses, which found that eighteen GO terms stood out among five plant species. These terms were related to biological defense, growth, reproduction, transcription, post-transcription, and intermembrane transportation, regarded as one of the fundamental results of using the utilized ice plant genome. Conclusions: The information that we characterized is useful for elucidation of the mechanism of growth promotion under salinity and reversible conversion of the photosynthetic type from C3 to Crassulacean Acid Metabolism (CAM).

    This study aims to examine the characteristics of vacuole ATPase (V–ATPase) activities and protein expression in vacuole fractions isolated from the common ice plant (Mesembryanthemum crystallinum L.) grown under control and salt–stressed conditions, using various centrifugation conditions of sucrose density gradient. The protein samples were analyzed using two–dimensional electrophoresis (2DE) gels. The results reveal that the vacuolar fractions collected at 20%, 30%, and 40% (w/w) sucrose density in the 0 mM, 100 mM, and 400 mM NaCl–stressed plants, respectively, exhibited the highest V–ATPase activities. Using the differential display with 2DE gels, the proteins in the vacuole fractions were analyzed, and the abundant proteins on each gel were identified based on their isoelectric points and molecular weights. The Arabidopsis database and the ExPASy–TagIdent tool were used to estimate the proteins. The results show 165, 194, and 199 abundant proteins with 41, 51, and 53 different proteins relating to the vacuole fractions among control samples (0 mM NaCl) and salt–stressed samples at 100 mM NaCl and 400 mM NaCl, respectively. Although the proteins on each 2DE gel varied among the samples, they were generally focused on ten physiological function groups, including pump proteins, transporter proteins, metal channel proteins, peptide transport proteins, channel proteins, stress response proteins, protease proteins, autophagic proteins, stress response proteins, and unknown proteins.

    The common ice plant (Mesembryanthemum crystallinum L.), a halophytic vegetable plant, shows the maximum growth in the salinity environment where almost all crops die. The NaCl–promoted growth enhancement called halophilism is an essential trait of increased salt tolerance. To elucidate the factors associated with halophilism, we investigated the effects of NaCl on respiration and genes of cell–division–related factors in suspension–cultured cells. Respiration rate increased with the increase of concentration of NaCl, being maximal at 100 mM NaCl. Metabolite analysis showed that contents of metabolites in glycolysis such as glucose, fructose 6–phosphate, and lactic acid were reduced in the NaCl treated cells. The contents of TCA cycle metabolites such as pyruvate (or oxaloacetic acid), citric acid (or isocitric acid), succinic acid, fumaric acid, and malic acid were reduced in NaCl–treated cells at 21 d after treatment, indicating that the metabolic flux of the TCA cycle is activated to increase respiration and production of NADH and FADH_2, which are electron donors for ATP synthesis. Expression analysis showed that the expression of eight genes for complex of ETC (electron transport chain complex), ATP synthase, and TCA cycle enzymes was induced in the salt–treated cells within 6–48 h after the onset of treatment. Moreover, we found that among five cell cycle–related genes that we newly annotated, the expression of a gene associated with the progression of the G1 (McCycD3;1), a gene for B2–type cyclin–dependent kinase (McCDKB2;1), and its cyclin partner forming an active kinase complex in G2/M phase (McCycD4;1) were stimulated by NaCl. We suggested that these factors contributed to the enhanced cell growth in the common ice plant halophilism.

    Many bacterial and fungal pathogens are known to affect lotus, which limits both flower quality and yield production. Treatments such as pesticides can reduce pathogens but can result in resistance. This study was conducted to identify the most common fungal, and bacterial pathogens associated with leaf spot diseases of white lotus (Nelumbo nucifera). Molecular–based identification using ITS and 16 s ribosomal DNA sequences revealed that the isolates belonged to Aspergillus aculeatus, Aspergillus fumigatus, and Klebsiella pneumoniae. We evaluated the effect of silver nanoparticles against pathogens isolated from infected white lotus leaves. According to the findings, AgNPs have antifungal activities against these plant diseases at different concentrations of 0.1,1, 10, 20, and 30 mg/L. Treatment of 30 mg/L silver nanoparticles on PDA showed the highest inhibition rate of plant pathogenic fungi Aspergillus sp., and bacteria Klebsiella pneumoniae. Based on this result, it is possible to suggest that silver nanoparticles synthesized could be an efficient, safe, cost–effective, and affordable alternative to control disease in white lotus.

    Halophytes are salt-tolerant plant that grows naturally in saline areas where almost all conventional crops die due to NaCl toxicity. The common ice plant, Mesembryanthemum crystallinum L., an annual halophyte native to South Africa, tolerates high salinity levels and accumulates NaCl in a shoot at a high level. To check the availability of the ice plant for desalinization of soils, we cultured the ice plant in soils collected from 16 sites located along coastal regions in the prefectures of Miyagi and Iwate, where were attacked by the tsunami disaster in the wake of the 2011 earthquake off the Pacific coast of Tohoku on 11 March 2011. In the soils obtained from some tsunami affected areas, the growth was better than that in the non-contaminated soil. The factors associated with growth inhibition were suggested to be water ratio (an index of water content) and soil water permeability. The ice plant’s estimated biological yield ranged from 0.33 to 14.6 kg m<sup>−2</sup>, equivalent to 2.3 to 101.7 t ha<sup>−1</sup>. The sum of Na<sup>+</sup> and Cl<sup>−</sup> was about 9.5 g in the shoot (31.8% on a dry weight basis), and the estimated total amount of these ions removed from salinized soil was 2.38 t ha<sup>−1</sup>. These results indicated that the common ice plant could be used as a crop under salinity and a tool for ameliorating NaCl from salinized soils.

    This research aims to modify crop photosynthesis to confer extreme drought and salt tolerance. To introduce CAM into C3 plants, the researchers will edit the transcriptional regulatory sequences of CAM genes in C3 plants to place the associated genes under the control of clock genes. To identify the editing sites, they will identify cis-elements and transcription factors that control CAM gene expression and elucidate the factors required for CAM activation. They identified transcriptional regulators of CAM-related genes in C3 plants and identified sequences to be modified by genome editing. They also created vectors for prime editing and identified cis-factors and transcription factors involved in the C3-CAM transition in the common ice plant.

Educational Activities

  • As part of graduate education, I am responsible for teaching the Advanced Plant Production Physiology and supervising both doctoral and master’s thesis research for graduate students in the Laboratory of Plant Production Physiology, including those enrolled in the international course. In undergraduate education, I teach Introduction to Agriculture, Plant Production Physiology, Agroecology, and Plant Production and Physiology I, and also share responsibility for specialized courses targeted at lower-year students. Additionally, I provide supervision for undergraduate thesis research conducted by students in the Laboratory of Plant Production Physiology.
