Outline of research: Main research titles of our laboratory are microbial control of insect pests, characterization of entomopathogenic microorganisms, mechanism of microbial infection in insects, defense reaction of insects against pathogens, and studies of invertebrate cell culture. I concentrated especially on protozoan infection in insects and studies of lepidopteran insect cell culture as follows: 1) Studies on infection and development of entomopathogenic microsporidians as microbial control agents using insect cell cultures. 2) Development of in vitro germination methods for poor germinators of the microsporidians. 3) Ultrastructursal study of host-microsporidian parasite interactions. 4) Clonig of the microsporidians and chromosomal study of those clones. 5) Study of a protozoan parasite isolated from the West Indian sweet potato weevil. 6) Establishemnt of insect cell lines having a highly phagocytic ability. Outline of education: 1) Lectures and tutorials of Insct Pathology and Microbial Control for the graduate students. 2) English lectures of Microbiology, Insct Pathology and Microbial Control for the undergraduate and the graduate students in special course on international development research (partial responsibility). 3) Lecture of Insct Pathology and Microbial Control for the undergraduate students (partial responsibility).

    The West Indian sweet potato weevil, Euscepes postfasciatus (Fairmaire), native from West India is one of the most important insect pests of sweet potato in Japan. The sterile insect technique eradication program is in progress in Okinawa Prefecture, Japan. In 2004, however, the neogregarine Farinocystis sp. parasite was detected from mass-reared colonies of E. postfasciatus at the facility of Okinawa Prefectural Plant Protection Center and it caused decrease in their longevity and fecundity. This protozoa were not detected from wild West Indian sweet potato weevils except 1 individual, so the original host of the neogregarine is unknown. In this study, the Coleopteran insects were injected with the protozoan oocysts and reared 2 weeks. They were investigated the infected or uninfected with the neogregarine using PCR and tissue fixation. As the results, Scarabaeidae, Chrysomeloidae, and Tenebrionidae were not infected with Farinocystis sp.. On the other hand, about Curculionoidae, the Farinocystis sp. specific products were amplified by PCR and trophozoites-like protozoan cells were observed by the tissue fixation. The protozoan cells after trophozoites in their life cycle, however, were not observed. These results suggested that Farinocystis sp. has very narrow host range.

    Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) is a serious pest of crops and vegetables in several Asian areas. Metarhizium rileyi is one of the most promising entomopathogenic fungi for controlling S. litura because of its specificity to lepidopteran larvae; however, little is known about its virulence against last instar larvae and pupae of S. litura although these stages are efficiently controlled by introducing entomopathogenic fungi into the soil. This study aimed to characterize the virulence of M. rileyi strain against the two stages of S. litura by comparing it with 13 strains of entomopathogenic fungi with broad host ranges. In inter-stage virulence comparisons, most fungal strains showed considerably stronger virulence against larvae than against pupae, with M. rileyi Nr4 showing the largest difference. Inter-strain virulence comparisons based on final mortality and mycosis proportion as well as integrative virulence scores revealed that M. rileyi Nr4 was the most virulent against larvae, whereas it was the weakest against pupae, but caused malformation in emerged adults. These results suggest that M. rileyi Nr4 is the most promising for controlling the last instar larvae and is less effective against pupae than the generalist fungi.

    A Bacillus thuringiensis strain, which is an entomopathogenic microorganism, was used for effective control of Lycoriella ingenua larvae, which are an intractable pest occurring during Agaricus bisporus cultivation. The effects of B. thuringiensis subsp. kyushuensis CAB464 strain that shows high insecticidal effects against L. ingenua larvae at low concentrations, which were sublethal concentrations, on the survival rate of L. ingenua larvae were reviewed and according to the results, the feeding and growth rates of the larvae were hindered, and it could be seen that while B. thuringiensis subsp. kyushuensis CAB464 strain directly acts on A. bisporus cultivation plots, it also has an insecticidal mechanism to inhibit feeding thereby starving the larvae to death. Pot tests were conducted to see the applicability and control effect of B. thuringiensis subsp. kyushuensis CAB464 strain on A. bisporus cultivation plots, and according to the results, B. thuringiensis subsp. kyushuensis CAB464 strain showed a control value of 80.1%, which was at least about 50% higher than that of the untreated control plot and showed a survival rate at least about 38% lower than that of the untreated control plot while the untreated control plot showed gradual increases in the survival rate. The control effects of B. thuringiensis subsp. kyushuensis CAB464 strain were tested in an A. bisporus cultivation plot in which L. ingenua in all age periods existed in mixture, and based on the results, the control value after two times of treatment was higher than that after one time of treatment as it was 75.6% after the first treatment and 81.7% after the second treatment, and it could be seen that the survival rate gradually decreased. The foregoing indicates that B. thuringiensis subsp. kyushuensis CAB464 strain is applicable as a novel microbial insecticide that can control L. ingenua.

    微胞子虫類は、菌類の1グループとして位置づけられる偏性細胞内寄生性の単細胞真核微生物で、宿主の約4割が昆虫で占められる。野外鱗翅目昆虫からはさまざまな微胞子虫種が検出されているが、これら微胞子虫は、野外昆虫個体群密度を制御する要因として重要な役割を果たしていると考えられている。
    そこで本研究では、野外鱗翅目昆虫における微胞子虫感染と、異なる鱗翅目昆虫種間での交叉感染試験による微胞子虫の病原性・病原力、感染増殖および伝播を調査し、微胞子虫感染によって宿主の野外個体群密度がどのように制御されているのか、また、微胞子虫の交叉感染が鱗翅目昆虫の種多様性を脅かす可能性があるのか、という問いに迫る。
