The Hisayama study, Alzheimer's and Dementia: Diagnosis, Assessment and Disease Monitoring, 10.1016/j.dadm.2019.04.008, 11, 415-423, 2019.12, [URL], Introduction: We have conducted the pathological cohort study of autopsied cases of Hisayama residents to reveal a recent trend of dementia-related pathology. We noticed a trend of putaminal involvement of Alzheimer's disease (AD) with parkinsonism. Then, we investigated the accurate prevalence of neurological diseases with putaminal AD pathology in the general population. Methods: We examined a series of 291 autopsies in the Hisayama study and performed image analysis of immunohistochemistry for microtubule-associated protein tau (MAPT) and amyloid β. Results: Approximately 65.6% and 36.1% of cases showed putaminal MAPT and amyloid deposits, respectively. Diffuse deposits of them were mainly found in the AD cases. Putaminal MAPT was highly associated with AD-related pathological criteria. Four of 22 cases with severe putaminal MAPT deposition were documented as having developed parkinsonism. Discussion: Severe MAPT accumulation in the basal ganglia was closely related to the development of AD pathology and could occur most frequently in AD cases without comorbidities..

In this pilot study, we measured electroencephalography (EEG) and eye gaze data of learners and analyzed to estimate which area they had difficulty to learn. The developed system realized simultaneous measurements of physiological data and subjective evaluations during learning. Using this system, we observed specific EEG activity in difficult pages. Integrating of eye gaze and EEG measurements raised a possibility to determine where a learner felt “difficult” in a page of learning materials. From these results, we could suggest that the multimodal measurements of EEG and eye gaze would lead to effective improvement of learning materials. For future study, more data collection using various materials and learners with different backgrounds is necessary. This study could lead to establishing a method to improve e-learning materials based on learners' mental states..

A dual-tablet user interface with simplified visual layers and more intuitive operations was proposed aiming to reduce the physical and mental loads of elderly learners. An initial prototype with basic functions of viewing learning material was developed based on a cross-platform framework. Two preliminary user experiments participated by elderly volunteers were carried out for formative evaluations, in order to improve the usability of the interface design iteratively. The prototype was modified based on the participants’ comments and observation of their operations during the experiments. Additional findings of the elderly users’ preference and tendency were discussed for further development..

As the first steps towards to build new learning environments, we developed a system for multimodal learning analytics using eye-tracker and EEG measurement, and inclusive user interface design for elderly learners by dual-tablet system. Multimodal learning analytics system can be supportive to extract where and how learners with varied backgrounds feel difficulty in learning process. The eye-tracker can retrieve information where the learners paid attention. EEG signals will provide clues to estimate their mental states during gazes in learning. We developed simultaneous measurement system of these multimodal responses and are trying to integrate the information to explore learning problems. A dual-tablet user interface with simplified visual layers and more intuitive operations was designed aiming to reduce the physical and mental loads of elderly learners. A prototype was developed based on a cross-platform framework, which is being refined by iterative formative evaluations participated by elderlies, in order to improve the usability of the interface design. We propose a system architecture applying the multimodal learning analytics and the user-friendly design for elderly learners, which couples learning analytics “in the wild” environment and learning analytics in controlled lab environments..

Kazuhiro Samura, Yasushi Miyagi, Minako Kawaguchi, Fumiaki Yoshida, Tsuyoshi Okamoto, Masatou Kawashima, Predictive factors of antiparkinsonian drug reduction after subthalamic stimulation for parkinson’s disease, Neurologia medico-chirurgica, 10.2176/nmc.oa.2019-0040, 59, 9, 331-336, 2019.06, [URL], Subthalamic nucleus deep brain stimulation (STN-DBS) improves motor symptoms in individuals with advanced Parkinson’s disease (PD) and enables physicians to reduce doses of antiparkinsonian drugs. We investigated possible predictive factors for the successful reduction of antiparkinsonian drug dosage after STN-DBS. We evaluated 33 PD patients who underwent bilateral STN-DBS. We assessed rates of reduction of the levodopa-equivalent daily dose (LEDD) and levodopa daily dose (LDD) by comparing drug doses before vs. 6-months post-surgery. We used correlation coefficients to measure the strength of the relationships between LEDD and LDD reduction rates and preoperative factors including age, disease duration, preoperative LEDD and LDD, unified Parkinson’s Disease Rating Scale part-II and -III, levodopa response rate, Mini-Mental State Examination score, dyskinesia score, Hamilton Rating Scale for depression, and the number of non-motor symptoms. The average LEDD and LDD reduction rates were 61.0% and 70.4%, respectively. Of the variables assessed, only the number of psychiatric/cognitive symptoms was significantly correlated with the LEDD reduction rate. No other preoperative factors were correlated with the LEDD or LDD reduction rate. A wide range of preoperative psychiatric and cognitive symptoms may predict the successful reduction of antiparkinsonian drugs after STN-DBS..

