Akiko Mizokami, Takahito Otani, Takashi Kanematsu, Eijiro Jimi, Masato Hirata, Role of G-protein coupled receptor GPRC6A in regulating adipose tissue metabolism, 第63回歯科基礎医学会学術大会, 2021.10, GPRC6A, a member of family C of G protein-coupled receptors, has been proposed to be a master regulator of metabolic processes. It has been reported that global deletion of GPRC6A increases susceptibility to develop diet-induced obesity and metabolic related disorders. However, due to its broad tissue distribution and its ability to respond to a variety of hormonal and nutritional signals, the underlying cellular and molecular mechanisms of the metabolic disorders are still unclear. We previously reported that long-term oral administration of osteocalcin, one of the GPRC6A ligands, markedly reduced adipocyte size and improved glucose tolerance in wild type mice. Thus, we generated adipocyte-specific GPRC6A knockout mice (adG6AKO) and found that these animals manifested increased adipose tissue weight, adipocyte hypertrophy, and adipose tissue inflammation under obesogenic environment. These effects were associated with reduced lipolytic activity due to down-regulation of lipolytic enzymes in adipose tissue of adG6AKO mice. Our results suggest that the constitutive activation of GPRC6A signaling in adipocytes plays a key role in adipose lipid handling and the prevention of obesity and related metabolic disorders..
