Agr., Kyushu Univ., 69, 1, 1-10, 2024.02, In Japan, Dickeya dianthicola, D. chrysanthemi, Pectobacterium atrosepticum, P. parmentieri, and P. brasiliense are known as blackleg disease pathogens of potato. A transient outbreak of the blackleg disease occurred in Nagasaki Prefecture, and the pathogens were identified as P. parmentieri and P. brasiliense. Because repetitive extragenic palindromic PCR showed clonal relatedness among strains in each species, two Nagasaki strains (NK5 and NK14) were selected as the representative strains, and their whole genome sequences were determined. Average nucleotide identity analysis supported the identification in the previous study. Multilocus sequence analysis with seven housekeeping genes indicated the relationship between NK14 and other strains isolated in Japan, Russia, China, Switzerland, Poland, and Belgium. NK5 was related to the strains from various geographic locations including Syria, China, Canada, South Korea, Netherlands, Algeria, South Africa, Mexico, and Belarus. Subsequently, core genome multilocus sequence typing was performed with publicly available sequences, indicating that NK14 and NK5 were most closely related to the strains Poland/Russian and Netherlands/Belarus/Russia, respectively. This study concerns the first phylogenetic analyses based on the whole genome sequences of Japanese strains of P. parmentieri and P. brasiliense, and contributes to the molecular epidemiological analysis of the pathogen of potato blackleg.

Although sterol demethylation inhibitor (DMI) fungicides have controlled this disease effectively, outbreaks have been frequent in some areas of Japan since 2008. We, therefore, evaluated fungal populations from Fukuoka Prefecture for DMI fungicide sensitivity in vivo. Aeciospores on diseased pear leaves were collected in 2013 from four orchards in Fukuoka Prefecture and one control orchard in Ibaraki Prefecture. Juniperus chinensis var. kaizuka Hort. test plants were sprayed with difenoconazole and fenarimol at recommended rates, and the plants were inoculated with aeciospores. Difenoconazole and fenarimol effectively controlled the population of rust fungus from Ibaraki Prefecture. In contrast, fenarimol and difenoconazole were found to be ineffective against fungal populations from Fukuoka Prefecture. Subsequently, telia were collected from diseased juniper twigs in five areas in Fukuoka Prefecture and one control area in Ibaraki Prefecture in 2013 and 2014. Similarly, test plants of Japanese pear ‘Kousui’ were treated with difenoconazole or fenarimol and inoculated with basidiospores taken from those telia. Both difenoconazole and fenarimol were largely ineffective against the populations from Fukuoka Prefecture. In addition, field trials conducted in an experimental orchard in 2014 and 2015 showed that difenoconazole and fenarimol were not effective in controlling this disease. These results indicate the occurrence of Japanese pear rust resistant to DMI fungicides in Fukuoka Prefecture..

Here, we identified and characterised an insecticidal protein from Enterobacter. The insecticidal protein was purified from the strain and inoculated into B. mori larvae. Intracellular proteins were prepared, purified and separated by preparative native polyacrylamide gel electrophoresis (PAGE); one protein band had insecticidal activity. Sodium dodecyl sulfate-PAGE showed the presence of several bands, indicating that the insecticidal protein formed a complex. Peptide mass fingerprinting of a prominent 255.3-kDa band revealed 64 peptides that matched one protein with 33.0% sequence coverage. This protein was a homologue of the A component of the toxin complex (Tc), and the VRP1 domain was conserved; thus, the gene was named itcA (insecticidal toxin complex A). In the itcA downstream region, B and C component gene homologues were found, and these genes were located on an 86.2-kb contig sequence. Two repA genes and 27 genes related to conjugation transfer of plasmids were located on the contig, suggesting that the contig originated from a mobilisable plasmid. Therefore, these findings suggested that the strain may have acquired the Tc genes by horizontal transfer. This is the first description of Tc produced by the genus Enterobacter..

Myanmar Rs strains were characterized by traditional and molecular methods. Polymerase chain reaction (PCR) test using Rs-specific primer set amplified one specific band (281-bp) from template DNA of all strains. Pathogenicity tests on the four solanaceous plants differentiated the strains into six pathogenic groups. Biovar determination tests showed that biovar 3 strains predominated (63%) among all Rs strains. Biovar 4 strains (7%) were obtained from both tomato and chili strains, whereas biovar 2 (30%) strains were isolated only from potato. Multiplex-PCR analysis indicated that tomato, eggplant and chili strains belonged to phylotype I, whereas potato strains comprised phylotype I and phylotype II. Strains in phylotype I, which was suggested to have originated from Asia, were the most prevalent in all surveyed areas. Phylogenetic analysis based on the endoglucanase (egl) gene sequences revealed that Myanmar strains partitioned into two major clusters that corresponded to phylotype I and II. Strains in phylotype I were further divided into seven subclusters, each corresponding to a distinct sequevar (15, 17, 46, 47, 48, unknown 1 or unknown 2). All strains in phylotype II belonged to sequevar 1. This is the first comprehensive report of the presence of diverse Rs strains in Myanmar..

Here, we re-evaluated the position of bacterial strains (532 and 652) belonging to the genus Hafnia, isolated from flacherie-diseased silkworms in 1965. Phylogenetic analysis based on the 16S rRNA gene sequences of these strains suggests that they belong to genus Enterobacter. Using multilocus sequence analy- sis (MLSA), these strains were further classified to MLSA group A, which is a “core” group of Enterobacter con- taining E. cloacae (the type species of the genus). Although these strains were closely related to E. mori, E. tabaci, and E. asburiae, they also had other MLSA characteristics that distinguished them from these neighbor- ing bacterial species. These data were supported by further biochemical analysis. Thus, it appears that the 532 and 652 strains isolated almost half a century ago belong to genus Enterobacter, and their unique characteristics strongly suggest that they are a novel bacterial species..

In contrast, information pertaining to bacterial protein expression using this system is not sufficient. Therefore, recombinant nucleases from Serratia liquefaciens (rSlNucAs) were expressed in a Baculovirus-B. mori protein expression system. rSlNucAs containing the native signal peptide (rSlNucA-NSP) or silkworm 30-K signal peptide (rSlNucA-30K) at the NH2-terminus were constructed to enable secretion into the extracellular fraction. Both rSlNucA-30K and rSlNucA-NSP were successfully secreted into hemolymph of B. mori larvae. Affinity-purified rSlNucAs showed high nuclease activity. Optimum pH was 7.5 and half of maximum activity was maintained between pH 7.0 and 9.5. Optimum temperature was 35 °C. rSlNucAs showed sufficient activity in twofold-diluted radioimmunoprecipitation assay buffer and undiluted, mild lysis buffer. Genomic DNA of Escherichia coli was efficiently digested by rSlNucAs in the bacterial lysate. The results in this study suggest that rSlNucAs expressed by the Baculovirus-B. mori protein expression system will be a useful tool in molecular biology. Functional recombinant protein of bacteria was produced by Baculovirus-B. mori protein expression system. This system may be highly suitable for bacterial extracellular protein secreted via Sec pathway..