How does citrus-like smell influence the memory of orange colour?, PloS one, 10.1371/journal.pone.0203876, 13, 9, 2018.09, [URL], Olfactory modulation of vision is not well understood whereas visual modulation of olfaction has been more fully investigated. This study aimed to reveal in a simple manner whether there is olfactory modulation of colour working memory using an odour that induces a citrus-like smell and is associated with orange colours. We assumed that the odour would have modulatory effects on the colour information stored in working memory. To clarify whether these effects are supportive or disruptive, during the colour working memory task we measured an event-related potential component, P3, which is involved in attentional processes of working memory. The results indicated that odour presentation mediated a decline in the rate of correct guesses for orange colours. Furthermore, the odour suppressed P3 during reddish-colour retrieval, including orange. These results suggest that colour working memory in orange can be disrupted via olfactory modulation with citrus-like odours..

, 10.1007/978-981-10-4361-1_73, 431-435, 2018.01, [URL], A general method for evaluating sleep is by identifying the stages of sleep from electroencephalogram (EEG), electrooculogram (EOG) and electromyogram (EMG). Unfortunately, visual sleep scoring is usually subjective and time consumptive. This was the reason for the development of automatic sleep staging based on multi EEG, EOG, and EMG. Recently, automatic sleep stages based on single channel EEG is also being developed to reduce the unnecessary number of wire. The problem is that this approach needs 90 min to complete the sleep stages cycle while normal short nap or day-time sleep experiment duration is usually less than 60 min. With this less than 60 min’ sleep duration issue, the purpose of this research is to develop a single channel EEG evaluation method by measuring changes in amplitude power of EEG at a specific time of day time sleep. The amplitude difference in term of a complexity of EEG will be used as an experiment index. We calculated EEG complexity with Multi-Scale Entropy (MSE) for day-time sleep experiment. The average entropy as the final result from the method could determine the EEG complexity of frequency power and have a negative correlation with the delta wave. Based on this result, average entropy was used and had demonstrated a significant difference. MSE method could be useful for single channel EEG evaluation and significantly perform better understanding of day-time sleep..

For instance, ubiquitous availability of smart devices and communication networks may have reduced the burden for people to remember factual information. At the same time, they may have increased the benefits to master the uses of new digital technologies. In the midst of such a social and technological shift, we could design novel integrated platforms that support people at all ages to learn, work, collaborate, and co-create easily. In this paper, we discuss our ideas and first steps towards building an extended learning analytics platform that elderly people and unskilled adults can use. By understanding the characteristics and needs of elderly learners and addressing critical user interface issues, we can build pervasive and inclusive learning analytics platforms that trigger contextual reminders to support people at all ages to live and learn actively regardless of age-related differences of cognitive capabilities. We discuss that resolving critical usability problems for elderly people could open up a plethora of opportunities for them to search and exploit vast amount of information to achieve various goals..

However, the relationship of these parameters to brain activity remains poorly understood. The objective of this study was to determine the effect of airflow on brain activity using electroencephalograms (EEG) of participants in a living environment under different airflow conditions. Before the recording, the room was set to a standardised air temperature and humidity. During the recording, each participant was required to perform a simple time-perception task that involved pressing buttons after estimating a 10-second interval. Cooling and heating experiments were conducted in summer and winter, respectively. A frequency analysis of the EEGs revealed that gamma and beta activities showed lower amplitudes under conditions without airflow than with airflow, regardless of the season (i.e., cooling or heating). Our results reveal new neurophysiological markers of the response to airflow sensation. Further, based on the literature linking gamma and beta waves to less anxious states in calm environments, we suggest that airflow may alter the feelings of the participants..

A magnetoencephalographic study, Hearing Research, 10.1016/j.heares.2016.10.027, 344, 82-89, 2017.01, [URL], In a previous magnetoencephalographic study, we showed both functional and structural reorganization of the right auditory cortex and impaired left auditory cortex function in people who stutter (PWS). In the present work, we reevaluated the same dataset to further investigate how the right and left auditory cortices interact to compensate for stuttering. We evaluated bilateral N100m latencies as well as indices of local and inter-hemispheric phase synchronization of the auditory cortices. The left N100m latency was significantly prolonged relative to the right N100m latency in PWS, while healthy control participants did not show any inter-hemispheric differences in latency. A phase-locking factor (PLF) analysis, which indicates the degree of local phase synchronization, demonstrated enhanced alpha-band synchrony in the right auditory area of PWS. A phase-locking value (PLV) analysis of inter-hemispheric synchronization demonstrated significant elevations in the beta band between the right and left auditory cortices in PWS. In addition, right PLF and PLVs were positively correlated with stuttering frequency in PWS. Taken together, our data suggest that increased right hemispheric local phase synchronization and increased inter-hemispheric phase synchronization are electrophysiological correlates of a compensatory mechanism for impaired left auditory processing in PWS..

Although understanding residual function is necessary for appropriate rehabilitation, the extent of its effects on cognitive improvement remains unknown. This study evaluated cognitive function in patients with severe motor and intellectual disabilities after early brain injuries due to cerebral hemorrhage or periventricular leukomalacia. We focused on neural responses to hearing the subject's own name (SON). According to previous studies, differences in response to SON are associated with several types of cognitive dysfunction. Methods: We examined healthy subjects (aged 21.4 6 1.10 years; control) and patients with a previous brain injury (aged 13-27 years at the time of our analysis) resulting in periventricular leukomalacia or a cerebral hemorrhage during the perinatal period or childhood. We recorded EEG responses to the SON and to other Japanese words, obtaining EEG-evoked potentials with wavelet transformations. Results: Compared with healthy controls, beta power (not alpha power) revealed differences in response to SON by patients with brain injury, especially those with cerebral hemorrhage. Conclusions: We suggest that alpha and beta power differences reflect different cognitive functions and that the SON response reveals more than one process. Beta powers may reflect the intellectual disability of cognitive function in response to selfrelevant stimuli, especially in patients with cerebral hemorrhage. Meanwhile, alpha powers did not differ from those of the healthy controls, suggesting that the patients perhaps paid attention to their own names..

The Hisayama Study, Journal of Alzheimer's Disease, 10.3233/JAD-160521, 55, 2, 613-624, 2017.01, [URL], Background: The Hisayama study is a prospective cohort study of lifestyle-related diseases that commenced in 1961. Through it, a significant increasing trend in the prevalence of Alzheimer's disease has been observed over the past 18 years. Objectives: We sought to investigate the increases in brain pathology related to Alzheimer's disease using automated MATLAB morphometric analyses for quantifying tau pathology. Methods: We examined a series of autopsied cases from Hisayama residents obtained between 1998 and 2003 (group A: 203 cases), and between 2009 and 2014 (group B: 232 cases). We developed custom software in MATLAB to analyze abnormal tau deposits quantitatively. Specimens were immunostained with both anti-amyloid-β-protein and anti-phosphorylated tau antibodies. Results: Both the Consortium to Establish a Registry for Alzheimer's Disease (CERAD) criteria for senile plaques and Braak stage for NFT were higher in group B. Morphometric analyses of the hippocampi also revealed a trend toward increased tau pathology in both men and women over 80 years of age in group B. The increases were also significant when the subjects were examined independently according to high or low CERAD scores and in all levels of AD neuropathologic change according to the National Institute on Aging-Alzheimer's Association guidelines (2012). Conclusion: We revealed a recent trend of increased tauopathy in the older people, which is partly independent of amyloid-β pathology..

A resting-state fMRI study, Cerebral Cortex, 10.1093/cercor/bht004, 24, 6, 1529-1539, 2014.06, [URL], It has been revealed that spontaneous coherent brain activity during rest, measured by functional magnetic resonance imaging (fMRI), self-organizes a small-world network by which the human brain could sustain higher communication efficiency across global brain regions with lower energy consumption. However, the state-dependent dynamics of the network, especially the dependency on the conscious state, remain poorly understood. In this study, we conducted simultaneous electroencephalographic recording with resting-state fMRI to explore whether functional network organization reflects differences in the conscious state between an awake state and stage 1 sleep. We then evaluated whole-brain functional network properties with fine spatial resolution (3781 regions of interest) using graph theoretical analysis. We found that the efficiency of the functional network evaluated by path length decreased not only at the global level, but also in several specific regions depending on the conscious state. Furthermore, almost two-thirds of nodes that showed a significant decrease in nodal efficiency during stage 1 sleep were categorized as the default-mode network. These results suggest that brain functional network organizations are dynamically optimized for a higher level of information integration in the fully conscious awake state, and that the default-mode network plays a pivotal role in information integration for maintaining conscious awareness..

Although the components of the oil and their biological activities have been studied extensively, the effect of the volatiles emitted from the roots of V. zizanioides on humans has so far remained unexplored. We investigated the effects of volatile compounds emitted from the cut roots of V. zizanioides (1.0 g, low-dose conditions; 30 g, high-dose conditions) on individuals during a visual display terminal task. Participants who breathed the volatile compounds emitted under low-dose conditions showed faster reaction times and stimulation of sympathetic nerve activity as measured by electrocardiography. These effects were not observed under high-dose conditions. The total amount of volatiles emitted during the experiment was 0.25 μg under low-dose conditions and 1.35 μg under high-dose conditions. These findings indicate that volatile compounds emitted from the roots of V. zizanioides under low-dose conditions may have helped subjects to maintain performance in visual discrimination tasks while maintaining high sympathetic nerve system activity..

There is evidence that the amplitude of the N20 component of median nerve somatosensory evoked potentials typically increases with age, probably because of cortical disinhibition. The secondary somatosensory cortex (SII) receives dual input from the SI and the thalamus. We quantified age-related changes both in SI and SII using magnetoencephalography (MEG). We recorded somatosensory evoked magnetic fields (SEFs) to median nerve stimulation in 15 young adults (aged 22-36 years, mean age 29.0±4.1) and 15 older adults (aged 52-67 years, mean age 61.9±5.4), and analysed major SEF components in SI and SII. The amplitude and equivalent current dipole (ECD) strength of the N20m were significantly increased in the older adults, consistent with the well-known electrophysiological change for cortical disinhibition in SI. The latency of N20m showed a trend for increase in older subjects, possibly reflecting slowing of conduction velocity in the peripheral nerves. In contrast, SII response (response peak at around 80-120 ms) showed a different change in aging. Latencies of the contralateral SII responses showed a trend for shortening in the older adults. There were no significant age-related changes for the amplitudes and ECD strengths. Thus, SI and SII are differently affected by aging. The shortening of the SII latency suggests age-related plastic-adaptive change in SII, which is mediated by the direct thalamocortical pathway..

To test whether spatial frequency (SF) information is responsible for this phenomenon, we recorded high-density event-related potentials (ERPs) with unfiltered or spatially filtered word stimuli in Japanese-speaking subjects. Kanji (early-learned, late-learned), Kana (word, non-word), and scrambled characters served as stimuli. Fourier analysis revealed that Kanji and Kana were characterized by high-SF (HSF) and low-SF (LSF) information, respectively. In ERPs with unfiltered stimuli, bilateral occipital P100, left occipitotemporal N170 and fronto-central N400 were elicited. Scrambled characters did not evoke left-lateralized N170 or clear N400. Under the LSF condition, P100 and N170 latencies for Kanji were significantly longer than those for Kana. In the HSF condition, P100 and N170 latencies for late-learned Kanji were significantly longer than those for early-learned Kanji. There was no significant difference in the N400 between Kanji and Kana in both SF conditions. These results suggest that early visual responses, but not the semantic component, are influenced by SF. This indicates a close link between Kana and LSF information, and between Kanji and HSF information. The differential effects of SF could underlie the neural basis of the differences between Kanji and Kana reading..

Additionally, we employed voxel-based morphometry to compare cortical gray matter (GM) volumes on structural MR images. PWS exhibited impaired left auditory sensory gating. The tonotopic organization in the right hemisphere of PWS is expanded compared with that of the controls. Furthermore, PWS showed a significant increase in the GM volume of the right superior temporal gyrus, consistent with the right tonotopic expansion. Accordingly, we suggest that PWS have impaired left auditory sensory gating during basic auditory input processing and that some error signals in the auditory cortex could result in abnormal speech processing. Functional and structural reorganization of the right auditory cortex appears to be a compensatory mechanism for impaired left auditory cortex function in PWS..

The differences in local organization of domains and pinwheels have been the subject of considerable controversy. Here, we focus on the differences in global organizations of domains and pinwheels. We analyzed the spatial distribution of orientation preferences in wide regions around domains and pinwheels in monkey V1. A particular distribution pattern of orientation preferences was found more clearly in the regions around domains than in the regions around pinwheels. Then we analyzed the phase-locking values (PLVs) of the induced gamma activity to assess neural synchrony within the somatosensory cortical network. We also assessed PLVs in patients with multiple sclerosis (MS) to validate our PLV analysis in evaluating the inter-areal functional connectivity, which can often be impaired in MS. The PLVs of the induced gamma activity were calculated for each pair of unaveraged MEG signals that represented the activities of the contralateral SI and bilateral SII areas. Analysis of PLVs between the SI and SII areas showed significantly increased PLVs for gamma-band activities, starting at an early post-stimulus stage in normal controls, whereas this increase in PLVs was apparently diminished in MS. The PLV analysis provided evidence for early-latency, gamma-band neuronal synchronization between the SI and SII areas in normal controls. Our study first demonstrates the gamma-band synchrony in the early-stage human somatosensory processing..

Methods: The borders of the STN were determined by microelectrode recording. The most eligible trajectory for the sensorimotor area of the STN was used for LFP recording while advancing the DBS electrode. Results: The low-frequency LFP power (θ- to β-band) increased from a few millimeters above the dorsal border of the STN defined by microelectrode recording; however, the low-frequency power kept the same level beyond the ventral border of the STN. Only high β-power showed close correlation to the dorsal and ventral borders of the STN. Conclusions: A spectral power analysis of LFP recording by DBS electrode helps with the final confirmation of the dorsal and ventral borders of the STN of Parkinson's disease in DBS implantation surgery..

Methods: The SEPs elicited by stimulating the right median nerve were recorded in 14 young and 16 older healthy subjects, while self-initiated movements of the right fingers were performed at 5-10 s intervals. The amplitudes of the major components of the SEPs at F3 and C3′ (2 cm posterior to C3) during the pre-movement period were analysed as the resting condition subserving the baseline. Results: The amplitudes at rest were significantly greater in the elderly than in the younger subjects. The amplitudes of P27, N35 and P45 at C3′ as well as N30 at F3 decreased significantly during the pre-movement period. However, the ratio of amplitudes in the pre-movement period to the resting period in the elderly was not significantly different from that in the younger subjects, except for the interaction of N30. Conclusions: The effect of age on the gating of N30 at F3 may indicate an altered preparatory processing of self-initiated movement in the elderly. The gating effect of older subjects at C3′ is almost comparable to that of young ones, which appears to be a compensatory mechanism to maintain the precise movements. Contextual modulation on orientation-contrast is one typical example of nonclassical neuronal behavior. This modulation by surrounding stimuli in a nonclassical receptive field is mainly thought to be mediated by short- and long-range horizontal connections within the primary visual cortex. Short-range connections are circularly symmetrical and relatively independent of orientation preferences, while long-range connections are patchy, asymmetrical, and orientation specific. Although this modulation can be explained by long-range specific connections qualitatively, recent studies suggest that long-range connections alone may be insufficient with respect to the balance between two types of connections. Here, in order to clarify the role of short-range connections in the process of contextual modulation, we propose a model of the primary visual cortex with isotropic short-range connections and a geometric orientation map. Computational simulations using the model have demonstrated that contextual modulation can be explained by short-range connections alone. This is due to the interaction between the spatial periodicity of orientation domains and the excitatory-inhibitory regions arising from the propagation of activities..
